本研究的目的是确定甲基cellosolve(mC)二甘醇(DEG)、甘醇(EG)和1,2-丙三醇(1、2Propanediol PG)对冷冻的牛体外受精胚胎在支持培养基中进行体外培养和直接移植对其直接用水气是否有益的作用。仅将具有致密卵丘且用已获能精子受精的牛卵母细胞在添加有5％超排牛血清(SCS)中,培养20—22小时(38.5℃,5％CO_2)。7—8日龄胚胎在1.3m甲基cellosolve(mC),1.1m二甘醇(DEG),1.8m甘醇(EG),1.6m丙三醇(PG)和1.1m buthylene glycol(BG)溶液中平衡10分钟,再装入0.25毫升塑料细管中,于0℃放进酒精冷冻机中,然后以-1℃/分钟从0℃降至-6℃植冰,保持10分钟,再以-0.3℃/分或-0.5℃/分降到-30℃。在将塑料管入液氮中保存。在30℃水浴中解冻后,把胚胎放入TCm-199中(内含5％SCS)培养48小时,进行胚胎的直接再水化。解冻后发育至较晚阶段且形态好的胚胎被认为是存活的。将放入每种冷冻保护剂中的某些胚胎在不去掉冷冻保护剂的情况下进行非手术移植,在冷冻和直接再水化期间,对于保护孵化胚胎的浓度来说1.8m EG(64.3％)和1.3m mC(54.5％),1.1m DEG(60.0％)要优于其他浓空。胚胎冷冻的速度中(0.3℃/分和0.5℃/分)没有明显差异(P≤0.05),但是以0.3℃降温时,体外培养后形成孵化胚胎的比率为64.6％(31/48);而以0.

Short-fetch high waves during the passage of 2019 Typhoon Faxai over Tokyo Bay

The risk of wind waves in a bay is often overlooked,owing to the belief that peninsulas and islands will inhibit high ***,during the passage of a tropical cyclone,a semi-enclosed bay is exposed to twodirectional waves:one generated inside the bay and the other propagated from the outer *** Faxai in 2019 resulted in the worst coastal disaster in Tokyo Bay in the last few *** authors conducted a post-disaster survey immediately after this *** modeling was also performed to reveal the mechanisms of unusual high *** significant high-wave damage occurred on coasts facing the Pacific *** contrast,Fukuura-Yokohama,which faces Tokyo Bay,suffered overtopping waves that collapsed *** precisely reproduce multi-directional waves,the authors developed an extended parametric typhoon model,which was embedded in the JmA mesoscale meteorological model(JmA-mSm).The peak wave height was estimated to be 3.4 m off the coast of Fukuura,in which the contribution of the outer-sea waves was as low as 10%–20%.A fetchlimited wave developed over a short distance in the bay is considered the primary mechanism of the high *** maximum wave occurred on the left-hand side of the typhoon track in the bay,which appears to be contrary to the common understanding that it is safer within the semicircle of a storm than on the opposite *** Faxai was a small typhoon;however,if the radius was tripled,it is estimated that the wave height would exceed 3 m over the entire bay and surpass 4 m off the coasts of Yokohama and Chiba.

Features of Boiling on an Artificial Surface—Bubble Formation, Wall Temperature Fluctuation and Nucleation Site Interaction

The artificial surfaces are applied to study the pool boiling features, including the bubble behaviors, the surface temperature fluctuation, the heat transfer characteristics and nucleate site interaction. Three sets of experiments are carried out to investigate the influences of cavity shape, cavity size, cavity spacing on the boiling phenomena. Experimental results reveal that bubbling from the cylindrical as well as reentrant cavity is generally stable. The influence of cavity diameter on the bubble behaviors and the temperature fluctuation seems very weak while the effect of cavity depth cannot be neglected. As for the two cavity conditions, the bubble behaviors show the different features depending on the dimensionless cavity spacing. Three significant factors (thermal interaction, hydraulic interaction, bubble coalescence) control the nucleation site interaction, and the competition and dominance of the factors yield four interaction regimes.

Hydrophobic Effect in a Continuum model of the Lipid Bilayer

We study a continuum paradigm of the lipid bilayer based on minimizing the free energy of a mixture of water and lipid *** paper extends previous work of Blom and Peletier[European ***.,15(2004),pp.487-508]in the following ways.(a)It formulates a more general model of the hydrophobic effect to facilitate connections with microscale simulations and first-principles analysis.(b)It clarifies the meaning and role of the model parameters.(c)It outlines a method for determining parameter values so that physically-realistic bilayer density profiles can be obtained,for example for use in macroscale ***(a)-(c)suggest that the model has potential to robustly connect some micro-and macroscale levels of multiscale blood flow *** mathematical modelling in point(a)is based upon a consideration of the underlying physics of inter-molecular *** governing equations thus obtained are minimized by gradient flows via a novel numerical approach;this enables point(b).The numerical results are shown to behave physically in terms of the effect of background concentration,in contrast to the earlier model which is shown here to not display the expected behaviour.A“short-tail”approximation of the lipid molecules also gives an analytical tool which yields critical values of some parameters under certain ***(c)involves the first quantitative comparison of the numerical data with physical experimental results.

microscopic level interaction between fusion-peptides and lipid bilayer membranes plays a crucial role in membrane fusion,a key step of viral *** this paper,we use coarse-grained molecular dynamics(CGmD)simulations to study the interaction between hemagglutinin fusion-peptides and phospholipid bilayer *** CGmD,we are able to simulate the interaction of fusion peptides with a relatively large piece of membrane for a sufficiently long time period,which is necessary for a detailed understanding of the fusion process.A conformation of the peptide with a kink at the level of phosphate group is obtained,consistent with NmR and EPR *** results show that the N-terminal segment of the peptide inserts more deeply into the membrane bilayer compared to the C-terminal segment,as observed in previous *** simulations also show that the presence of fusion peptides inside the membrane may cause bilayer thinning and lipid molecule ***,our results reveal that peptides tend to aggregate,indicating cluster formation as seen in many experiments.

High Precision Ultrasonic Guided Wave Technique for Inspection of Power Transmission Line

Due to the merits of high inspection speed and long detecting distance, Ultrasonic Guided Wave（UGW） method has been commonly applied to the on-line maintenance of power transmission line. However, the guided wave propagation in this structure is very complicated, leading to the unfavorable defect localization accuracy. Aiming at this situation, a high precision UGW technique for inspection of local surface defect in power transmission line is proposed. The technique is realized by adopting a novel segmental piezoelectric ring transducer and transducer mounting scheme, combining with the comprehensive characterization of wave propagation and circumferential defect positioning with multiple piezoelectric elements. Firstly, the propagation path of guided waves in the multi-wires of transmission line under the proposed technique condition is investigated experimentally. Next, the wave velocities are calculated by dispersion curves and experiment test respectively, and from comparing of the two results, the guided wave mode propagated in transmission line is confirmed to be F（1,1） mode. Finally, the axial and circumferential positioning of local defective wires in transmission line are both achieved, by using multiple piezoelectric elements to surround the stands and send elastic waves into every single wire. The proposed research can play a role of guiding the development of highly effective UGW method and detecting system for multi-wire transmission line.

Phenomenological aspects of possible vacua of a neutrino flavor model

We discuss a supersymmetric model with discrete flavor symmetry A4×Z3. The additional scalar fields which contribute masses of leptons in the Yukawa terms are introduced in this model. We analyze their scalar potential and find that they have various vacuum structures. We show the relations among 24 different vacua and classify them into two types. We derive expressions of the lepton mixing angles, Dirac CP violating phase and majorana phases for the two types. The model parameters which are allowed by the experimental data of the lepton mixing angles are different for each type. We also study the constraints on the model parameters which are related to majorana phases. The different allowed regions of the model parameters for the two types are shown numerically for a given region of two combinations of the CP violating phases.
