Ferroptosis inhibitors reduce celastrol toxicity and preserve its insulin sensitizing effects in insulin resistant HepG2 cells

Objective:Research has shown that celastrol can effectively treat a variety of diseases,yet when passing a certain dosage threshold,celastrol becomes toxic,causing complications such as liver and kidney damage and erythrocytopenia,among *** this dichotomy in mind,it is extremely important to find ways to preserve celastrol’s efficacy while reducing or preventing its ***:In this study,insulin-resistant Hep G2(IR-Hep G2)cells were prepared using palmitic acid and used for in vitro ***-Hep G2 cells were treated with celastrol alone or in combination with Nacetylcysteine(NAC)or ferrostatin-1(Fer-1)for 12,24 or 48 h,at a range of *** counting kit-8assay,Western blotting,quantitative reverse transcription-polymerase chain reaction,glucose consumption assessment,and flow cytometry were performed to measure celastrol’s cytotoxicity and whether the cell death was linked to ***:Celastrol treatment increased lipid oxidation and decreased expression of anti-ferroptosis proteins in IR-Hep G2 *** downregulated glutathione peroxidase 4(GPX4)m *** docking models predicted that solute carrier family 7 member 11(sLC7A11)and GPX4 were covalently bound by ***,we found for the first time that the application of ferroptosis inhibitors(especially NAC)was able to reduce celastrol’s toxicity while preserving its ability to improve insulin sensitivity in IR-Hep G2 ***:One potential mechanism of celastrol’s cytotoxicity is the induction of ferroptosis,which can be alleviated by treatment with ferroptosis *** findings provide a new strategy to block celastrol’s toxicity while preserving its therapeutic effects.

Techno-Economic Optimization of Novel stand-Alone Renewable Based Electric Vehicle Charging station near Bahria Town Karachi,sindh Pakistan

Electric vehicles (EVs) are the most interesting and innovative technology in the 21st century because of theirenormous advantages, both technically and economically. Their emissions rate compared to fuel-based vehicles isnegligible as they do not consume fuel and hence do not emit any harmful gases. However, their bulk production,adoption and lack of charging stations increase the stress of power stations due to modern-day lifestyles. If Electricvehicles demand increases drastically then conventional power stations will not bear their demand and if theygenerate electricity by conventional means it will be very costly and may further add greenhouse gases. Therefore,this research provides the techno-economic assessment of a stand-alone renewable-dependent electric vehiclecharging station, excluding any burden on electrical utility. The proposed study is carried out in Bahria Town,Karachi, a city in Pakistan. In this study, HOmER Pro software was utilized for techno-economic assessment. Ahybrid system comprising solar Photovoltaic/Wind Turbine/Fuel cells and battery storage is included in the *** and wind resources were taken from NAsA’s website, where charging stations will be integrated. The overallresults show promising in terms of total Net Present Cost and the Cost of Energy which are 2.72m $ and 0.237 $,respectively. The total system generation is 3,598 megawatt hours per year, and the total energy consumption is 885megawatt hours per year.

The availability of nitrogen(N)is crucial for both the productivity of terrestrial and aquatic ecosystems ***,the overuse of artificial fertilizers and the energy required to fix nitrogen have pushed the global nitrogen cycle(N-cycle)past its safe operating limits,leading to severe nitrogen pollution and the production of significant amounts of greenhouse gas nitrous oxide(N2O).The anaerobic ammonium oxidation(anammox)mechanism can counteract the release of ammonium and N2O in many oxygenlimited situations,assisting in the restoration of the homeostasis of the Earth’s N *** this work,we looked into the characteristics of the anammox hotspots’distribution across various types of ecosystems *** hotspots are present at diverse oxic-anoxic interfaces in terrestrial systems,and they are most prevalent at the oxic-anoxic transition zone in aquatic *** on the discovery of an anammox hotspot capable of oxidizing ammonium anoxically into N2 without N2O by-product,we then designed an innovative concept and technical routes of nature-based anammox hotspot geoengineering for climate change,biodiversity loss,and efficient utilization of water *** 15 years of actual use,anammox hotspot geoengineering has proven to be effective in ensuring clean drinking water,regulating the climate,fostering plant and animal diversity,and enhancing longterm environmental *** sustainable biogeoengineering of anammox could be a workable natural remedy to resolve the conflicts between environmental pollution and food security connected to N management.
