Osteoarthritis(OA)is one of the most common chronic diseases in the ***,current treatment modalities mainly relieve pain and inhibit cartilage degradation,but do not promote cartilage *** this study,we show that G protein-coupled receptor class C group 5 member B(GPRC5B),an orphan G-protein-couple receptor,not only inhibits cartilage degradation,but also increases cartilage regeneration and thereby is protective against *** observed that Gprc5b deficient chondrocytes had an upregulation of cartilage catabolic gene expression,along with downregulation of anabolic genes in ***,mice deficient in Gprc5b displayed a more severe OA phenotype in the destabilization of the medial meniscus(Dmm)induced OA mouse model,with upregulation of cartilage catabolic factors and downregulation of anabolic factors,consistent with our in vitro *** of Gprc5b by lentiviral vectors alleviated the cartilage degeneration in Dmm-induced OA mouse model by inhibiting cartilage degradation and promoting *** also assessed the molecular mechanisms downstream of Gprc5b that may mediate these observed effects and identify the role of protein kinase B(AKT)-mammalian target of rapamycin(m TOR)-autophagy signaling ***,we demonstrate an integral role of GPRC5B in OA pathogenesis,and activation of GPRC5B has the potential in preventing the progression of OA.

In the present work,we studied dissolution behavior of three glass ceramics samples each having 34 SiO2–14.5 P2O5–1 CaF2–0.5 mgF2(%wt)and ratio of CaO/mgO varying from 11.5:1 to 1:11.5 in conventional SBF(SBF-K9) and revised SBF(r-SBF) that has ionic concentration exactly equal to that of human blood *** that purpose,samples were immersed in fluids for different time periods upto 25 *** film XRD analysis revealed the diffusive nature of the phases on the surfaces of samples after soaking for different time periods in *** showed the poor precipitation and small thickness of the HCAp layer on the samples as compared to that in SBF-K9,thus indicating the fitness and sensitivity of r-SBF for in-vitro characterization of ***,FTIR and EDS revealed slow bonding rate on the surfaces of the samples in r-SBF than that in SBF-K9 that showed the dependence of bond formation on the composition of the materials as well as on the physiological fluid used for in-vitro *** rate of HCAp formation was slower in r-SBF due to more competitive adsorption of CO3 ions to Ca and mg ions owing to greater amount of CO3 in r-SBF than that in *** shows the importance of CO3 content in the physiological fluids for the in-vitro assessment of ***,r-SBF is recommended to be used for assessment of samples to clearly understand their behavior in-vivo.

The putative Jambil meta-carbonatites of Swat,northern Pakistan,occur as discrete intrusions into the Proterozoic manglaur Formation,which are difficult to be distinguished from nearby calc-silicate marble because both rock types experienced regional metamorphism during Himalayan orogenesis that resulted in similar mosaic textures and mineral *** are often significant repositories of economic mineral resources and,therefore,are important to be distinguished from calc-silicate *** present new geochemical and geochronology data to distinguish between the two rock types and interpret the petrogenesis and tectonic evolution of the Jambil *** rock chemical data from the Jambil meta-carbonatites show characteristically high rare earth element(REE),Sr contents and lack of negative Eu anomaly,consistent with average calcio-carbonatite values worldwide and an igneous *** than 0.5 wt.% SrO in the metacarbonatites and SrO> 0.15 wt.% in constituent rock forming calcite are discriminating signatures of the Jambil ***,the Jambil meta-carbonatites are relatively depleted in Rb,Nb,Ta,Ti,Zr and Hf,relatively enriched in Ba,Th,Sr,and have a high LREE/HREE ratio when normalized to primitive *** carbon and oxygen isotope compositions vary from-3.5‰ to-4.3‰and from 9.7‰ to 12.3‰,*** geochemical characteristics indicate generation of the carbonatites through small degree of partial melting from a carbonated eclogitic ***-situ,U/Pb analysis of titanite indicates that the Jambil meta-carbonatites were emplacement at 438 ±3 *** combined with regional geological observations,we interpret the emplacement of the Jambil metacarbonatites to have taken place during the Silurian back arc extension within greater Gondwana and mark a transition from a compressional tectonic regime,brought about by collision of microcontinental blocks along the northern margin of Gondwana,to post-orogenic

Hindukush is an active subduction zone where at least one earthquake occurs on daily *** seismic hazard studies,it is important to develop a local magnitude scale using the data of local seismic *** have computed local magnitude scale for Hindukush earthquakes using data from local network belonging to Center for Earthquake Studies(CES)for a period of three years,i.e.2015–2017.A total of 26,365 seismic records pertaining to 2,683 earthquakes with magnitude 2.0 and greater,was used with hypocentral distance less than 600 *** scale developed by using this data comes to be m_(L)=logA+0.929logr+*** magnitude determined through formulated relation was compared with that of standard relation for Southern California and relation developed by the same authors for local network for Northern *** was observed that Hindukush region has high attenuation as compared to that of Southern California and Northern Punjab which implies that Hindukush is tectonically more disturbed as compared to the said regions,hence,seismically more active as *** have calculated station correction factors for our *** correction factors do not show any pattern which probably owes to the geological and tectonic complexity of this *** deviation and variance of magnitude residuals for CES network determined using Hutton and Boore scale and scale developed in this study were compared,it showed that a variance reduction of 44.1%was *** of magnitude residuals for different distance ranges was almost zero which showed that our magnitude scale was stable for all distances up to 600 *** developed magnitude scale will help in homogenization of earthquake *** has been observed that b-value of CES catalog decreases when magnitude is calculated by using newly developed magnitude scale.

The separate spin evolution quantum hydrodynamics(SSE-QHD)model is used to investigate the energy behavior for ion acoustic waves in degenerate quantum *** results show that the energy flow speed decreases with spin polarization *** is also shown that it decreases with the increasing rate up to a certain range of wave number and then it goes to zero *** is observed that Bohm potential suppresses the energy flow *** is also noticed that the energy flow speed deviates from the group velocity even in the absence of Bohm potential ***,the contribution of of Bohm poential effect in spin polarized plasma reduces the extent of deviation.

Nitriding of aluminium alloy （***0.8） in nitrogen and nitrogen-helium mixture was carried out by using 100 Hz-pulsed DC glow discharge. Samples are treated for different durations, namely 4 h, 8 h and 12 h, in nitrogen plasma as well as in a mixture of nitrogen- helium plasma for the same processing duration of 4 h. All nitriding treatments are carried out at an input power of 100 W, filling pressure of 1 mbar and substrates temperature of 250~C. X-ray diffraction （XRD） results show an expansion in lattice spacing and consequently shift of diffraction peaks towards lower angle with the addition of helium in nitrogen plasma. Surface morphology of the treated sample is investigated by scanning electron microscopy （SEm） and energy dispersive X-ray analysis （EDX）. Vickers micro-hardness testing results show increases in surface hardness with processing duration as well as with the addition of helium in nitrogen plasmas. This increase of surface hardness may be attributed to the diffusion of nitrogen content in the surface layer generating internal stresses. It is observed that the addition of helium positively affects the nitriding of samples.

Application of an LC–mS/mS method for the analysis of amlodipine,valsartan and hydrochlorothiazide in polypill for a bioequivalence study

A sensitive and selective method has been proposed for the simultaneous determination of amlodipine(AmL),valsartan(VAL) and hydrochlorothiazide(HCTZ) in human plasma by liquid chromatography–tandem mass spectrometry(LC–mS/mS). The analytes and their deuterated analogs were quantitatively extracted from100 μL human plasma by solid phase extraction on Oasis HLB cartridges. The chromatographic separation of the analytes was achieved on a Chromolith RP18 e(100 mm × 4.6 mm) analytical column within 2.5 min. The resolution factor between AmL and VAL, AmL and HCTZ, and VAL and HCTZ was 2.9, 1.5 and 1.4, respectively,under isocratic conditions. The method was validated over a dynamic concentration range of 0.02–20.0 ng/m L for AmL, 5.00–10,000 ng/m L for VAL and 0.20–200 ng/m L for HCTZ. Ion-suppression/enhancement effects were investigated by post-column infusion technique. The mean IS-normalized matrix factors for AmL, VAL and HCTZ were 0.992, 0.994 and 0.998, respectively. The intra-batch and inter-batch precision(% CV) across quality control levels was ≤ 5.56% and the recovery was in the range of 93.4%–99.6% for all the analytes. The method was successfully applied to a bioequivalence study of 5 mg AmL + 160 mg VAL + 12.5 mg HCTZ tablet formulation(test and reference) in 18 healthy Indian males under fasting. The mean log-transformed ratios of C max, AUC0–120 h and AUC0-inf and their 90% CIs were within 90.2%–102.1%. The assay reproducibility was demonstrated by reanalysis of 90 incurred samples.

In this work, the excess water-stagnation issue in the high current region in direct methanol fuel cells(DmFCs) is resolved by using atomic precision modulated nitrogen-crafted graphene(NG) in the cathode microporous layer by utilizing simplistic,industrial-expansive and ecological strategy. Few-layer 2D-graphene(~2–5 nm thickness) is prepared by bath sonication approach from abundant feedstock-graphite and is treated with nitric acid to yield 1.8 wt.% uniformly dispersed nitrogen containing NG. Specifically, 1:4 weight ratio NG:carbon-black(CB) hybrid architecture, displays 0.252 V in 370 mA cm^(-2) with the peak power density of 93.4 mW cm^(-2), improving cell power density by 45.6% compared with standard one at 60℃ and 1 mol/L methanol/oxygen conditions at ultra-low catalyst loadings and displaying exceptional stability. Atomic insights into NG reveal that interplay between bonding configurations, altered hydrophobic/hydrophilic porosity of graphene(10.6% less wettability from contact angle and 13.1% high electrode porosity measurements) contribute to the better mass-transport-porogenic effect(16.3% high oxygen-permeability), mildly affecting the electron pathway(6.5% reduced in-plane electrical conductivity),overall significantly improving cell performance. Altogether, this work delivers multiple advantages, i.e., the usage of material from facile, sustainable and cost-effective routes, while improving DmFC performance with potential industrial promise.

The Kohistan Terrane in N. Pakistan straddles the suture zone between the collided Indian and Eurasian plates in the NW Himalayas of N. Pakistan. Initiated as an intra\|oceanic island\|arc crust in Neotethys, the terrane accreted with the southern margin of the Karakoram Plate in the Late Cretaceous (about 90ma). Continued subduction at its southern margin resulted in Andean\|type magmatism and tectonics, until collision with the Indian Plat in the Early *** much of the central and eastern Kohistan expose Cretaceous volcanic and sedimentary rocks of intra\|oceanic island arc setting, the western Kohistan in Swat, Dir, Chitral and upper Ghizer valleys preserves remnants of the Late Paleocene—Early Eocene volcanism and sedimentation of Andean\|type setting. These volcano\|sedimentary lithologies in western Kohistan are geographically divisible into two, separated by the Lowari Pluton; (1) Dir\|Swat Belt, (2) Drosh\|Shamran Belt. Late Paleocene\|Early Eocene siliceous volcanics termed Utror Volcanics and Shamran Volcanics are common in the two belts and so are the plutons intrusive into these volcanics (Late Stage\|II of the Kohistan Batholith). The difference is in the sedimentary successions presented in the two belts. Whereas in the Dir\|Swat belt, the sedimentary succession comprises Baraul Banda Slate Formation, deposited in deep\|water fore\|arc setting and contemporaneous in age with that of the nearby Utror Volcanic Formation (about 55～60ma), the sediments in the Drosh\|Shamran belt are distinctly red beds of fluvial origin (the Purit Formation) those overlie unconformably on top of the Shamran Volcanics and their equivalents.
