Hormone-based pharmacotherapy for metabolic dysfunction-associated fatty liver disease

metabolic dysfunction-associated fatty liver disease(mAFLD)has reached epidemic proportions globally in parallel to the rising prevalence of *** its significant burden,there is no approved pharmacotherapy specifically tailored for this *** potential drug candidates for mAFLD have encountered setbacks in clinical trials,due to safety concerns or/and insufficient therapeutic ***,several investigational drugs that mimic the actions of endogenous metabolic hormones,including thyroid hormone receptorβ(THRβ)agonists,fibroblast growth factor 21(FGF21)analogues,and glucagon-like peptide-1 receptor agonists(GLP-1RAs),showed promising therapeutic efficacy and excellent safety *** them,resmetirom,a liver-targeted THRβ-selective agonist,has met the primary outcomes in alleviation of metabolic dysfunction-associated steatohepatitis(mASH),the advanced form of mAFLD,and liver fibrosis in phase-3 clinical *** hormone-based pharmacotherapies not only exhibit varied degrees of therapeutic efficacy in mitigating hepatic steatosis,inflammation and fibrosis,but also improve metabolic ***more,these three hormonal agonists/analogues act in a complementary manner to exert their pharmacological effects,suggesting their combined therapies may yield synergistic therapeutic *** in-depth studies on the intricate interplay among these metabolic hormones are imperative for the development of more efficacious combination therapies,enabling precision management of mAFLD and its associated comorbidities.

Role of Air Staging in a Batch-Type Fixed Bed Biomass Combustor under Constant Primary Air

Staged combustion of biomass is the most suitable thermo-chemical conversion for achieving lower gaseous emissions and higher fuel conversion *** a staged fixed bed combustion of biomass,combustion air is supplied in two *** the first stage,primary air is provided below the fuel,whereas in the later stage,secondary air is supplied in the freeboard *** available literature on the effects of air staging(secondary air location) at a constant primary air flow rate on combustion characteristics in a batch-type fixed bed combustor is limited and hence warrants further *** study resolves the effect of air staging,by varying the location of secondary air in the freeboard at five secondary to total air ratios in a batch-type fixed bed *** are reported for the effects of these controlled parameters on fuel conversion rate,overall gaseous emissions(CO_(2),CO and NO_x) and temperature *** fuel used throughout was densified hardwood *** show that a primary freeboard length(distance between fuel bed top and secondary air injection) of200 mm has higher fuel conversion rates and temperatures as well as lower CO emissions,at a secondary to total air ratio of 0.75 as compared to primary freeboard length of 300 ***,NO_x emissions were found to be lower for a primary freeboard length of 300 mm as compared to 200 *** increase in secondary to total air ratio from 0.33 to 0.75 resulted in higher freeboard temperatures and lower CO as well as NO_x *** outcomes of this study will be helpful in the effective design of commercial scale biomass combustors for more efficient and environmentally friendly combustion.

Grain refinement of mg-Ca alloys by native mgO particles

In mg-Ca alloys the grain refining mechanism,in particular regarding the role of nucleant substrates,remains the object of *** native mgO is being recognised as a nucleating substrate accounting for grain refinement of mg alloys,the possible interactions of mgO with alloying elements that may alter the nucleation potency have not been elucidated ***,we design casting experiments of mg-xCa alloys varied qualitatively in number density of native mgO,which are then comprehensively studied by advanced electron *** results show that grain refinement is enhanced as the particle number density of mgO *** native mgO particles are modified by interfacial layers due to the co-segregation of Ca and N solute atoms at the mgO/mg *** aberration-corrected scanning transmission electron microscopy and electron energy loss spectroscopy,we reveal the nature of these Ca/N interfacial layers at the atomic *** of the crystallographic termination of mgO,Ca and N co-segregate at the mgO/mg interface and occupy mg and O sites,respectively,forming an interfacial structure of a few atomic *** interfacial structure is slightly expanded,less ordered and defective compared to the mgO matrix due to compositional deviations,whereby the mgO substrate is altered as a poorer template to nucleate mg *** solidification in a TP-1 mould,the impotent mgO particles account for the grain refining mechanism,where they are suggested to participate into nucleation and grain initiation processes in an explosive *** work not only reveals the atomic engineering of a substrate through interfacial segregation but also demonstrates the effectiveness of a strategy whereby native mgO particles can be harnessed for grain refinement in mg-Ca alloys.

Polypropylene(PP)fibres have primarily used to control shrinkage cracks or mitigate explosive spalling in concrete structures exposed to fire or subjected to impact/blast loads,with limited investigations on capacity *** study unveils the possibility of using PP micro-fibres to improve the impact behaviour of fibre-reinforced ultra-high-performance concrete(FRUHPC)*** show that the addition of fibres significantly improves the impact behaviour of FRUHPC columns by shifting the failure mechanism from brittle shear to favourable flexural *** addition of steel or PP fibres affected the impact responses *** fibres considerably increased the peak impact force(up to 18%)while PP micro-fibres slightly increased the peak(3%-4%).FRUHPC significantly reduced the maximum midheight displacement by up to 30%(under 20impact)and substantially improved the displacement recovery by up to 100%(under 20impact).FRUHPC with steel fibres significantly improved the energy absorption while those with PP micro-fibres reduced the energy absorption,which is different from the effect of PP-macro fibre reported in the *** optimal fibre content for micro-PP fibres is 1%due to its minimal fibre usage and low peak and residual *** study highlights the potential of FRUHPC as a promising material for impact-resistant structures by creating a more favourable flexural failure mechanism,enhancing ductility and toughness under impact loading,and advancing the understanding of the role of fibres in structural performance.

BACKGROUND Unhealthy maternal diet leads to heavy metal exposures from the consumption of ultra-processed foods that may impact gene behavior across generations,creating conditions for the neurodevelopmental disorders known as autism and attention deficit/hyperactivity disorder(ADHD).Children with these disorders have difficulty metabolizing and excreting heavy metals from their bloodstream,and the severity of their symptoms correlates with the heavy metal levels measured in their *** may play a key role in helping parents reduce their ultra-processed food and dietary heavy metal intake by providing access to effective nutritional epigenetics *** To test the efficacy of nutritional epigenetics instruction in reducing parental ultra-processed food *** The study utilized a semi-randomized test and control group pretest-posttest pilot study design with participants recruited from parents having a learning-disabled child with autism or ***-two parents who met the inclusion criteria were randomly selected to serve in the test(n=11)or control(n=11)*** test group participated in the six-week online nutritional epigenetics tutorial,while the control group did *** efficacy of the nutritional epigenetics instruction was determined by measuring changes in parent diet and attitude using data derived from an online diet survey administered to the participants during the pre and post intervention *** intake scores were derived for both ultra-processed and whole/organic *** sample t-tests were conducted to determine any differences in mean diet scores within each *** There was a significant difference in the diet scores of the test group between the pre-and post-intervention *** parents in the test group significantly reduced their intake of ultra-processed foods with a preintervention diet score of 70(mean=5.385,SD=2.534)and a post-intervention diet score of 113(mean=8.692,SD=1.750)and the

The brain's extracellular matrix(ECm),which is comprised of protein and glycosaminoglycan(GAG)scaffolds,constitutes 20%-40% of the human brain and is considered one of the largest influencers on brain cell functioning(Soles et al.,2023).Synthesized by neural and glial cells,the brain's ECm regulates a myriad of homeostatic cellular processes,including neuronal plasticity and firing(miyata et al.,2012),cation buffering(moraws ki et al.,2015),and glia-neuron interactions(Anderson et al.,2016).Considering the diversity of functions,dynamic remodeling of the brain's ECm indicates that this understudied medium is an active participant in both normal physiology and neurological diseases.

Physics-constrained graph modeling for building thermal dynamics

In this paper,we propose a graph model embedded with compact physical equations for modeling the thermal dynamics of *** principles of heat flow across various components in the building,such as walls and doors,fit the message-passing strategy used by Graph Neural networks(GNNs).The proposed method is to represent the multi-zone building as a graph,in which only zones are considered as nodes,and any heat flow between zones is modeled as an edge based on prior knowledge of the building ***,the thermal dynamics of these components are described by compact models in the *** are further employed to train model parameters from collected *** model training,our proposed method enforces physical constraints(e.g.,zone sizes and connections)on model parameters and propagates the penalty in the loss function of *** constraints are essential to ensure model robustness and *** evaluate the effectiveness of the proposed modeling approach on a realistic dataset with multiple *** results demonstrate a satisfactory accuracy in the prediction of multi-zone ***,we illustrate that the new model can reliably learn hidden physical parameters with incomplete data.
