Intestine is responsible for the biotransformation of many orally-exposed *** constitutive androstane receptor(CAr/Nr1i3) is known to up-regulate many genes encoding drugmetabolizing enzymes and transporters(drug-processing genes/DPGs) in liver,but less is known regarding its effect in ***-day-old wild-type and Car / mice were administered the CArligand TCPOBOP or vehicle once daily for 4 *** wild-type mice,Car m rNA was down-regulated by TCPOBOP in liver and *** / mice had altered basal intestinal expression of many DPGs in a section-specific *** with the liver data(Aleksunes and Klaassen,2012),TCPOBOP upregulated many DPGs(Cyp2b10,Cyp3a11,Aldh1a1,Aldh1a7,Gsta1,Gsta4,Gstm1-m4,Gstt1,Ugt1a1,Ugt2b34,Ugt2b36,and mrp2–4) in specific sections of small intestine in a CAr-dependent ***,the m rNAs of Nqo1 and Papss2 were previously known to be up-regulated by TCPOBOP in liver but were not altered in ***,many known CAr-target genes were highest expressed in colon where CAr is minimally expressed,suggesting that additional regulators are involved in regulating their *** conclusion,CAr regulates the basal expression of many DPGs in intestine,and although many hepatic CAr-targeted DPGs were bona fide CAr-targets in intestine,pharmacological activation of CAr in liver and intestine are not identical.

The approval of immune checkpoint inhibitors(ICIs)has changed the treatment landscape in many aspects of urothelial cancer(UC),in both non-muscle-invasive bladder cancer and muscle-invasive bladder cancer and has introduced the concept of long-term remission for some patients in the metastatic ***-line chemotherapy remains superior at achieving initial control of disease compared to front-line immune ***,long-term durable responses are limited by chemotherapy *** maintenance approach,sequencing chemotherapy with ICIs,could be considered a best of both worlds approach,achieving initial control with chemotherapy,which is maintained in some individuals with ***,outcomes for patients with metastatic UC remain *** are three steps to improving outcomes for these patients;the first is to develop better drugs and combinations of therapies,the second is the development of novel biomarkers and techniques to better select patients for treatment,and the third area of development is to give the drugs in the most optimal setting.

Acute liver failure (ALF) is a devastating clinical syndrome characterised by progressive encephalopathy, coagulopathy, and circulatory dysfunction, which commonly leads to multiorgan failure and death. Central to the pathogenesis of ALF is activation of the immune system with mobilisation of cellular effectors and massive production of cytokines. As key components of the innate immune system, monocytes and macrophages are postulated to play a central role in the initiation, progression and resolution of ALF. ALF in humans follows a rapidly progressive clinical course that poses inherent difficulties in delineating the role of these pivotal immune cells. Therefore, a number of experimental models have been used to study the pathogenesis of ALF. Here we consider the evidence from experimental and human studies of ALF on the role of monocytes and macrophages in acute hepatic injury and the ensuing extrahepatic manifestations, including functional monocyte deactivation and multiple organ failure.

Atlantic halibut(Hippoglossus hippoglossus,Linnaeus,1758)are a data-poor stock within the waters of the United *** study evaluated the use of otolith measurements to back-calculate lengths of Atlantic halibut at previous ***-calculations have proven useful for estimating length at age and growth rates of other *** the best knowledge of the authors,this study is the first to document the use of this method for Atlantic *** back-calculations rely on a few key assumptions,such as proportionality of fish length and otolith length,which are not always *** study shows that backcalculations using the Fraser-Lee method can provide reasonable estimates of Atlantic halibut length at previous ages,especially when samples from young halibut are included to improve estimates of the intercept of the linear *** on back-calculated estimates,female and male halibut in the Gulf of maine showed different growth rates after age *** was no evidence of changes in growth rates over an approximately 15 year time *** caught in the Gulf of maine and on the neighboring Scotian Shelf showed some differences in growth rates;however,the results did not support strong conclusions about differences between the two regions as the direction of the differences was not consistent between the sexes and previous tagging studies have shown extensive movement between the two *** finding of reasonably accurate back-calculated lengths at previous ages is important for this data-poor species,as back-calculations increase the amount of information that can be obtained from otoliths.
