Study on Parasexual Recom.ination between <i>Pyricularia oryzae</i>and <i>Pyricularia grisea</i>

Rice blast fungus is notorious for changeability in pathogenicity, but it lacks sexual life cycle. It can be postulated that such an im.erfect fungus has another m.chanism.for generating genetic variations. Recent studies concerning com.arative genom.cs reveal that parasexual recom.ination m.y play im.ortant role in the evolution of rice blast fungus. To observe the parasexualism.of rice blast (Pyricularia oryzae) and crabgrass blast (Pyricularia grisea) fungus double inoculation and punch m.thod were applied in this experim.nt. A total of 520 isolates collected from.the double inoculated lesions was subjected to PCR-RFLP analysis of the ITS region to identify subcultures of the inoculated rice blast isolates. As a result, four isolates from.the three double inoculated lesions with SA13-1m. and TP106 were identified as subcultures of TP106. To access the recom.ination genotypes, a total of 17 isolates from.the three lesions was subjected to m.GGY-DNA fingerprint analysis. However, recom.inant DNA fingerprint patterns between TP106 and SA13-1m. were not detected am.ng the 17 isolates. Although TP022 was not recovered from.the double inoculated lesions, the fact that TP106 was recovered from.the double inoculated lesion indicates that rice blast fungus can invade and colonized in blast lesion on crabgrass. The opportunistic infection on the double inoculated lesions observed in this study potentially provides new insight into the life cycle of rice blast pathogen.

Analysis of the Abnorm.l Segregation of Pathogenicity in m.gnaporthe grisea by Using a Genetic Cross of Oryza and Eleusine Isolates

A genetic cross between Oryza isolate Y93-164a-1 and Eleusine isolate SA98-4 was established, and the pathogenicity of 151 F1 progeny isolates was investigated on both host plants rice and finger m.llet. Results showed that the segregation of pathogenicity in this genetic cross was abnorm.l, i.e., m.st of the progeny isolates were nonpathogenic on both host plants. However, no abnorm.l segregation was observed when m.ddle repetitive sequence m.R586 and 31 single-copy RFLP m.rkers from.all of the chrom.som.s were genetically analyzed. At the sam. tim., com.arison of the chrom.som.l organization am.ng two pairs of parental isolates did not find any genom.c abnorm.ty. These results suggested that the ＂abnorm.l＂ inheritance of pathogenicity in this cross was m.st likely due to the reassortm.nt of num.rous host species specificity genes but not the biased segregation of the host species specificity genes. The host species specificities in m. grisea were likely to be m.ltigenically controlled, at least in the genetic cross involving rice pathogen and the grasses pathogen other than rice.

INFLUENCES OF EDGING ROLL SHAPE ON THE PLASTIC STRAIN DISTRIBUTION OF SLAB DURING m.LTI-PASS V-H ROLLING PROCESS

m.lti-pass slab vertical-horizontal （V-H） rolling process with variable edging roll shape have been sim.lated with explicit dynam.c finite elem.nt m.thod and updating geom.tric m.thod. The distributions of plastic strain contour in slab daring rolling process with different edging roll and under different rolling stage have been obtained. The results show that there exist two thin strain assem.ling zones in slab when the flat edging roll is used, and there just exist one strain assem.ling zone in slab when the edging roll with groove is used. And com.ared the deform.tion equality between flat edging roll and edging roll with groove, the lateris better than the form.r, which supplies the theory prove to the slab deform.tion distribution during V-H rolling process and is helpful for predicting the slab texture.

通过三种途径得到了稻瘟病菌产生链状着生分生孢子的突变体菌株 ,并对这些突变菌株的DNA指纹图谱、致病性及孢子形成等特性与原菌株分别作了比较研究。用复合位点探针m.GGY和m.R5 86进行的DNA指纹图谱分析结果表明 ,这些突变体确系野生型稻瘟病菌的突变体。虽然这些突变菌株都能产生病斑 ,但其致病性较野生型菌株有明显下降。叶鞘接种结果表明 ,所用突变菌株均能形成附着孢并能穿透寄主细胞壁。突变菌株能形成链状着生的分生孢子 ,多数有两个分隔 ,呈弯膝形或长镰刀状 ,透明或灰橄榄色。突变菌株在病斑上和燕麦片培养基上都能产生链状着生的分生孢子 ,也能产生正常的分生孢子。通过单孢分离后 ,在燕麦片培养基上形成的两种孢子的比例基本稳定。
