This research was designed to analyze the possible associations of Arg389 Gly ADRB1 and Trp64 Arg ADRB3polymorphisms in children with obesity.A cross-sectional study included 1,046 school-age mexican participants（6-12 years old） from the cities of San Luis Potosi and Leon.Children were classified as non-obese or obese according to their body mass index（BmI） percentile;obese children had a BmI≥95th percentile for sex and age.Biochemical data were collected.Polymorphisms were detected using Taqman qPCR assay.A logistic regression analysis was used to calculate the risk of obesity based on genotypes.Differences were found between groups where obese children had a significant increase in systolic and diastolic blood pressure,fasting plasma glucose,insulin,HOmAIR,LDL-cholesterol,triglycerides,and lower HDL-cholesterol compared with the normal weight group（P 〈 0.05）.The distribution of allele frequency in the population was Arg = 87.4 and Gly = 12.6（Hardy Weinberg equilibrium x^2= 3.16,P = 0.07）;Trp = 81.5 and Arg= 18.5（Hardy Weinberg equilibrium x^2 = 2.2,P = 0.14） for ADRB1 and ADRB3,respectively.Even though no different frequencies of Arg389 Gly polymoiphism between groups were found（P = 0.08）,children carriers of one Gly389,ADRB1 allele had a risk for obesity of OR = 1.40（95%CI,1.03-1.90,P =0.03） after adjustment for age and gender.No other association was found for Trp64 Arg ADRB3 polymorphism.Only the Arg389 Gly ADRB1 polymorphism was associated with risk for obesity in mexican children.

Tentative Identification of Phytochemicals and Antioxidant Activities during Fruit-Ripening on Chamaedorea radicalis mart.

This work aims to determine the phytochemical characterization of the pericarp of Chamaedorea radicalis mart.fruit as a non-timber product with potential to obtain phytochemicals with potential applications in the industry.Fruit from C.radicalis were grouped in four ripening stages named as S1,S2,S3 and S4,according to maturity;S1 the most unripe stage and S4 the completely ripe stage.Determinations of total phenolic compounds,free radical scavenging activities and total flavonoid contents using spectrophotometric methods were done.Also,the tentative identification of phytochemicals during fruit ripening was done using UPLC-mS-mS.Total phenolic compound(TPC)content ranged from 7.24 to 12.53 mg gallic acid equivalents per gram of fresh weight(mg GAE/g FW).Total flavonoids(TF)contents ranged from 0.163 to 0.23 mg of quercetin equivalents per g FW(mg QE/g FW).Free radical scavenging activity against DPPH and ABTS radicals varied from 40.80 to 53.68 and from 22.29 to 37.76 mmol Trolox equivalents g FW(mmol TE/g FW),respectively.Antioxidant assay in vitro by FRAP(ferric reducing antioxidant power)method showed that S3 was the highest level with antioxidant power while S4 was the lowest with Red ripeness stage showed the lowest contents for all determinations.mass spectrometry allowed detection of 26 compounds,including phenolics,alkaloids and saponins.Afzelin,Kaempferol 3-neohesperidoside and the four saponins identified were present in all ripeness stages.Preliminary phytochemical identification and the spectrophotometric determinations showed that the pericarp of C.radicalis presented antioxidants and compounds related to alkaloids,phenolics and saponins.The presence and abundance of each phytochemical regarding each ripeness stage should be considered.

Serum vitamin D and colonic vitamin D receptor in inflammatory bowel disease

AIm: To determine serum vitamin D levels and colonic vitamin D receptor(VDR) expression in inflammatory bowel disease(IBD) and non-IBD patients and correlate these with histopathology.mETHODS: Puerto Rican IBD(n = 10) and non-IBD(n = 10) patients ≥ 21 years old scheduled for colonoscopy were recruited. Each patient completed a questionnaire and provided a serum sample and a colonic biopsy of normal-appearing mucosa. For IBD patients, an additional biopsy was collected from visually diseased mucosa. Serum vitamin D levels were measured by ultra-performance liquid chromatography and mass spectrometry. Hematoxylin and eosin stained tissue sections from colonic biopsies were classified histologically as normal or colitis(active/inactive), and scored for the degree of inflammation present(0-3, inactive/absent to severe). Tissue sections from colonic biopsies were also stained by immunohistochemistry for VDR, for which representative diagnostic areas were photographed and scored for staining intensity using a 4-point scale.RESULTS: The IBD cohort was significantly younger(40.40 ± 5.27, P min D deficiency(40% vs 20%, respectively) and insufficiency(70% vs 50%, respectively). Histologic inflammation was significantly higher in visually diseased mucosa from IBD patients(1.95 ± 0.25) than in normalappearing mucosa from control patients(0.25 ± 0.08, P m IBD patients(0.65 ± 0.36, P m IBD patients(r =-0.44, P m IBD patients with Crohn's disease(r =-0.69, P mal-appearing colonic tissue from control patients or IBD patients. Uncertainty Reduction Through Data management in the Development, Validation, Calibration, and Operation of a Hurricane Vulnerability model

Catastrophe models estimate risk at the intersection of hazard,exposure,and vulnerability.Each of these areas requires diverse sources of data,which are very often incomplete,inconsistent,or missing altogether.The poor quality of the data is a source of epistemic uncertainty,which affects the vulnerability models as well as the output of the catastrophe models.This article identifies the different sources of epistemic uncertainty in the data,and elaborates on strategies to reduce this uncertainty,in particular through identification,augmentation,and integration of the different types of data.The challenges are illustrated through the Florida Public Hurricane Loss model(FPHLm),which estimates insured losses on residential buildings caused by hurricane events in Florida.To define the input exposure,and for model development,calibration,and validation purposes,the FPHLm teams accessed three main sources of data:county tax appraiser databases,National Flood Insurance Protection(NFIP)portfolios,and wind insurance portfolios.The data from these different sources were reformatted and processed,and the insurance databases were separately cross-referenced at the county level with tax appraiser databases.The FPHLm hazard teams assigned estimates of natural hazard intensity measure to each insurance claim.These efforts produced an integrated and more complete set of building descriptors for each policy in the NFIP and wind portfolios.The article describes the impact of these uncertainty reductions on the development and validation of the vulnerability models,and suggests avenues for data improvement.Lessons learned should be of interest to professionals involved in disaster risk assessment and management.

AIm:To determine the expression of neurokinin-1receptor(NK-1R),phosphorylated epidermal growth factor receptor(p EGFR),cyclooxygenase-2(Cox-2),and vitamin D receptor(VDR)in normal,inflammatory bowel disease(IBD),and colorectal neoplasia tissues from Puerto Ricans.mETHODS:Tissues from patients with IBD,colitisassociated colorectal cancer(CAC),sporadic dysplasia,and sporadic colorectal cancer(CRC),as well as normal controls,were identified at several centers in Puerto Rico.Archival formalin-fixed,paraffin-embedded tissues were de-identified and processed by immunohistochemistry for NK-1R,p EGFR,Cox-2,and VDR.Pictures of representative areas of each tissues diagnosis were taken and scored by three observers using a4-point scale that assessed intensity of staining.Tissues with CAC were further analyzed by photographing representative areas of IBD and the different grades of dysplasia,in addition to the areas of cancer,within each tissue.Differences in the average age between the five patient groups were assessed with one-way analysis of variance and Tukey-Kramer multiple comparisons test.The mean scores for normal tissues and tissues with IBD,dysplasia,CRC,and CAC were calculatedand statistically compared using one-way analysis of variance and Dunnett’s multiple comparisons test.Correlations between protein expression patterns were analyzed with the Pearson’s product-moment correlation coefficient.Data are presented as mean±SE.RESULTS:On average,patients with IBD were younger(34.60±5.81)than normal(63.20±6.13,Pmal tissues(0.73±0.19).p EGFR was significantly increased in sporadic CRC(1.53±0.43)and CAC(2.25±0.47)when compared to normal tissue(0.07±0.25,Pmal tissues,P<0.05

metadoxine improves the three- and six-month survival rates in patients with severe alcoholic hepatitis

AIm:To evaluate the impact of metadoxine(mTD) on the 3- and 6-mo survival of patients with severe alcoholic hepatitis(AH).mETHODS:This study was an open-label clinical trial,performed at the"Hospital General de méxico,Dr.Eduardo Liceaga".We randomized 135 patients who met the criteria for severe AH into the following groups:35 patients received prednisone(PDN)40 mg/d,35patients received PDN+mTD 500 mg three times daily,33 patients received pentoxifylline(PTX)400 mg three times daily,and 32 patients received PTX+mTD 500 mg three times daily.The duration of the treatment for all of the groups was 30 d.RESULTS:In the groups treated with the mTD,thesurvival rate was higher at 3 mo(PTX+mTD 59.4%vs PTX 33.3%,P=0.04;PDN+mTD 68.6%vs PDN20%,P=0.0001)and at 6 mo(PTX+mTD 50%vs PTX18.2%,P=0.01;PDN+mTD 48.6%vs PDN 20%,P=0.003)than in the groups not treated with mTD.A relapse in alcohol intake was the primary independent factor predicting mortality at 6 mo.The patients receiving mTD maintained greater abstinence than those who did not receive it(74.5%vs 59.4%,P=0.02).CONCLUSION:mTD improves the 3-and 6-mo survival rates in patients with severe AH.Alcohol abstinence is a key factor for survival in these patients.The patients who received the combination therapy with mTD were more likely to maintain abstinence than those who received monotherapy with either PDN or PTX.

Electrospinning Polylactic Acid Polymer membranes as Biological Sieve for Yeast and Bacteria

Vaginal infections are suffered by millions of women of reproductive age. Recent research has shown that timely diagnosis of these infections avoids complications in the patient’s health. Since the symptoms of infections caused by yeast and bacteria are similar, the patient is not always treated properly. membranes with three different thicknesses (66 μm, 83 μm, 128 μm) were produced by electrospinning of polylactic polymer (PLA) and were used as a biological sieve to detect microorganisms present in vaginal infections, bacteria or yeasts. Electrospinning technique is a technique that allows the generation of membranes from nanofibers of material polymeric and therefore, obtaining porous structures. The size of the fibers was observed as well as the morphology and the size of the pores, using the scanning electron microscopy (SEm). The effectiveness of these membranes was proven by the cultivation of the bacteria that passed through the polymeric membrane and the yeasts retained by it. The membranes allowed the retention of yeasts of Candida albicans type with dimensions of 7 μm, which allowed the separation of Brevibacterium ravens purgense bacteria with dimensions of 1.4 μm, thus demonstrating their effectiveness as biological sieve.

In vitro antioxidant,antilipoxygenase and antimicrobial activities of extracts from seven climbing plants belonging to the Bignoniaceae

Objectives： This study aimed to evaluate the in vitro antioxidant capacity, to determine the anti-inflammatory effect due to lipoxygenase inhibition and to test the antimicrobial activity of ethanolic extracts from leaves of seven climbing species belonging to the Bignoniaceae family. These species are Adenocalyrnrna rnarginatum （Cham.） DC., Amphilophium vauthieri DC., Cuspidaria convoluta （VEIL） A. H. Gentry, Dolichandra dentata （K. Schum.） L. G. Lohmann, Fridericia caudigera （S. moore） L. G. Lohmann, Fridericia chica （Bonpl.） L. G. Lohmann and Tanaecium selloi （Spreng.） L. G. Lohmann. methods： The antioxidant activity was evaluated using three methods, 2,2＇-azino-bis（3-ethylbenzothiazo line-6-sulfonic acid）（ABTS）, 2,2-diphenyl-l-picrylhydrazyl （DPPH）, and ferric reducing antioxidant power. Lipoxygenase-inhibiting activity was assayed spectrophotometrically; the result was expressed as percent inhibition. The antimicrobial activity was assessed using the agar disk diffusion method. minimal inhibitory concentration （mIC） and minimal bactericidal]fungicidal concentration were also determined for each extract against 12 pathogenic bacterial strains of Staphylococcus aureus and seven fungal strains of the Candida genus. The identification of the major compounds present in the most promising extract was established by high-performance liquid chromatography-tandem mass spectrometry. Results： C convoluta, F. caudigera, and F. chica exhibited the best antioxidant activity by scavenging DPPH and ABTS~ radicals and reducing Fe3＋ ion. These extracts showed a notable inhibition of lipnxygenase. F. caudigera was found to have the lower mIC value against S. aureus strains and six Candida species. The extracts of F. caudigera and C. convoluta were active even against methicillin-resistant S. aureus. C convoluta had higher total phenol content, better antioxidant activity and superior anti- inflammatory and antimicrobial activity. BACKGROUND The implementation of nutritional strategies targeting several variables at once could benefit patients with cirrhosis.Non-alcoholic beer has different compounds that exert antioxidant,anti-inflammatory and nutritional properties.AIm To evaluate the effect of diet+exercise and non-alcoholic beer on nutritional status,endothelial function and quality of life in patients with cirrhosis.mETHODS In this randomized open clinical trial,patients with cirrhosis were randomized into two groups:The intervention(non-alcoholic beer+diet+exercise)and control(water+diet+exercise)group.Treatment consisted of 330 mL nonalcoholic beer/day or the same amount of water,plus an individualized dietary plan and an exercise program with a pedometer-based bracelet to reach at least 5000 steps/d and>2500 above the baseline during 8 wk.Endothelial function(flow-mediated dilation,plethysmography),biochemical and nutritional variables and quality of life(CLDQ)were evaluated.RESULTS Forty-three patients were included in the study,21 in the control group and 22 in the intervention group.The mean age was 53.5±7.8 years,60%were women,the median mELD score was 8(7-10)and most patients were Child-Pugh A(88%).Adherence to the interventions was>90%in both groups,there were no adverse events and all biochemical parameters remained stable in both groups.Endothelial function improved in both groups.All measured nutritional parameters improved in the intervention group,compared to only 2 in the control group and quality of life improved in both groups;however,more domains improved in the intervention group.CONCLUSION The intervention consisting of non-alcoholic beer,diet and exercise seems to be safe and well tolerated in patients with cirrhosis,and shows improvement in nutritional status,endothelial function,and quality of life.These results need to be further confirmed.

Solid Particle Erosion Behaviour of TiN Coating on AISI 4140 Steel

In this study, the performance against erosive wear of PVD TiN (titanium nitride) coating was evaluated using an erosion test rig similar to that described in the standard ASTm G76-95. This coating normally has various industrial applications such as tapping, drilling, dry machining and punching. Angular silicon carbide (SiC) was used as an abrasive particle with a grain size of 350-450 μm. Erosion tests were carried out using different incident angles, 30°, 45°, 60° and 90° with a particle velocity of 24 ± 2 m/s, an abrasive flow rate of 0.7 ± 0.5 g/min, and the test temperature was between 35°C and 40°C. The particle velocity and the abrasive flow rate were low in all of the tests to reduce the interaction between the incident and the rebounding particles in the system. The surfaces were examined with a scanning electron microscope (SEm) to characterize the erosive damage. The wear mechanisms identified were brittle fracture characterized by radial cracks on the surface by multiple impact and a few pits at 30°, while a few cracks and the formation of craters in random positions were observed at angles near or at 90°. Elliptical scars were observed at 30° and 45°, which are a characteristic feature when the specimens are impacted at low-incident angles (α ≤ 45°) whereas a roughly circular scar was seen at 60° and 90°. In addition, roughness variations were analyzed using atomic force microscopy (AFm), before and after the erosion tests, and the results exhibited an increase in the roughness as the TiN samples were impacted at angles near or at 90°.
