SBL-JP-0004: A promising dual inhibitor of JAK2 and PI3KCD against gastric cancer

Background:Gastric cancer (GC) remains a global health burden and is often characterized by heterogeneous molecular profiles and resistance to conventional therapies. The phosphoinositide 3-kinase and PI3K and Janus kinase (JAK) signal transducer and activator of transcription (JAK-STAT) pathways play pivotal roles in GC progression, making them attractive targets for therapeutic interventions.methods:This study applied a computational and molecular dynamics simulation approach to identify and characterize SBL-JP-0004 as a potential dual inhibitor of JAK2 and PI3KCD kinases. KATOIII and SNU-5 GC cells were used for evaluation.Results:SBL-JP-0004 exhibited a robust binding affinity for JAK2 and PI3KCD kinases, as evidenced by molecular docking scores and molecular dynamics simulations. Binding interactions and Gibbs binding free energy estimates confirmed stable and favorable interactions with target proteins. SBL-JP-0004 displayed an half-maximal inhibitory concentration (IC) value of 118.9 nm against JAK2 kinase and 200.9 nm against PI3KCD enzymes. SBL-JP-0004 exhibited potent inhibition of cell proliferation in KATOIII and SNU-5 cells, with half-maximal growth inhibitory concentration (GI) values of 250.8 and 516.3 nm, respectively. A significant elevation in the early phase apoptosis (28.53% in KATOIII cells and 26.85% in SNU-5 cells) and late phase apoptosis (17.37% in KATOIII cells and 10.05% in SNU-5 cells) were observed with SBL-JP-0004 treatment compared to 2.1% and 2.83% in their respective controls.Conclusion:The results highlight SBL-JP-0004 as a promising dual inhibitor targeting JAK2 and PI3KCD kinases for treating GC and warrant further preclinical and clinical investigations to validate its utility in clinical settings.

The single-phase high-entropy alloy film is difficult to meet severe friction conditions due to its low hardness and high friction ***-composite structure film is composed of at least two separated phases,showing the properties of strength and toughness integration and excellent wear *** design of nanocomposite structures can effectively improve the mechanical properties and tribological properties of high-entropy alloy *** this study,the(CuNiTiNbCr)C_(x) nanocomposite high-entropy films(HEFs)integrated with high hardness,high toughness,and self-lubrication were synthesized by the double-target co-sputtering *** effect of carbon content on microstructure,mechanical properties,and tribological properties of(CuNiTiNbCr)C_(x) films was *** the increase of carbon content in the HEFs,the carbon atoms preferentially react with Ti,Nb,and Cr to form a(TiNbCr)C ceramic-reinforced phase,and then the excess carbon atoms precipitate in the form of amorphous carbon(a-C)lubricating phase in the *** structure of the HEFs changes from an amorphous structure to a nanocomposite structure of amorphous(amorphous CuNiTiNbCr phase+a-C phase)/nanocrystalline(TiN-bCr)C *** the carbon content is about 21.2 at.%,the carbide phase in the film reaches saturation and the hardness and modulus of the films are highest,which are 18 GPa and 228 GPa,*** HEFs with a carbon content of 44.0 at.%show the best toughness and tribological properties with a friction coefficient of 0.16 and a wear rate of 2.4×10^(-6) mm^(3)/(N m),which is mainly attributed to the excellent resistance to fatigue crack growth and the interfacial lubricating layer formed in the friction *** nanocomposite(CuNiTiNbCr)C_(x) HEFs show very promising application prospect in the field of friction protection.

metabolic adaptation of cancer cells is a key tactic for maintaining their tumorigenicity and viability in the tumor immune microenvironment （TImE）.Concurrent modulation on the metabolic pathway and immune system could change the living condition of cancer cells and potentiate the efficacy of anticancer drugs.A copper complex （DDCu） was designed to influence the energy metabolism of cancer cells and reshape the *** restrained the production and transportation of lactate by inhibiting the expression of lactate dehydrogenase and monocarboxylate transporter 4,thereby altering the metabolic pathway of cancer cells and improving the acidic tumor ***,DDCu was reduced to Cu+by cellular glutathione to react with lipoylated proteins of the tricarboxylic acid cycle and destabilize the Fe–S cluster proteins,leading to the aggregation of dihydrolipoamide S-acetyltransferase and cuproptosis in cancer ***,DDCu promoted the polarization of macrophages from the m2 to m1 phenotype,activated CD4+and CD8+T cells,and reversed the *** a result,DDCu inhibited the tumor growth through a synergy between metabolic regulation and immunomodulation.

Formation and Dissociation of Bamboo-like ErD_2/ErD_3 Grains

The formation and dissociation of Er deuteride grains were investigated by using scanning electron microscopy, in-situ x-ray diffraction and thermal desorption spectroscopy. Pure Er film shows classic columnar structure, while the Er deuteride film was observed as bamboo-like morphology. The in-situ x-ray diffraction results demonstrate that the diffraction peaks of 27.39°, 28.43°, 31.71°, and 39.95° are ascribed to the crystal planes of （002）, （110）, （111）, and （112） of ErD3 phase, respectively. It is also confirmed that by in-situ x-ray diffraction technique, ErD3 grains would be dissociated at temperature exceeding 400 ℃. This result is consistent with the conclusion obtained by thermal desorption spectroscopy.

Design process of A/ADm-BCn type directional eutectic composites was instanced by investigating Al-mg-Si-Li and Ni-mo-C-Al systems in this study by thermoanalysis and directional solidification techniques. It was demonstrated that A(D)-BCn eutectic alloys could be obtained more conveniently by using Smith′s thermoanalysis method to trace change of the eutectic Points of A-BCn induced by addition of components D. A/ADm-BCn directional eutectic composites could be produced from directionally solidified A/ADm-BCn alloys after proper heat treatments if components D have higher solid solubility in components A at high temperature and stable of metastable phases ADm could be formed at room temperature.

Nonlinear forced vibration in a subcritical regime of a porous functionally graded pipe conveying fluid with a retaining clip

This study examines the nonlinear behaviors of a clamped-clamped porous pipe made of a functionally graded material(FGm) that conveys fluids and is equipped with a retaining clip, focusing on primary resonance and subcritical dynamics. The nonlinear governing equations for the FGm pipe are derived by the extended Hamilton's principle, and subsequently discretized through the application of the Galerkin *** direct method of multi-scales is then used to solve the derived equations. A thorough analysis of various parameters, including the clip stiffness, the power-law index, the porosity, and the clip location, is conducted to gain a comprehensive understanding of the system's nonlinear dynamics. Through the analysis of the first natural frequency, the study highlights the influence of the flow velocity and the clip stiffness, while the comparisons with metallic pipes emphasize the role of FGm composition. The examination of the forced response curves reveals saddle-node bifurcations and their dependence on parameters such as the detuning parameter and the power-law index, offering valuable insights into the system's nonlinear resonant behavior. Furthermore, the frequency-response curves illustrate the hardening nonlinearities influenced by factors such as the porosity and the clip stiffness, revealing nuanced effects on the system response and resonance characteristics. This comprehensive analysis enhances the understanding of nonlinear behaviors in FGm porous pipes with a retaining clip, providing key insights for practical engineering applications in system design and optimization.

A survey on distributed compressed sensing： theory and applications

The compressed sensing （CS） theory makes sample rate relate to signal structure and content. CS samples and compresses the signal with far below Nyquist sampling frequency simultaneously. However, CS only considers the intra-signal correlations, without taking the correlations of the multi-signals into account. Distributed compressed sensing （DCS） is an extension of CS that takes advantage of both the inter- and intra-signal correlations, which is wildly used as a powerful method for the multi-signals sensing and compression in many fields. In this paper, the characteristics and related works of DCS are reviewed. The framework of DCS is introduced. As DCS＇s main portions, sparse representation, measurement matrix selection, and joint reconstruction are classified and summarized. The applications of DCS are also categorized and discussed. Finally, the conclusion remarks and the further research works are provided.

Effect of Preparation Technique on microstructure and Hydrogen Storage Properties of LaNi(3.8)Al(1.0)mn(0.2) Alloys

La Ni3.8Al1.0mn0.2 alloy was prepared by vacuum induction melting and *** effects of different preparation techniques of the as-cast,cast then annealed,as-spun and spun then annealed alloys on the microstructure and hydrogen storage properties were *** results indicated that the non-Ca Cu5 phases in the alloy became tinier and more dispersive after annealing or melt-spinning compared to those of the as-cast *** in the spun then annealed alloy,the non-Ca Cu5 phases disappeared and only a single-phase with Ca Cu5 type structure was *** all the alloys,the cell volume was increased in an order of as-cast 〈 spun then annealed 〈 cast then annealed 〈 as-spun,and the change of plateau pressure showed the opposite trend with that of the cell *** plateau could be flattened after melt-spinning or annealing,and the spun then annealed alloy showed the minimum plateau *** absorption kinetics of the alloy was promoted after melt-spinning or *** is suggested that the change in cell volume and compositional homogeneity resulting from different preparation techniques contribute to the difference of the hydrogen storage properties of the investigated alloys.

LiBH_(4) containing 18.5 wt.%H_(2) is an attractive high-capacity hydrogen storage material,however,it suffers from high operation temperature and poor ***,a novel and low-cost bifunctional additive,waxberry-like Fe_(3)O_(4) secondary nanospheres assembled from ultrafine primary Fe_(3)O_(4) nanoparticles,is synthesized,which exhibits significant destabilization and bidirectional catalyzation towards(de)hydrogenation of LiBH_(4).With an optimized addition of 30 wt.% waxberry-like Fe_(3)O_(4),the system initiated dehydrogenation below 100℃ and released a total of 8.1 wt.%H_(2) to 400℃.After 10 cycles,a capacity retention of 70% was achieved,greatly superior to previously reported oxides-modified *** destabilizing and catalyzing mechanisms of waxberry-like Fe_(3)O_(4) on LiBH_(4) were systematically analyzed by phase and microstructural evolutions during dehydrogenation and hydrogenation cycling as well as density functional theory(DFT)*** present work provides new insights in developing advanced nano-additives with unique structural and multifunctional designs towards LiBH4 hydrogen storage.

A Boundary Condition-Implemented Immersed Boundary-Lattice Boltzmann method and Its Application for Simulation of Flows Around a Circular Cylinder

A boundary condition-implemented immersed boundary-lattice Boltzmann method(IB-LBm)is presented in this *** present approach is an improvement to the conventional *** the conventional IB-LBm,the no-slip boundary condition is only approximately *** a result,there is flow penetration to the solid *** drawback of conventional IB-LBm is the use of Dirac delta function interpolation,which only has the first order of *** this work,the no-slip boundary condition is directly implemented,and used to correct the velocity at two adjacent mesh points from both sides of the boundary *** velocity correction is made through the second-order polynomial interpolation rather than the first-order delta function ***,the two drawbacks of conventional IB-LBm are removed in the present *** important contribution of this paper is to present a simple way to compute the hydrodynamic forces on the boundary from Newton’s second *** validate the proposed method,the two-dimensional vortex decaying problem and incompressible flow over a circular cylinder are *** shown in the present results,the flow penetration problem is eliminated,and the obtained results compare very well with available data in the literature.
