Background: Two trials were perform.d to evaluate a partially defatted Herm.tia illucens(HI) larvae m.al as potential feed ingredient in rainbow trout(Oncorhynchus m.kiss Walbaum. diets. In the first trial, 360 trout(178.9 ± 9.8 g of m.an initial body weight) were random.y divided into three experim.ntal groups(4 tanks/treatm.nt, 30 fish/tank). The fish were fed for 78 days with isonitrogenous, isolipidic and isoenergetic diets containing increasing levels of HI, on as fed basis: 0%(HI0, control diet), 25%(HI25) and 50%(HI50) of fish m.al substitution, corresponding to dietary inclusion levels of 0, 20% and 40%. In the second trial, 36 trout(4 tanks/treatm.nt, 3 fish/tank) were used to evaluate the in vivo apparent digestibility coefficients(ADC) of the sam. diets used in the first ***: Survival, growth perform.nce, condition factor, som.tic indexes, and dorsal fillet physical quality param.ters were not affected by diet. The highest dietary inclusion of HI larvae m.al increased dry m.tter and ether extract contents of trout dorsal fillet. The use of HI larvae m.al induced a decrease of valuable polyunsaturated fatty acids(PUFA) even if differences were only reported at the highest level of HI inclusion. The insect m.al worsened the lipids health indexes of the sam. m.scle. Dietary inclusion of insect m.al did not alter the villus height of the fish. No differences were found am.ng treatm.nts in relation to ADC of ether extract and gross energy, while ADC of dry m.tter and crude protein were higher in HI25 if com.ared to ***: The obtained results showed that a partially defatted HI larvae m.al can be used as feed ingredient in trout diets up to 40% of inclusion level without im.acting survival, growth perform.nce, condition factor, som.tic indexes, dorsal fillet physical quality param.ters, and intestinal m.rphology of the fish. Expression of Toll-Like Receptor-2 and -4 in Am.iochorion m.m.ranes of Preterm.Delivery in the Presence of Histologic Chorioam.ionitis

Expression of Toll-like receptors at the m.ternal-fetal interface during norm.l and com.licated pregnancies has aroused interest in the last few years. However, despite the im.ortance of TLR-2 and TLR-4, which recognizes m.st m.croorganism. presenting in the am.iotic cavity infections associated with prem.turity, com.arison of the expressions of these receptors is rare in the literature. Thus, the purpose of this study was to com.are the gene expression between TLR-2 and TLR-4 in am.iochorion m.m.ranes of pregnant wom.n with preterm.delivery in the presence of histologic chorioam.ionitis (HCA). Am.iochorion m.m.ranes were collected from.40 pregnant wom.n with preterm.delivery;20 presented HCA and 20 did not. Fragm.nts of the m.m.ranes were subm.tted to total RNA extraction, followed by cDNA production by reverse transcription. Real tim. quantitative PCR was used to quantify the gene expression of the TLRs. m.NA concentrations between TLR-2 and TLR-4 were com.ared using the nonparam.tric m.nn-Whitney test. TLR-2 expression was higher than TLR-4 expression in the presence of HCA. No difference was observed between TLR-2 and TLR-4 expression in m.m.ranes in the absence of inflam.atory infiltrate. In conclusion, am.iochorion m.m.ranes express TLR-2 and TLR-4 and higher TLR-2 expression in the presence of histologic chorioam.ionitis suggests that m.croorganism. recognizable by TLR-2 play an im.ortant role in the physiopathology of preterm.labor.

Objective: The goal of this study was to determ.ne the prevalence of C. trachom.tis in wom.n diagnosed with infertility attending the Outpatient Clinic of Infertility from.Botucatu m.dical School, UNESP, Brazil. Patients and m.thods: This m.lecular study enrolled a total of 112 wom.n. Am.ng these patients, 62 presented prim.ry infertility while 50 presented secondary infertility. The criteria for eligibility included wom.n who were: reproductive-aged;no prior report of seroconversion for HIV;no antibiotic or vaginal cream.used in the preceding 30 days;and abstinence from.sexual intercourse for 72 hours before the visit. The wom.n were subm.tted to a gynecological exam.nation and cervical sam.les were collected with an endocervical cytobrush for m.lecular analysis of C. trachom.tis. Results: The prevalence of chlam.dial infection was 8% with sim.lar prevalence between prim.ry (8.1%) and secondary (8.0%) infertility. Conclusion: Considering the asym.tom.tic nature of chlam.dial infection and its association with tubal factor infertility, there is a pressing need to incorporate the screening of C. trachom.tis infection as part of the routine investigation for infertility. The early diagnostic by screening can m.nim.ze com.lications and reduce Public Health costs with Assisted Reproductive Technology.

Exosom.-like sm.ll-extracellular vesicles (sEVs) are extracellular vesicles that act in intercellular com.unication and are involved in several biologic and pathologic processes. While sEVs increase the stability of their cargo m.lecules, there is still a need for standardization of sam.ling and isolation of these m.crovesicles. We aim.d to determ.ne the best sam.ling m.thod for isolation of sEVs from.peripheral blood from.reproductive-aged wom.n. m.terial and m.thods: We included sam.les of plasm. from.our biobank collected in 2014 by venipuncture in heparin tubes and stored at -80°C. We also included blood sam.les collected in heparin tubes and Ethylenediam.ne tetraacetic acid (EDTA) tubes and stored at -80°C for one to two weeks prior processing. All blood sam.les were collected from.the sam. nine reproductive-aged fem.le volunteers. sEVs were isolated from.plasm. by ultracentrifugation and filtration and indirectly quantified using Pierce BCA Protein Assay kit. Transm.ssion electron m.croscopy (TEm. and Nano Tracking Analysis (NTA) were perform.d to confirm.the isolation of sEVs. Results and Discussion: TEm.and NTA confirm.d the isolation of sEVs. Protein concentration of short-tim. stored heparin sam.les was not statistically different from.long-tim. stored heparin sam.les (1847.2 ± 651.4 vs. 2363.2 ± 1025.1, p = 0.14). There was no difference between heparin and EDTA plasm. sam.les recently collected (2363.2 ± 1025.1 vs. 2044.8 ± 653.2, p = 0.44). In conclusion, blood sam.les m.y be collected using heparin or EDTA for isolation of sEVs. Long-tim. stored plasm. sam.les m.intain sEVs integrity and m.y be used, especially in com.arative studies.
