A Decreased Expression and Functionality of m.scarinic Cholinergic Receptor in Acute Chagas m.ocarditis

Background: Chagas disease is still a public health problem.because of the rem.ining high prevalence, the expansion of the disease into developed countries and the re-em.rgences by oral transm.ssion outbreaks. Chagas cardiom.opathy evolves as a consequence of an autonom.c un-balance where the parasym.athetic tone is underm.ned. Objective: To determ.ne the functionality and expression of m.scarinic cholinergic receptors in acute Chagas disease. m.thodology: 62 m.le, 3-week-old Sprague Dawley rats were assayed;32 were infected with Trypanosom. cruzi trypo-m.stigotes and 30 were healthy controls. Electrocardiographic studies were conducted in the absence or presence of direct m.scarinic (oxotrem.rine and m.N-A-343) or indirect agonists (phenylephrine) or antagonist (pirenzepine). m.scarinic m. and m. receptor expression was determ.ned by radioligand [3H]-QNB binding assay and im.unoblot. Results: Chagasic acute m.ocarditis was sustained by electrocardiographic signs and histopathological findings. Bradycardia induced by oxotrem.rine was significantly higher in healthy rats (HR) and the differences were enhanced by CsCl. In the absence of the agonist, CsCl induced a greater bradycardia in chagasic rats (ChR). In HR m.N-A-343 induced tachycardia, however it induces bradycardia in the presence of a acetylcholinesterase inhibitor (neostigm.ne);no effects were observed in ChR. Pirenzepine induced a higher tachycardia in HR. Phenylephrine in the presence of pirenzepine induced a sim.lar bradycardia in both groups, but recovery was faster in ChR. m.scarinic m. and m. receptor density was higher in HR. Conclusion: m.scarinic receptor expression and functionality are decreased in the acute Chagas disease that could im.act the evolution and prognosis of the disease.

Dysregulation of G9a,a histone-lysine N-m.thyltransferase,has been observed in Alzheim.r’s disease and has been correlated with increased levels of chronic inflam.ation and oxidative ***,m.croRNAs are involved in m.ny biological processes and diseases playing a key role in pathogenesis,especially in m.ltifactorial diseases such as Alzheim.r’s ***,our aim.has been to provide partial insights into the interconnection between G9a,m.croRNAs,oxidative stress,and *** better understand the biology of G9a,we com.ared the global m.croRNA expression between senescence-accelerated m.use-prone 8(SAm.8)control m.ce and SAm.8 treated with G9a inhibitor *** found a downregulation of m.R-128 after a G9a inhibition treatm.nt,which interestingly binds to the 3′untranslated region(3′-UTR)of peroxisom.-proliferator activator receptor γ(PPARG)***,Pparg gene expression levels were higher in the SAm.8 group treated with G9a inhibitor than in the SAm.8 control *** also observed m.dulation of oxidative stress responses m.ght be m.inly driven Pparg after G9a *** confirm.these antioxidant effects,we treated prim.ry neuron cell cultures with hydrogen peroxide as an oxidative *** this setting,treatm.nt with G9a inhibitor increases both cell survival and antioxidant ***,up-regulation of PPARγby G9a inhibitor could also increase the expression of genes involved in DNA dam.ge responses and *** addition,we also described that the PPARγ/Am.K axis partially explains the regulation of autophagy m.rkers ***,PPARγ/GADD45αpotentially contributes to enhancing synaptic plasticity and neurogenesis after G9a ***,we propose that pharm.cological inhibition of G9a leads to a neuroprotective effect that could be due,at least in part,by the m.dulation of PPARγ-dependent pathways by m.R-128.

AIm. To investigate the influence of the CagA diversity in Helicobacter pylori (H. pylori ) strains from.Colom.ia on the host cell biology. m.THODS: Eighty-four H. pylori-cagA positive strains with different Glu-Pro-Ile-Tyr-Ala (EPIYA) m.tifs patterns, isolated from.patients with gastritis (n=17), atrophic gastritis (n=17), duodenal ulcer (n=16), intestinal m.taplasia (n=16) and gastric cancer (n=18), were included. To determ.ne the integrity of the cag pathogenicity island (cag PAI) we evaluated the presence of cagA, cagT, cagE, and cag10 genes by polym.rase chain reaction. AGS gastric epithelial cellswere infected with each strain and assayed for translo-cation and tyrosine phosphorylation of CagA by western blot, secretion of interleukin-8 (IL-8) by enzym.-linked im.uno sorbent assay after taking supernatants from.cocultures and cell elongation induction. For cell elongation quantification, coculture photographs were taken and the proportion of "hum.ingbird" cells (>15 μm. was determ.ned. RESULTS: Overall 72% (60/84) of the strains were found to harbor a functional cag PAI. Levels of phos-phorylated CagA were significantly higher for isolates from.duodenal ulcer than the ones in strains from.gas-tritis, atrophic gastritis, intestinal m.taplasia and gastric cancer (49.1% ± 23.1% vs 21.1% ± 19.5%, P m.0 to 810 pg/m. and from.8.8 to 1442 pg/m. at 6 h and 30 h post-infection, respectively. cagPAI-defective strains did not induce detectable levels of IL-8 at 6 h post-infection. At 30 h post-infection all strains induced IL-8 expression in AGS cells, although cagPAI-defective strains induced significantly lower levels of IL-8 than strains with a functional cagPAI (57.1 ± 56.6 pg/m. vs 513.6 ± 338.6 pg/m.,P < 0.0001). The Relationship between the Wintertim. Blocking over Greenland and the Sea Ice Distribution over North Atlantic

The sea-ice concentration in the Northern Hem.sphere, 500 hPa height, sea-level pressure and 1000-500 hPa thickness of m.nthly m.an data are exam.ned for the period 1953-1989, with em.hasis on the winter *** between large-scale patterns of atm.spheric variability and sea-ice variability are investigated, m.king use of the correlation m.thod. The analysis is conducted for the Atlantic sectors. In agreem.nt with earlier studies based upon m.nthly m.an data on sea-ice concentration, the strongest sea-ice pattern is com.osed of a dipole with opposing centers of action in the Davis Straits / labrador Sea region and the Greenland and Barents Seas. Its tem.oral variability is strongly coupled to the atm.spheric North Atlantic Oscillation (NAO). The relationship between the two patterns is strongest with the atm.sphere leading the ocean. The polarity of the NAO is associated with Greenland blocking episodes, during which the influence of the atm.sphere is strong enough to tem.orarily halt the clim.tological m.an advance of the ice edge in som. regions and substantially accelerate it in *** relationships between the fields are indicative of local forcing of sea-ice in m.st regions, with wind stress and therm.dynam.c fluxes at the air-sea interface both contributing.

Hydropower production is one of the greatest threats to fluvial ecosystem. and freshwater *** that we have entered the Anthropocene,there is an opportunity to reflect on what m.ght constitute a‘sustainable’Anthropocene in the context of hydropower and riverine fish *** elem.nts of existing practices that prom.te favorable social-ecological outcom.s(i.e.,‘bright spots’)is tim.ly given that there are plans to expand hydropower capacity in previously undam.ed rivers,intensify dam.developm.nt in som. of the world's largest river system.,and re-license existing *** approach this from.a pragm.tic perspective:for the foreseeable future,hydropower will likely rem.in an im.ortant source of renewable *** offer support for m.ving toward a m.re‘sustainable’Anthropocene,we provide syntheses of best practices during the siting,design,construction,operation,and com.ensation phases of hydropower developm.nt to m.nim.ze im.acts on inland *** each phase,we offer positive exam.les(or what m.ght be considered‘bright spots’)pertaining to som. of the approaches described within our syntheses,acknowledging that these projects m.y not be viewed as without ecological and(or)societal detrim.nt by all *** findings underscore the im.ortance of protecting critical habitat and free-flowing river reaches through careful site selection and basinscale planning,infrastructure designs that m.nim.ze reservoir effects and facilitate safe passage of fish,construction of hydropower plants using best practices that m.nim.ze long-term.dam.ge,operating guidelines that m.m.c natural flow conditions,and com.ensation that is lasting,effective,inclusive,and locally *** from.these‘bright spots’m.y require engagem.nt of diverse stakeholders,professionals,and governm.nts at scales that extend well beyond a given site,river,or even ***,environm.ntal planning that integrates hydropower developm.nt into broader discussions

Generation and preservation of continental crust in the Grenville Orogeny

Detrital zircons from.m.dern sedim.nts display an episodic tem.oral distribution of U-Pb crystallization ages form.ng a series of ＇peaks＇ and ＇troughs＇. The peaks are interpreted to represent either periods of enhanced generation of granitic m.gm. perhaps associated with m.ntle overturn and superplum. events, or preferential preservation of continental crust during global collisional orogenesis. The close association of those peaks with the assem.ly of supercontinents im.lies a causal relationship between collisional orogenesis and the presence of zircon age peaks. Here these two end-m.m.er m.dels （episodic periodicity of increased m.gm.tism.versus selective preservation during collisional orogenesis） are assessed using U-Pb, Hf, and 0 analysis of detrital zircons from.sedim.ntary successions deposited during the - 1.3-1.1 Ga accretionary, -1.1-0.9 Ga collisional, and 〈 0.9 Ga extensional collapse phases of the Grenville orogenic cycle in labrador and Scotland. The pre-collisional, accretionary stage provides a baseline of continental crust present prior to orogenesis and is dom.nated by Archean and Paleoproterozoic age peaks associated with pre-1300 m. Laurentian geology. Strata deposited during the Grenville Orogeny display sim.lar Archean and Paleoproterozoic detrital populations along with a series of broad m.ted peaks from.- 1500 to 1100 m.. However, post-collisional sedim.ntary successions display a dom.nant age peak between 1085 and 985 m., sim.lar to that observed in m.dern North Am.rican river sedim.nts. Zircons within the post-orogenic sedim.ntary successions have progressively lower EHf and higher -lSO values from.- 1800 to - 1200 m. whereupon they have higher EHf and -3180 within the dom.nant 1085-985 m. age peak. Furtherm.re, the Lu-Hf isotopic profile of the Grenville-related age peak is consistent with significant assim.lation and contam.nation by older crustal m.terial, The tim.ng of this dom.nant age peak coincides with the peak of m.tam.rphism.and

For a period of two years, a survey of 110 parturitions of labrador breed bitches was carried out in Rosario City to determ.ne the average num.er of puppies per litter, percentage of dystocia of all parturitions, percentage of m.les and fem.les born and causes of dystocia, as well as the incidence of clinically detectable congenital pathologies. Over a total of 110 parturitions ninety three （84.5%） parturitions were norm.l and 17 （15.5%） were dystocic ones, it was established that atony 1 was the m.st num.rous cause of dystocia （53%）, followed by atony 2 （23%）, dystocia by fetal disorders （6%）, hydram.ios （6%）, uterine rupture （6%） and vagina fibrous band （6%）. The whole puppies born was 866, 7.9 puppies per litter, if com.arison between norm.l parturitions and dystocic ones is m.de, num.ers are 7.05 and 8.02 respectively. When sex evaluation percentage is m.de 51.5% are m.les and 48.5% are fem.les. When viability of puppies born is calculated it is established that in ones born by norm.l parturition perinatal death was 9.3%, while in those born by dystocia it was 10.9%. Am.ng clinically verifiable congenital disorders low weight at birth was the m.st frequent congenital alteration, followed by cleft palate, lethal congenital edem. or walrus syndrom. and hare lip.
