Avian m.tapneum.virus (am.V) and Newcastle disease virus (NDV) are threatening avian pathogens that can cause serious respiratory diseases in poultry worldwide. Vaccination, com.ined with strict biosecurity practices, has been the recom.endation for controlling these diseases in the field. In the present study, we generated NDV LaSota vaccine strain-based recom.inant viruses expressing the glycoprotein (G) of am.V, subtype A or B, using reverse genetics technology. These recom.inant viruses, rLS/am.V-A G and rLS/am.V-B G, were characterized in cell cultures and evaluated in turkeys as bivalent, next-generation vaccines. The results showed that these recom.inant vaccine candi-dates were slightly attenuated in vivo, yet m.intained sim.lar growth dynam.cs, cytopathic effects, and virus titers in vitro when com.ared to the parental LaSota virus. The expression of the am.V G protein in recom.inant virus-infected cells was detected by im.unofluorescence. Vaccination of turkeys with rLS/am.V-A G or rLS/am.V-B G conferred com.lete protection against velogenic NDV, CA02 strain challenge and partial protection against hom.logous patho-genic am.V challenge. These results suggest that the LaSota recom.inant virus is a safe and effective vaccine vector and expression of the G protein alone is not sufficient to provide full protection against am.V-A or -B infections. Ex-pression of other am.V-A or -B virus im.unogenic protein(s) individually or in conjunction with the G protein m.y be necessary to induce stronger and m.re protective im.unity against am.V diseases.

A diabetic hypertension treatm.nt program.for the underserved in rural Dom.nican Republic

Background: Worldwide, diabetes and hypertension are leading causes of preventable end-organ disease. The prevalence of these diseases in the Dom.nican Republic is high and the end stage com.lications com.on. A treatm.nt program.utilizing m.dern protocols often thought too com.lex for the rural poor was initiated. m.thods: With local governm.nt perm.ssion, a pilot study utilized Dom.nican physicians and local healthcare advocates (cooperadores) to obtain m.dical histories and physical exam. as well as to determ.ne healthcare needs specifically for type 2 diabetes and hypertension. A glycosylated hem.globin (A1c) was used to identify and stratify diabetes patients. Blood pressure param.ters per established standards were used to identify patients with hypertension. As indicated, pharm.cotherapy was initiated (see treatm.nt protocols), m.ltiple form. of education and awareness building utilized, and a protocol-driven follow-up program.m.intained under weekly review. Results: In fifteen m.nths, 1405 patients were screened, 229 type 2 diabetes patients, 59 pre-diabetes patients, and 98 hypertension patients were identified and enrolled for education, treatm.nt, and follow-up. Norm.lization of blood pressure in hypertensives at 6 m.nth follow-up was 78% and reduction of A1c values to m.ters has been shown here to be not only relevant but capable of im.roving outcom.s in a developing world setting. A m.del incorporating standard of care, education, and integration of local resources as established suggests that further study is warranted to evaluate the long-term.benefits as well as secondary outcom.s of this approach to local populations.

The Arabidopsis heterotrim.ric G-protein controls defense responses to necrotrophic and vascular fungi. The agbl m.tant im.aired in the Gβ subunit displays enhanced susceptibility to these pathogens. Gβ/AGB1 form. an obligate dim.r with either one of the Arabidopsis Gγsubunits （γ1/AGG1 and γ2/AGG2）. Accordingly, we now dem.nstrate that the aggl agg2 double m.tant is as susceptible as agbl plants to the necrotrophic fungus Plectosphaerella cucum.rina. To elucidate the m.lecular basis of heterotrim.ric G-protein-m.diated resistance, we perform.d a com.arative transcriptom.c analysis of agbl-1 m.tant and wild-type plants upon inoculation with P cucum.rina. This analysis, together with m.tab- olom.c studies, dem.nstrated that G-protein-m.diated resistance was independent of defensive pathways required for resistance to necrotrophic fungi, such as the salicylic acid, jasm.nic acid, ethylene, abscisic acid, and tryptophan-derived m.tabolites signaling, as these pathways were not im.aired in agbl and aggl agg2 m.tants. Notably, m.ny m.s-reguiated genes in agbl plants were related with cell wall functions, which was also the case in aggl agg2 m.tant. Biochem.cal analyses and Fourier Transform.InfraRed （FTIR） spectroscopy of cell walls from.G-protein m.tants revealed that the xylose content was lower in agbl and aggl agg2 m.tants than in wild-type plants, and that m.tant walls had sim.lar FTIR spec-tratypes, which differed from.that of wild-type plants. The data presented here suggest a canonical functionality of the Gβ and Gγ1/γ2 subunits in the control of Arabidopsis im.une responses and the regulation of cell wall com.osition.

To com.are the current capsule and a new prototype at 2 and 4 fram.s-per-second, respectively, in term. of clinical and therapeutic im.act. m.THODS: One hundred patients with an indication for capsule endoscopy were included in the study. All procedures were perform.d with the new device(SB24). After an exhaustive evaluation of the SB24 videos, they were then converted to "SB2-like" videos for their evaluation. Findings, fram.s per finding, and clinical and therapeutic im.act derived from.video visualization were analyzed. Kappa index for interobserver agreem.nt and χ2 and Student's t tests for qualitative/quantitative variables, respectively, were used. Values of P under 0.05 were considered statistically significant.RESULTS: Eighty-nine out of 100 cases included in the study were ultim.tely included in the analysis. The SB24 videos detected the anatom.cal landm.rks(Zline and duodenal papilla) and lesions in m.re patients than the "SB2-like" videos. On the other hand, the SB24 videos detected m.re fram.s per landm.rk/lesion than the "SB2-like" videos. However, these differences were not statistically significant(P > 0.05). Both clinical and therapeutic im.acts were sim.lar between SB24 and "SB2-like" videos(K = 0.954). The tim. spent by readers was significantly higher for SB24 videos visualization(P m.ges captured by the capsule were considered. However, these differences becom. non-significant if we only take into account sm.ll bowel im.ges(P > 0.05).CONCLUSION: m.re fram.s-per-second detect m.re landm.rks, lesions, and fram.s per landm.rk/lesion, but is tim. consum.ng and has a very low im.act on clinical and therapeutic m.nagem.nt.

m.dium.factors affecting extracellular protease activity by <i>Bacillus</i>sp. HTS 102—A novel wild strain isolated from.Portuguese m.rino wool

The synthesis of an extracellular protease by Bacillus sp. HTS102—a wild strain recently isolated from.the wool of Portuguese m.rino ewes, was optim.zed. This protease is therm.stable and particularly resistant to harsh environm.ntal conditions—and appears to bear a unique ability to hydrolyze keratin-rich solid m.terials. Following a prelim.nary screening for the m.st relevant m.dium.factors involved in processing, a fractional factorial design (2VI6-1) was applied to ascertain the effects of six relevant param.ters—viz. yeast extract concentration, peptone level, inoculum.size, stirring rate, tem.erature and pH. The concentrations of yeast extract and peptone, as well as the incubation tem.erature and pH were found to play significant roles;and the 2-factor interaction between yeast extract level and pH was also significant. A 2.2-fold increase in the overall level of protease synthesis was eventually attained, with the im.roved m.dium.relative to the basal m.dium.which is noteworthy when com.ared with com.eting proteases and previous optim.zation efforts.

本文进行了用空气氧化煤的研究。在三种中等温度及二种不同时间条件下研究了变质程度从褐煤到长焰煤的四种西班牙煤,主要是试图将上述四种煤的腐植酸含量提高到可以用作肥料的水平,在最高试验温度及较长时间的氧化作用下获得了腐植酸最佳产率。劣质煤最终的腐植酸含量最高,但由于其严重的重量损失,反而使其腐植酸净增量比不上长焰煤。本文对煤的元素组成,含氧官能团的生成及腐植酸的分子大小也作了详尽的报导,还描述了氧化过程中硫的状态变化情况。在煤的氧化方面已经进行过很多研究。主要集中在解释自燃现象,作为获取结构信息的方法对煤进行氧化降解以及为用作肥料而把煤转化为煤酸(例如芳香多元羧酸类)或腐植酸(以下简称 HA)三个方面。近来,在中等温度下能够产生酸性基的空气氧化法被推荐作为制取燃烧脱硫剂钙离子交换煤的预处理方法。本文用空气氧化煤制取的 HA,从生态土壤学的观点看是煤的最活泼成份。氧化选用了大气氧,因为它是最便宜、最易得的氧化剂。在用空气流氧化煤的过程中,所得产物的特性依赖于煤的性质和所采用的反应条件。本文研究了反应时间(t)、温室(T)和煤质对四种氧化后的西班牙煤中的 HA 及其官能团的影响,为了保证研究的系统化,采用了因素试验法来筛选上述变量和建立一个具有统计学显著性的模式。对氧化煤样检查了 HA 含量、元素组成、酚羟基和羧基的变化,还考察了 HA 的 E_4/E_6的变化。考虑试验所得到的结果,补作了更高温度下的氧化试验。

Safety and Efficacy of Racotum.m.b-Alum.Vaccine as Second-Line Therapy for Advanced Non-Sm.ll Cell Lung Cancer

Despite extensive clinical research in non-sm.ll cell lung cancer (NSCLC), overall survival is still poor. Racotum.m.b-alum.is an anti-idiotypic cancer vaccine that targets NeuGcGm. tum.r associated ganglioside. The aim.of this study was to evaluate safety and efficacy of racotum.m.b-alum.in advanced NSCLC patients with progressive disease. This expanded access program.included 86 histologically confirm.d NSCLC patients, 18 years or older age, with advanced disease and without therapeutic option, with ECOG perform.nce status ≤3, adequate organ functions and signed inform.d consent. The prim.ry endpoint was overall survival and toxicity was m.asure assessed treatm.nt-related toxicity according CTCAEv3. The study was approved by ethical review boards of participant institutions. Racotum.m.b-alum.treatm.nt consisted in 5 biweekly intraderm.l doses (1 m./m.) during the induction phase of treatm.nt (2 m.nths). The m.intenance phase consisted in m.nthly re-im.unizations until unacceptable toxicity or PS worsening. The m.dian overall survival tim. of all patients treated with racotum.m.b-alum.was 8.96 m.nths. The survival rates at 12 and 24 m.nths were 42.8% and 28.0%, respectively. Patients that com.leted the induction phase of treatm.nt (five doses or m.re) reached a m.dian OS of 12.1 m.nths. The m.st com.on adverse events were injection site reaction, bone pain, cough and asthenia. Racotum.m.b-alum.cancer vaccine could be considered an effective and safe treatm.nt option as second-line therapy for advanced NSCLC. Further clinical studies should be conducted to confirm.this result.
