The Association between Researchers’Conceptions of Research and Their Strategic Research Agendas

Purpose:In studies of the research process,the association between how researchers conceptualize research and their strategic research agendas has been largely overlooked.This study aims to address this gap.Design/methodology/approach:This study analyzes this relationship using a dataset of more than 8,500 researchers across all scientific fields and the globe.It studies the associations between the dimensions of two inventories:the Conceptions of Research Inventory(CoRI)and the multi-Dimensional Research Agenda Inventory—Revised(mDRAI-R).Findings:The findings show a relatively strong association between researchers’conceptions of research and their research agendas.While all conceptions of research are positively related to scientific ambition,the findings are mixed regarding how the dimensions of the two inventories relate to one another,which is significant for those seeking to understand the knowledge production process better.Research limitations:The study relies on self-reported data,which always carries a risk of response bias.Practical implications:The findings provide a greater understanding of the inner workings of knowledge processes and indicate that the two inventories,whether used individually or in combination,may provide complementary analytical perspectives to research performance indicators.They may thus offer important insights for managers of research environments regarding how to assess the research culture,beliefs,and conceptualizations of individual researchers and research teams when designing strategies to promote specific institutional research focuses and strategies.Originality/value:To the best of the authors’knowledge,this is the first study to associate research agendas and conceptions of research.It is based on a large sample of researchers working worldwide and in all fields of knowledge,which ensures that the findings have a reasonable degree of generalizability to the global population of researchers.

This study compared three cryopreservation protocols on sperm functions, IVF outcomes, and embryo development. Epididymal spermatozoa cryopreserved using slow-cooling （18% w/v raffinose, RS-C） were compared with spermatozoa vitrified using 0.25 m sucrose （SV） or 18% w/v raffinose （RV）. The motility, vitality, and DNA damage （TUNEL assay） of fresh control （FC） spermatozoa were compared with post-thawed or warmed RS-C, RV, and SV samples. mouse oocytes （n = 267） were randomly assigned into three groups for insemination： RV （n = 102）, RS-C （n = 86）, and FC （n = 79）. The number and the proportion of two-cell embryos and blastocysts from each treatment were assessed. Sperm motility （P 〈 0.01） and vitality （P 〈 0.05） were significantly reduced after vitrification compared with slow-cooled spermatozoa. However, DNA fragmentation was significantly reduced in spermatozoa vitrified using sucrose （15 - 1.8% [SV] vs 26 - 2.8% [RV] and 27 - 1.2% [RS-C]; P 〈 0.01）. Although the number of two-cell embryos produced by RS-C, RV, and FC spermatozoa was not significantly different, the number of blastocysts produced from two-cell embryos using RV spermatozoa was significantly higher than FC spermatozoa （P = 0.0053）. This simple, small volume vitrification protocol and standard insemination method allows successful embryo production from small numbers of epididymal spermatozoa and may be applied clinically to circumvent the need for ICSI, which has the disadvantage of bypassing sperm selection.

Cuba has been leading marine protected area(mPA)designation in the Caribbean region to ensure conservation of its valuable marine ecosystems.Yet,an efficient monitoring program for mPAs is still to be implemented and will benefit from pre-existing information.The highly regulated mPA of Punta Francés National Park is one of the first Cuban mPAs and was established mainly to protect species and ecosystems for tourism purposes.monitoring of protection effects on commercial fish species was lacking in this mPA.This study aimed at increasing local scientific knowledge by providing a baseline study about the most commercially fished families(Haemulidae,Lutjanidae and Serranidae)around Punta Francés mPA.Data collected represent only a limited period but can be used as a base point to support future monitoring.Fish abundance in number and biomass,and size were collected to test for differences between inside and adjacent areas outside the Punta Francés mPA,in different coral reef types.The main result of this study was the significantly larger size and biomass of snappers and groupers outside the mPA where intense fisheries occur.An observation consistent with a large spawning aggregation was also recorded outside the mPA.Even with a limited set of data,these results suggest that,at least temporarily,the most targeted species[and sizes]are still highly vulnerable to fisheries.Relevant habitats in the adjacent area,that are apparently missing within the mPA,may support some of the results found.Extending the limits of the Punta Francés mPA to include those habitats outside may be critical to ensure the effectiveness of this mPA in contributing to protect the most commercial species of the region.This should be done together with efficient fisheries management measures in the region,such as the significantly increase of minimum legal sizes and temporal closures during spawning migrations.

The biological properties of therapeutic peptides,such as their pharmacokinetics and pharmacodynamics,are correlated with their structure and aggregation properties.Herein,we studied the aggregation properties of a therapeutic peptide(CIGB-814),currently in phase 2 clinical trial,for the treatment of rheumatoid arthritis over a wide range of concentrations(μm-mm).We applied spectroscopic techniques(fluorescence,circular dichro-ism,resonance,and dynamic light scattering),atomic force microscopy,and molecular dynamics simulations to determine the aggregation mechanism of CIGB-814.We found that the hierarchical aggregation of CIGB-814 at micromolar concentrations was initiated by the formation of peptide oligomers.Subsequently,the peptide oligomers trigger the nucleation and growth of peptide nanostructures(cac=123μm),ultimately leading to the fibrillization of CIGB-814(cac’=508μm).These results pave the way for a deeper understanding of the CIGB-814 therapeutic activity and may give important insights on its pharmacokinetics.

The complexity of multi-dimensional climate, environmental and human health information complicates bringing together scientists, civil society, and decision makers to provide adequate mitigation and adaptation options for the consequences of global environmental change. To provide an effective pathway to conciliate (integrate) these datasets, we present PULSE-Brazil as the Platform for Understanding Long-term Sustainability of Ecosystems and human health. The overarching aim of this paper is to focus on two study cases in the Brazilian state of Acre, 1) analyzing recent socio-environmental disasters such as the impacts of droughts and consequent increases in fire detections on the incidence of respiratory diseases, and 2) evaluating the impacts of temperature increases in combination with rainfall seasonality upon the incidence of Dengue fever. Based on data available in PULSE-Brazil platform, we also aim to provide insights on the consequences of future climate variability on ecosystems and human health. Finally, we provide a discussion on the collaborative process between scientists and policymakers that defined the PULSE-Brazil platform specifications and datasets and propose a conceptual pathway for promoting the interaction between science and policy during the process of producing a tool that allows the understating of climate-relate processes. Our results clearly demonstrated that ecosystems are under increased fire risk in the future that will, in turn, affect the health of human populations. moreover, humans are also exposed to critical Dengue fever outbreaks with the projected increase in minimum temperatures. Therefore, minimizing the impacts of these potentially ascending socio-environmental problems is the first step for adapting to a changing climate in the Amazon region. The perceptions of fishers towards the Arrábida marine Park,a marine protected area(mPA)in the west coast of Portugal,were studied through face-to-face interviews in two different moments of the mPA life cycle.Fishers'perceptions about the mPA and the impact it had on the fishing activity over time were identified just before the full implementation of the zoning and regulations of the management plan and 10 years later.This study aimed to investigate fishers'knowledge,acceptance and perceptions about the mPA changed with time,if support for the mPA was linked to the impact of the mPA on the fishing activity,and if fishers'perceptions about the impact of the mPA on the fishing activity match with local landings trends.Results show that although knowledge about the marine park significantly improved over time,fishers'acceptance did not.A decrease on fishers'support was not substantial but occurred.Issues such as the disagreement with regulations reinforced concerns raised during the implementation of the marine park,particularly in relation to the top-down decision-making,which commonly confers minor participation,recognition and legitimacy to fishers.Apparently,fisheries benefits were still not perceived by local fishers,though they are central for fishers'support.Further,the perceived negative impacts of the park seemed to be more related to social aspects and individual interests than to impacts on catches.Addressing adequate management,enforcement and participation of local fishers is still possible and are advocated here as to contribute to the expected socioecological outcomes and respective support,leading to the future successful performance of the Arrábida marine Park.Assessing fishers’perceptions towards an mPA over time is central and should be included on periodical socioecological monitoring and inform an effective adaptive management.

Forensic aesthetic facial damage—AFD expert’s valuation is a concerning matter in court as expressively vary in between experts. In fact, differences can be quite significant suggesting examiners age, sex and professional qualification may influence this valuation. Aiming to offer a systematic and more objective evaluation, some methods for assessing AFD had been proposed lately. Known as Aesthetic Impairment Impact Perception (AIPE), Cobo-Plana’s revised methodology (2010) was idealized to minimize the examiner’s subjectivity by means of answering an ordered sequence of questions related to the specific damage perception. This research aims to access AFD perception differences (simulated cases) in between Lawyers, Heath professionals (Physicians and Dental surgeons) and general professionals (laypeople) under AIPE methodology in order to allow a better comprehension of AFD when dealing with forensic expertise cases. Within Groups perceptions varied (p mportant” severity grade. Raters over 35 Years age scored higher AFD lesions, and rater’s sex (as an independent variable) did not express any significant difference in AFD scoring. The tendency of under-rating AFD lesions below very important severity grade was observed being a matter of major concern considered their prevalence and the effective consequences involved to the victim. These results recommend extra caution when performing AFD evaluation and consequently expertise reports, claiming a more judicious analysis based on consolidated standards and suggest, examiners to be more perceptive to physical damage evaluation in order to include social perceptions and reactions, mainly, victims feelings and constrains over their aesthetic loss.

Transcriptome analysis of the <i>Tityus serrulatus</i>scorpion venom gland

The Tityus serrulatus scorpion is considered the most dangerous scorpion in Brazil and is responsible for several cases of human envenomation annually. In this study, we performed transcriptome profiling of the T. serrulatus venom gland. In addition to transcripts with housekeeping functions, such as those related to protein synthesis, energy supply and structural processes, transcripts from thirty-five families of venom peptides or proteins were identified. These transcripts included three new complete sequences of toxins and more than a dozen putative venom gland proteins/peptides. The venom gland transcriptome profile was verified by comparison with the previously determined proteomic profile. In conclusion, this transcriptome data provides novel insights into the putative mechanisms underlying the venomous character of T. serrulatus. The collected data of scorpion transcripts and proteins/peptides described herein may be an important resource for identifying candidate targets of molecular therapies and preventative measures.

Curimat?-pioa (Prochilodus costatus) and curimat?-pacu (Prochilodus argenteus) are migratory fish species endemic to the S?o Francisco River Basin in Brazil. Both species play important roles in local fisheries and ecology in the Paraopeba River. A dam was recently constructed on this river and to help in the development and conservation programs, we characterized the genetic variation of both species before dam construction. Complex hypervariable repeats micro-satellite was used to asses genetic variation for both species within and between the five collection sites in order to detect population substructuring. Nucleotide substitutions and insertion/deletion polymorphisms (indels) resulted in 35 P. costatus haplotypes (sample size = 89) and 22 P. argenteus haplotypes (sample size = 32). Significant genetic diversity and population differentiation was detected between five sampling sites for both species. Therefore, each of the five sites should be regarded as a group comprising significant genetic differences in species conservation and maintenance plans. Comparing these results to genetic diversity measures after dam construction will be critical for future management in this region.
