Bioactive compounds fractionated from endophyte Streptomyces SUK 08 with promising ex-vivo antimalarial activity

Objective: To determine ex vivo antimalarial activity and cytotoxicity of endophytic Streptomyces SUK 08 as well as the main core structure fractionated from its crude ***: The activities of SUK 08 crude extract were evaluated by using the Plasmodium lactate dehydrogenase assay and synchronization test against rodent malaria parasite Plasmodium berghei, instead of human malarial parasite Plasmodium falciparum. The cytotoxicity of the crude extract was determined by mTT assay. The crude extract was analyzed by thin-layer chromatography and gas chromatography–mass ***: The ethyl acetate crude extract showed very promising antimalarial activity with IC50 of 1.25 mg/m l. The synchronization tests showed that ethyl acetate extraction could inhibit all stages of the Plasmodium life cycle, but it was most effective at the Plasmodium ring stage. On the basis of a mTT assay on Chang liver cells, ethyl acetate and ethanol demonstrated IC50 values of >1.0 mg/m l. The IC50 of parasitemia at 5% and30% for this extract was lower than chloroquine. Thin-layer chromatography, with 1: 9 ratio of ethyl acetate: hexane, was used to isolate several distinct compounds. Based on gas chromatography–mass spectrophotometry analysis, three core structures were identified as cyclohexane, butyl propyl ester, and 2,3-heptanedione. Structurally, these compounds were similar to currently available antimalarial ***: The results suggest that compounds isolated from Streptomyces SUK 08 are viable antimalarial drug candidates that require further investigations.

The changing dynamics,non-linearity of soil moisture content,as well as other weather and plant variables requires real-time monitoring and accurate predictive model for effective irrigation and crop *** this paper,an improved monitoring and datadriven modelling of the dynamics of parameters affecting the irrigation of mustard leaf plant is *** IoT-based monitoring framework is implemented using ESPresso lite V2.0 module interfaced with different soil moisture sensors(VH-400),flowmeter(YF-S201)as well as Davis vantage pro 2 weather station to measure soil moisture content,irrigation volume,and computation of the reference evapotranspiration(ETo).The data collected including plant images were transmitted to the Raspberry Pi 3 controller for onward online storage and the data are displayed on the IoT *** combination of both soil moisture and ETo values was used for scheduling a drip irrigated plant grown in a greenhouse for 35 days.A total number of 20,703 experimental data samples are collected from the IoT-based platform was further used for data driven modelling through system identification in *** result shows the development of different predictive models for soil moisture content *** ARX prediction model is found to perform better than the ARmX,BJ and State space model in terms of estimated fit of 91.31%,91.09%,91.08%,and 90.75%***,a robust monitoring framework for irrigation system has been developed,while the performance of the identified ARX model is promising to predict the volumetric soil water content.

Nutritive and mineral Composition in a Collection of Cucurbita pepol Grown in Kosova

In Kosovo, in most cases C. pepo is grown both in maize fields and vegetable gardens as well as in other more intensive systems. Our expedition has identified different location t of Kosovo where we were collected a samples at altitudes from 545 till 748 m.a.s.l. The aim of the present investigation was carried out to study morphological diversity, protein and lipids content. Protein content (PC) was determined by Kjeldahl method. lipids were determined by extraction using Soxhlet apparatus and diethyl ether was used as extraction solvent. Pumpkin populations show great diversity for quality traits. The greater part of variance was accounted for Protein Content (PC) and lipid Content (lC). The estimation overall value μ in seed for PC and lC were 14.59%, and 32.99% respectively. The genetic diversity studies of the investigated of C. pepo showed significant genetic variation on chemical parameters, which could be largely due to that they were of different origins. This evaluation of traits variability can assist breeders to identify populations with desirable characteristics for inclusion in variety breeding programs.

Iodine and Selenium Intake in a Sample of Women of Childbearing Age in Palmerston North, New Zealand after mandatory Fortification of Bread with Iodised Salt

Iodine deficiency is a worldwide public health problem, which has long been observed in many parts of the world, including New Zealand (NZ). The aim of this study was to assess iodine and selenium intake among women of childbearing age in Palmerston North, New Zealand post mandatory fortification of bread with iodised salt. Fifty women of childbearing age completed a researcher-led questionnaire, including a semi-quantitative food frequency questionnaire. Iodine and selenium were analysed in 24-hour urine samples. The median urinary iodine concentration (UIC) was 65 μg/l with 30% below 50 μg/l;representing mild iodine deficiency according to the World Health Organization. The estimated median daily iodine intake (130 μg/day) was higher than the Estimated Average Requirement (100 μg/day) and higher than seen in women prior to fortification. The median excretion of selenium (32 μg/day) was slightly above level suggested as adequate (30 μg/day) and estimated median intake (57 μg/day) was higher than Estimated Average Requirement (50 μg/day). Selenium and iodine excretion were significantly correlated (Spearman’s rank order;r(50) = 0.547, p 0.001). The major contributors to iodine intake were milk (36%), bread (25%) and fish/seafood (15%). Participants had a mean intake of 2.5 slices of bread/day, which contributed approximately 14 to 20 μg of iodine. The majority of participants (74%) had iodised salt at home, but less than half (48%) used iodised salt exclusively. In conclusion, despite the mandatory fortification of bread with iodised salt in NZ, UIC of the study population indicates iodine deficiency although their estimated dietary intakes appear adequate. It is essential that government initiatives to improve iodine status are evaluated for their efficacy.

In Vitromicropropagation of a Valuable medicinal Plant, Plectranthus amboinicus

The effect of the plant growth regulators benzyl amino purine (BAP), 1-naphthaleneacetic acid (NAA) and kinetin (KIN) on in vitro shoot induction and proliferation of Plectranthus amboinicus was examined. Explants obtained from lateral shoots and apical shoots of P. amboinicus were inoculated on murashige and Skoog (mS) culture medium supplemented with different concentrations of BAP, NAA and KIN. When the effect of each growth regulator was considered singly, the highest rate of shoot induction (80% of explants producing shoots) and highest number of shoots produced (2.4 shoots per explant) were obtained from lateral shoot explants cultured on mS media supplemented with 3.0 mg/l BAP within 6 - 7 weeks. Better results were obtained using mS medium supplemented with 1 mg/l BAP + 5 mg/l NAA. Shoot proliferation rose to 85%, while 5.7 shoots per explants were recorded. Among the different media tested for rooting, mS medium supplemented with 1.0 mg/l IBA was the most effective for root induction. The quality of the roots obtained was better than that obtained using mS media supplemented with NAA or IAA.

背景：接受非心脏手术的老年患者出现红细胞压积异常和心血管并发症的危险较高。尽管患者术前几乎都要进行红细胞压积筛查，但是有关术前贫血及红细胞增多症的不良影响的证据较为有限。目的：于实施非心脏大手术的退伍老兵中评估术前贫血和红细胞增多症的发生率及其对术后30天预后的影响。设计：利用全国退伍军人事务委员会（VA）于术质量改进计划数据库进行回顾性队列研究。根据术前红细胞压积水平将患者分为三类：贫血（红细胞压积〈39．0％）、正常（红细胞压积39．0％～53．9％）和红细胞增多症（红细胞压积≥54％）。对红细胞压积异常与术后30天心血管事件发生率及死亡率的关系进行评估。地点及患者：纳入1997至2004年于全美132个退伍军人医疗中心接受非心脏大手术的310311例患者（年龄≥65岁）。主要观测指标：主要观测指标为术后30天死亡率，次要观测指标包括术后30天死亡率或心脏事件（心脏骤停或Q波心肌梗死）发生率。结果：与红细胞压积正常者相比，红细胞压积发生正向或负向偏离者术后30天死亡率和心脏事件发生率均有所增加。研究发现，红细胞压积水平较正常范围每增减1个百分点，术后30天死亡率就可增加1．6％（95％可信区间，1．1％-2．2％）。进一步分析显示，当红细胞压积降至39％以下或超过51％时，校正后30天死亡风险和心脏病患病风险就开始增加。结论：接受非心脏大手术的老年患者（绝大多数为男性退伍军人）术前轻度贫血及红细胞增多症均可增加术后发生死亡和心脏事件的危险。今后的研究应明确上述发现可否见于其他人群，明确术前纠正贫血和红细胞增多症能否降低术后死亡风险。
