Throughout its 40-year history,the field of gene therapy has been marked by many *** has seen great strides in combating human disease,has given hope to patients and families with limited treatment options,but has also been subject to many *** of patients with this class of investigational drugs has resulted in severe adverse effects and,even in rare cases,*** the heart of this dichotomous field are the viral-based vectors,the delivery vehicles that have allowed researchers and clinicians to develop powerful drug platforms,and have radically changed the face of *** the past 5 years,the gene therapy field has seen a wave of drugs based on viral vectors that have gained regulatory approval that come in a variety of designs and purposes,these modalities range from vector-based cancer therapies,to treating monogenic diseases with life-altering *** present,the three key vector strategies are based on adenoviruses,adeno-associated viruses,and *** have led the way in preclinical and clinical successes in the past two ***,despite these successes,many challenges still limit these approaches from attaining their full *** review the viral vector-based gene therapy landscape,we focus on these three highly regarded vector platforms and describe mechanisms of action and their roles in treating human disease.

Removal of cadmium using MnO_2 loaded D301 resin

MnO2 loaded weak basic anion exchange resin D301 （Anion exchange resin, macroreticular weak basic styrene） as adsorbent has been prepared and applied to the removal of cadmium. The adsorption characteristics have been investigated with respect to effect of pH, equilibrium isotherms, removal kinetic data, and interference of the coexisting ions. The results indicated that the Cd^2＋ could be efficiently removed using MnO2 loaded D301 resin in the pH range of 3-8 from aqueous solutions with the co-existence of high concentration of alkali and alkaline-earth metals ions. The saturate adsorption capacity of the Cd^2＋ was 77.88 mg/g. The adsorption process followed the pseudo first-order kinetics. The equilibrium data obtained in this study accorded excellently with the Langmuir adsorption isotherm.

Hearing impairment(HI)affects 1/1000 children and over 2%of the aged *** have previously reported that mutations in the gene encoding gap junction protein connexin-31(Cx31)are associated with *** pathological mechanism of the disease mutations remains ***,we show that expression of Cx31 in the mouse inner ear is developmentally regulated with a high level in adult inner hair cells and spiral ganglion neurons that are critical for the hearing *** transfected cells,wild type Cx31 protein(Cx31wt)forms functional gap junction at *** contrast,two HIassociated Cx31 mutants,Cx31R180X and Cx31E183K resided primarily in the ER and Golgi-like intracellular punctate structures,respectively,and failed to mediate lucifer yellow *** of Cx31 mutants but not Cx31wt leads to upregulation of and increased association with the ER chaperone BiP indicating ER stress ***,the HI-associated Cx31 mutants are impaired in trafficking,promote ER stress,and hence lose the ability to assemble functional gap *** study reveals a potential pathological mechanism of HI-associated Cx31 mutations.

Effects of Rice Straw and Its Biochar Addition on Soil Labile Carbon and Soil Organic Carbon

Whether the biochar amendment could affect soil organic matter （SOM） turnover and hence soil carbon （C） stock remains poorly understood. Effects of the addition of ^13C-labelled rice straw or its pyrolysed biochar at 250 or 350℃ to a sugarcane soil （Ferrosol） on soil labile C （dissolved organic C, DOC; microbial biomass C, MBC; and mineralizable C, MC） and soil organic C （SOC） were investigated after 112 d of laboratory incubation at 25℃. Four treatments were examined as （1） the control soil without amendment （Soil）; （2） soil plus ^13C-labelled rice straw （Soil＋Straw）; （3） soil plus 250℃ biochar （Soil＋B250） and （4） soil plus 350℃biochar （Soil＋B350）. Compared to un-pyrolysed straw, biochars generally had an increased aryl C, carboxyl C, C and nitrogen concentrations, a decreased O-alkyl C and C：N ratio, but similar alkyl C and δ^13C （1 742- 1 877 %）. Among treatments, significant higher DOC, MBC and MC derived from the new C （straw or biochar） ranked as Soil＋Straw〉Soil＋B250〉Soil＋B350, whilst significant higher SOC from the new C as Soil＋B250〉Soil＋Straw≈Soil＋B350. Compared to Soil, DOC and MBC derived from the native soil were decreased under straw or biochar addition, whilst MC from the native soil was increased under straw addition but decreased under biochar addition. Meanwhile, native SOC was similar among the treatments, irrespective of the straw or biochar addition. Compared to Soil, significant higher total DOC and total MBC were under Soil＋Straw, but not under Soil＋B250 and Soil＋B350, whilst significant higher total MC and total SOC were under straw or biochar addition, except for MC under Soil＋B350. Our results demonstrated that the application of biochar to soil may be an appropriate management practice for increasing soil C storage.

The tapetum,a crucial innermost layer encompassing male reproductive cells within the anther wall,plays a pivotal role in normal pollen *** transcription factors (TFs) bHLH010/089/091 redundantly facilitate the rapid nuclear accumulation of DYSFUNCTIONAL TAPETUM 1,a gatekeeper TF in the ***,the regulatory mechanisms governing the activity of bHLH010/089/091 remain *** this study,we reveal that caffeoyl coenzyme A O-methyltransferase 1 (CCoAOMT1) is a negative regulator affecting the nuclear localization and function of bHLH010 and bHLH089,probably through their K259 *** findings underscore that CCoAOMT1 promotes the nuclear export and degradation of bHLH010 and ***,elevated CCoAOMT1 expression resulted in defective pollen development,mirroring the phenotype observed in bhlh010 bhlh089 ***,our investigation revealed that the K259A mutation in the bHLH089 protein disrupted its translocation from the nucleus to the cytosol and impeded its degradation induced by ***,transgenic plants with the probHLH089::bHLH089^(K259A)construct failed to rescue proper pollen development or gene expression in bhlh010 bhlh089 ***,these findings emphasize the need to maintain balanced TF homeostasis for male *** firmly establish CCoAOMT1 as a pivotal regulator that is instrumental in achieving equilibrium between the induction of the tapetum transcriptional network and ensuring appropriate anther development.

An Algorithm for Cavity Reconstruction in Electrical Impedance Tomography

We consider the inverse problem of finding cavities within some object from electrostatic measurements on the boundary. By a cavity we understand any object with a different electrical conductivity from the background material of the body. We give an algorithm for solving this inverse problem based on the output nonlinear least-square formulation and the regularized Newton-type iteration. In particular, we present a number of numerical results to highlight the potential and the limitations of this method.

The transient photoluminescence(PL)of DO-PPV(poly-(2,5-dioctyloxy-1,4-phenylene vinylene))solution in chloroform was investigated by picosecond time-resolved PL *** ultrafast rise of PL and the following single exponential decay with a time constant about 400ps were assigned to the formation of intrachain exciton and its decay process,*** redshift of the PL emission spectrum with time was caused by the subsequent exciton migration among the different conjugated segments in the DO-PPV polymer.

Tea consumption and risk of stroke:a dose-response meta-analysis of prospective studies

Objective:To determine the association between tea consumption and the risk of stroke. Methods:We searched the PubMed database from January 1966 to march 2012 and reviewed reference lists of retrieved articles to identify relevant studies. Studies were included if they reported relative risks (RRs) and corresponding 95% confidence intervals (CIs) of stroke with respect to three or more categories of tea consumption. A random-effects model was used to combine the study-specific risk estimates. Results:Fourteen studies, consisting of 513 804 participants with a median follow-up of 11.5 years, were included in this meta-analysis. We observed a modest but statistically significant inverse association between tea consumption and risk of stroke. An increase of three cups/d in tea consumption was associated with a 13% decreased risk of stroke (RR 0.87; 95% CI, 0.81-0.94). The decreased risk of stroke with tea consumption was consistent among most subgroups. Based on the three studies that provided results for stroke subtypes, tea consumption was also inversely associated with the risk of ischemic stroke (RR 0.76; 95% CI, 0.69-0.84), but not cerebral hemorrhage (RR 0.96; 95% CI, 0.82-1.11) or subarachnoid hemorrhage (RR 0.81; 95% CI, 0.57-1.16). Conclusions:Tea consumption is associated with a decreased risk of stroke, particularly ischemic stroke. More well-designed, rigorously conducted studies are needed in order to make confident conclusions about the association between tea consumption and stroke subtypes.
