Spastin and alsin protein interactome analyses begin to reveal key canonical pathways and suggest novel druggable targets

Developing effective and long-term treatment strategies for rare and complex neurodegenerative diseases is challenging. One of the major roadblocks is the extensive heterogeneity among patients. this hinders understanding the underlying disease-causing mechanisms and building solutions that have implications for a broad spectrum of patients. One potential solution is to develop personalized medicine approaches based on strategies that target the most prevalent cellular events that are perturbed in patients. Especially in patients with a known genetic mutation, it may be possible to understand how these mutations contribute to problems that lead to neurodegeneration. Protein–protein interaction analyses offer great advantages for revealing how proteins interact, which cellular events are primarily involved in these interactions, and how they become affected when key genes are mutated in patients. this line of investigation also suggests novel druggable targets for patients with different mutations. Here, we focus on alsin and spastin, two proteins that are identified as “causative” for amyotrophic lateral sclerosis and hereditary spastic paraplegia, respectively, when mutated. Our review analyzes the protein interactome for alsin and spastin, the canonical pathways that are primarily important for each protein domain, as well as compounds that are either Food and Drug Administration–approved or are in active clinical trials concerning the affected cellular pathways. this line of research begins to pave the way for personalized medicine approaches that are desperately needed for rare neurodegenerative diseases that are complex and heterogeneous.

Dirac r-matrix calculations of photoionization cross sections of Ni Ⅻ and atomic structure data of Ni 

We performed r-matrix calculations for photoionization cross sections of the two ground state configuration 3s^23p^5（~2P_（3/2,1/2）~o） levels and 12 excited states of Ni Ⅻ using relativistic Dirac Atomic r-matrix Codes（DArC） across the photon energy range between the ionizations thresholds of the corresponding states and well above the thresholds of the last level of the Ni XIII target ion. Generally, a good agreement is obtained between our results and the earlier theoretical photoionization cross sections. moreover, we have used two independent fully relativistic GrASP and FAC codes to calculate fine-structure energy levels, wavelengths, oscillator strengths, transitions rates among the lowest 48 levels belonging to the configuration（3s^23p^4, 3s3p^5, 3p^6, 3s^23p^33d） in Ni XIII. Additionally, radiative lifetimes of all the excited states of Ni XIII are presented. Our results of the atomic structure of Ni XIII show good agreement with other theoretical and experimental results available in the literature. A good agreement is found between our calculated lifetimes and the experimental *** present results are useful for plasma diagnostic of fusion and astrophysical plasmas.

An ab initio investigation of vibrational, thermodynamic, and optical properties of Sc_2AlC mAX compound

the structural vibrational, thermodynamical, and optical properties of potentially technologically important, weakly coupled mAX compound, Sc2 Al C are calculated using density functional theory(DFt). the structural properties of ScAlC are compared with the results reported earlier. the vibrational, thermodynamical, and optical properties are theoretically estimated for the first time. the phonon dispersion curve is calculated and the dynamical stability of this compound is investigated. the optical and acoustic modes are observed clearly. We calculate the Helmholtz free energy(F), internal energy(E), entropy(S), and specific heat capacity(Cv) from the phonon density of states. Various optical parameters are also calculated. the reflectance spectrum shows that this compound has the potential to be used as an efficient solar reflector.

Aims Understanding the factors that control biodiversity distributions at different spatial scales has been a key challenge for conservation *** biodiversity,reflected in differences in species compositions among sites(beta diversity),can be derived from species replacement(turnover)and is driven by multiple ***,we sought to tackle this issue through two questions related to threatened Brazilian seasonally dry forests:(i)what is the contribution of species turnover to beta diversity?and(ii)which factors drive variations in species compositions among forest patches?methods We sampled tree species and environmental variables(soils and climate)in 17 dry forest patches spaced almost 300 km *** used the beta diversity partitioning framework to determine the contribution of *** used redundancy analysis,with properly controlled spatial structure,to assess the contributions of the environmental and spatial factors to the variations of the species *** Findings Beta diversity among the patches was mainly represented by the turnover component(98.2%),with Simpson dissimilarity superior to other regions of the country(means of 0.89 and 0.71 in multiple site and pairwise measures,respectively).the environmental factors measured explained more than space,representing 30.3%of the variation of the species composition,of which 28.1%was nonspatially *** suggest that 300 km represents a threshold at which edaphic and climatic predictors have similar effects in determining community turnover(14.9%and 12.6%,respectively,without spatial structure).thus,conservation strategies should be considered across landscapes to effectively protect tropical forest diversity,as even considering the different climatic aspects covered by the scale,landscaped edaphic varieties are important drivers of species turnover.

A framework for analysis of extended fuzzy logic

We address a framework for the analysis of extended fuzzy logic(FLe) and elaborate mainly the key characteris-tics of FLe by proving several qualification theorems and proposing a new mathematical tool named the A-granule. Specifically, we reveal that within FLe a solution in the presence of incomplete information approaches the one gained by complete infor-mation. It is also proved that the answers and their validities have a structural isomorphism within the same context. this rela-tionship is then used to prove the representation theorem that addresses the rationality of FLe-based reasoning. As a conse-quence of the developed theoretical description of FLe, we assert that in order to solve a problem, having complete information is not a critical need; however, with more information, the answers achieved become more specific. Furthermore, reasoning based on FLe has the advantage of being computationally less expensive in the analysis of a given problem and is faster.
