Enhancement in antinociceptive and anti-inflammatory effects of tramadol by transdermal proniosome gel

Oral therapy of tramadol,an opiate analgesic,undergoes extensive hepatic metabolism and requires frequent *** therapy by virtue can overcome these issues and can improve the efficacy and reduce abuse liability of *** aim of this research was to investigate the possibility of transdermal delivery of tramadol by formulating proniosome gel and evaluate its therapeutic potential in *** effect of formulation composition as well as amount of drug on physicochemical characteristics of prepared proniosomes were *** proniosome gel(F4)was selected and evaluated for drug release,stability and transdermal efficacy by ex vivo and in vivo *** vesicles demonstrated optimal properties including spherical shape,nanosize with good entrapment efficiency,adequate zeta potential,higher stability and greater transdermal *** amorphization and dispersion of tramadol in the aqueous core of proniosome vesicles was confirmed by differential scanning *** profile of F4 was distinct(Pm control and displayed steady and prolonged tramadol release by Fickian *** therapy of F4 showed prominent reduction of induced twitches(Pmice and edema(Pmpared to oral *** improvement in clinical efficacy of tramadol in transdermal therapy is correlated with the pharmacokinetic data *** conclusion,the observed improvement in antinociceptive and anti-inflammatory effects from proniosome carriers signifies its potential to be a suitable alternative to oral therapy of tramadol with greater efficacy.

Peripheral nerve injury impairs motor, sensory, and autonomic function, incurring substantial financial costs and diminished quality of life. For large nerve gaps, proximal lesions, or chronic nerve injury, the prognosis for recovery is particularly poor, even with autografts, the current gold standard for treating small to moderate nerve gaps. In vivo elongation of intact proximal stumps towards the injured distal stumps of severed peripheral nerves may offer a promising new strategy to treat nerve injury. This review describes several nerve lengthening strategies, including a novel internal fixator device that enables rapid and distal reconnection of proximal and distal nerve stumps.

In vitro and in vivo bactericidal activity of Vitex negundo leaf extract against diverse multidrug resistant enteric bacterial pathogens

Objective:To investigate in vitro and in vivo antibacterial potentials of Vitex negundo(V. negundo) leaf extracts against diverse enteric ***:Water and methanol extracts of *** leaves were evaluated against enteric bacterial pathogens by using standard disc diffusion,viable bacterial cell count methods,determination of minimum inhibitory concentrations(mIC) and minimum bactericidal concentrations(mBC).Results:methanol extract of *** leaves showed potent antibacterial activity(inhibition zone:9.9- 22.6 mm,mIC: 200-3 200μg/mL,mBC:200-6 400μg/mL) against all the pathogenic enleric bacteria(Vibrio cholerae,Vibrio parahaemolyticus,Vibrio mimicus,Echerichia *** spps.,and Aeromonas spps) *** extract of *** leaves showed potent bactericidal activity both in vitro laboratory conditions(mBC,200-400μg/mL) and in the intestinal environment(Dose,1-2 mg/mL) of infant mice against pathogenic Vibrio cholerae,the major causative agent of cholera. Furthermore,assays using the mice cholera model showed that *** methanol extract can protect mice from Vibrio cholerae infection and significantly decrease the mortality rate(Pme we showed that methanol extract of *** leaves exhibited strong vibriocidal activity both in vitro and in vivo ***,it will be useful to identify and isolate the active compounds of this extract that could be a good alternative of antibiotics to treat cholera.

Vector control schemes have recently been used to drive linear induction motors(LIm)in high-performance *** trend promotes the development of precise and efficient control schemes for individual *** research aims to present a novel framework for speed and thrust force control of LIm using space vector pulse width modulation(SVPWm)*** framework under consideration is developed in four *** begin,mATLAB Simulink was used to develop a detailed mathematical and electromechanical *** research presents a modified SVPWm inverter control *** tuning the proportional-integral(PI)controller with a transfer function,optimized values for the PI controller are *** the subsystems mentioned above are integrated to create a robust simulation of the LIm’s precise speed and thrust force control *** reference speed values were chosen to evaluate the performance of the respective system,and the developed system’s response was verified using various data *** the low-speed range,a reference value of 10m/s is used,while a reference value of 100 m/s is used for the high-speed *** speed output response indicates that themotor reached reference speed in amatter of seconds,as the delay time is between 8 and 10 *** maximum amplitude of thrust achieved is less than 400N,demonstrating the controller’s capability to control a high-speed LIm with minimal thrust *** to the controlled speed range,the developed system is highly recommended for low-speed and high-speed and heavy-duty traction applications.

Atrial fibrillation(AF)is the most common form of cardiac arrhythmia,affecting 3 to 6 million people in the US,with an increased incidence in women and elderly,with its prevalence estimated to increase from 5.1 million in 2000 to 15.9 million in 2050 in the US population.[1]Age,hypertension,heart failure,myocardial infarction(mI),genetic factors,and associated systemic inflammatory states are well-established risk factors for the development of AF.[2–6]Lifestyle factors including alcohol consumption,smoking,obesity,sedentary lifestyle,and vigorous physical activity have also been implicated to contribute to the development of AF.[2,7–9]Studies suggest that AF drives up medical expenditure.[10]The European Society of Cardiology,based on the duration,responsiveness to treatment,and therapeutic attitude of the patient and physician,classifies AF into 4 types;when AF occurs and terminates by itself or by intervention within 7 days of onset,it is called paroxysmal AF;if sustained for more than a week,with or without intervention.

Positron-Acoustic Shock Waves in a Degenerate multi-Component Plasma

A theoretical investigation on the propagation of positron-acoustic shock waves （PASWs） in an unmagnetized, collisionless, dense plasma （containing non-relativistic inertial cold positrons, non-relativistic or ultra-relativistic degenerate electron and hot positron fluids and nondegenerate positively charged immobile ions） is carried out by employing the reductive perturbation method. The Burgers equation and its stationary shock wave solution are derived and numerically analyzed. It is observed that the relativistic effect （i.e., the presence of non/ultra- relativistic electrons and positrons） and the plasma particle number densities play vital roles in the propagation of PASWs. The implications of our results in space and interstellar compact objects including non-rotating white dwarfs, neutron stars, etc. are briefly discussed.

Fast and sensitive LC–mS/mS method for the simultaneous determination of lisinopril and hydrochlorothiazide in human plasma

A sensitive and rapid liquid chromatography-tandem mass spectrometry(LC–mS/mS) method has been developed for the simultaneous determination of lisinopril(LIS) and hydrochlorothiazide(HCTZ) in human plasma using their labeled internal standards(ISs). Sample pre-treatment involved solid phase extraction on Waters Oasis HLB cartridges using 100 μL of plasma, followed by liquid chromatography on Hypersil Gold C_(18)(50 mm×3.0 mm, 5 μm) column. The analytes were eluted within 2.0 min using acetonitrile-5.0 m m ammonium formate, p H 4.5(85:15, v/v) as the mobile phase. The analytes and ISs were analyzed in the negative ionization mode and quantified using multiple reaction monitoring. The method showed excellent linearity over the concentration range of 0.50–250.0 ng/m L for both the analytes. The intra-batch and inter-batch precision(% CV) was ≤5.26% and their extraction recoveries were in the range of 96.6%–103.1%. matrix effect evaluated in terms of IS-normalized matrix factors ranged from 0.97 to 1.03 for both the analytes. The validated method was successfully applied to determine the plasma concentration of the drugs using 10 mg lisinopril and 12.5 mg hydrochlorothiazide fixed dose formulation in 18 healthy Indian volunteers.
