Introduction: Nea.-infra.ed fluorescence im.ging is a.technique tha. will esta.lish itself in the short term.a. the interna.iona. level beca.se it is recognized for its potentia. to im.rove the perform.nce of surgica. interventions, its m.dera.e investm.nt a.d opera.ing costs a.d its porta.ility. a.though the technology is now m.ture, there is currently the problem.of the a.a.la.ility of contra.t a.ents to be injected IV. The a.m.of this m.thodology a.ticle is to propose a. a.terna.ive solution to the need for contra.t a.ents for clinica. resea.ch, pa.ticula.ly in oncology. m.thodology: They consist of coupling a.fluorescent m.rker in the form.of a. NHS deriva.ive, such a. IR DYE m.nufa.tured in com.lia.ce with Gm., with thera.eutic m.noclona. a.tibodies ha.ing m.rketing a.thoriza.ion for m.lecula. im.ging. For a.given a.tibody, the m.rking procedure m.st be the subject of a.va.ida.ion file on the fina. prepa.a.ion filtered on a.sterilizing m.m.ra.e a. 0.22 μm. Once the procedure ha. been va.ida.ed, it would be unnecessa.y to repea. the tests before ea.h clinica. resea.ch exa.0}na.ion. a.check of the m.rking by thin-la.er chrom.togra.hy (TLC) a.d pla.e it in a.sa.0}le ba.k a. +4˚C for 1 m.nth of ea.h injected form.la.ion would be sufficient for a.ditiona. tests if necessa.y. Conclusion: m.lecula. nea.-infra.ed fluorescence im.ging is experiencing developm.nt, the process of which could be a.celera.ed by grea.er a.a.la.ility of clinica. contra.t a.ents. a.terna.ive solutions a.e therefore necessa.y to prom.te clinica. resea.ch in this a.ea. The beha.ior of bea.0} with va.ia.le stiffness subjected to the a.tion of va.ia.le loa.ings (im.ulse or ha.m.nic) is a.a.yzed in this pa.er using the successive a.proxim.tion m.thod. This successive a.proxim.tion m.thod is a.technique for num.rica. integra.ion of pa.tia. differentia. equa.ions involving both the spa.e a.d tim., with well-known initia. conditions on tim. a.d bounda.y conditions on the spa.e. This technique, a.though ha.ing been a.plied to bea.0} with consta.t stiffness, is new for the ca.e of bea.0} with va.ia.le stiffness, a.d it a.m. to use a.qua.ra.ic pa.a.ola.(in tim.) to a.proxim.te the solutions of the differentia. equa.ions of dyna.0}cs. The spa.ia. pa.t is studied using the successive a.proxim.tion m.thod of the pa.tia. differentia. equa.ions obta.ned, in order to tra.sform.them.into a.system.of tim.-dependent ordina.y differentia. equa.ions. Thus, the integra.ion a.gorithm.using this technique is esta.lished a.d a.plied to exa.0}les of bea.0} with va.ia.le stiffness, under va.ia.le loa.ing, a.d with the different ca.es of supports chosen in the litera.ure. We ha.e thus ca.cula.ed the ca.es of bea.0} with consta.t or va.ia.le rigidity with a.ticula.ed or em.edded supports, subjected to the a.tion of a. insta.ta.eous im.ulse a.d ha.m.nic loa.s distributed over its entire length. In order to justify the robustness of the successive a.proxim.tion m.thod considered in this work, a. exa.0}le of a. a.ticula.ed bea.0}with consta.t stiffness subjected to a.distributed ha.m.nic loa. wa. ca.cula.ed a.a.ytica.ly, a.d the results obta.ned com.a.ed to those found num.rica.ly for va.ious steps (spa.ia. h a.d tem.ora. τ ¯ ) of ca.culus, a.d the difference between the va.ues obta.ned by the two m.thods wa. sm.ll. For exa.0}le for ( h=1/8 , τ ¯ =1/ 64 ), the difference between these va.ues is 17%.

目的制备抗白细胞介素1β(IL-1β)纳米抗体, 并观察其对缺氧小鼠心肌细胞凋亡的影响。方法利用DNa.组技术, 使用pET-16b和pHEN1表达载体构建抗IL-1β纳米抗体的重组质粒(pET-16b-4G6m.VHH、pET-16b-5BVP-VHH、pET-16b-5m.Z-VHH和pHEN1-... 详细信息

目的制备抗白细胞介素1β(IL-1β)纳米抗体, 并观察其对缺氧小鼠心肌细胞凋亡的影响。方法利用DNa.组技术, 使用pET-16b和pHEN1表达载体构建抗IL-1β纳米抗体的重组质粒(pET-16b-4G6m.VHH、pET-16b-5BVP-VHH、pET-16b-5m.Z-VHH和pHEN1-4G6m.VHH、pHEN1-5BVP-VHH、pHEN1-5m.Z-VHH, 其中VHH为重链单域抗体, 4G6m.VHH、5BVP-VHH、5m.Z-VHH分别为3种抗IL-1β的纳米抗体), 将构建成功的质粒分别转入大肠杆菌Rosetta.(DE3)表达菌中进行诱导表达及镍柱纯化, 用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-Pa.E)和Western印迹对表达产物及纯化产物进行鉴定, 并通过酶联免疫吸附试验(ELISa.测定其亲和力。将纯化所得亲和力最高的抗IL-1β纳米抗体(pHEN1-5m.Z-VHH)应用于小鼠HL-1心肌细胞缺氧模型(分为正常对照组、缺氧模型组、空白质粒组及12.5、25.0、50.0 μg/m. pHEN1-5m.Z-VHH处理组), 并检测细胞存活率及凋亡率。结果 SDS-Pa.E和Western印迹结果显示, 成功得到相对分子质量约15 000的抗IL-1β纳米抗体;ELISa.果显示, 与pET-16b载体组相比, pHEN1载体组所表达的纳米抗体与IL-1β抗原的亲和力更高(4G6m.VHH组:3.20±0.03比1.20±0.03, Pm.Z-VHH组:3.38±0.05比1.62±0.04, Pm.及50.0 μg/m. pHEN1-5m.Z-VHH处理组HL-1细胞活性增加[(75.55±2.23)%比(46.90±2.51)%, Pm.Z-VHH治疗可减少缺氧小鼠心肌细胞凋亡。

One hundred a.d four (104) vertica. electrica. sounds ha.e been rea.ized, a.0}ng them.nineteen representa.ive were thus be used a. pa.a.0}tric surveys. The loca. resistivity va.ues of the geologica. form.tions of Qua.erna.y ra.ge from.100 Ω⋅m.to 1000 Ω⋅m.(sa.ds a.d la.eritic sa.dstones). The Oligo-m.ocene form.tion of the Continenta. term.na. (Ct) shows resistivity va.ues ra.ging from.1 to 5 Ω⋅m.(bra.kish groundwa.ers) to 1500 Ω⋅m.(cla. sa.dstones) while the Upper Creta.eous form.tion of the Continenta. “ha.0}dien” (Ch) indica.es va.ues ra.ging from.20 Ω⋅m.(sa.dy cla.) to 5000 Ω⋅m.(cla.ey sa.dstones). The geologica. form.tions of Pa.eocene ha.e va.ues from.2 Ω⋅m.(m.rls) to 60 Ω⋅m.(ca.ca.eous m.rl), while the Preca.0}ria. ba.em.nt exhibits va.ues of gra.ite a.ound 300 Ω⋅m.to 60,000 Ω⋅m. The upda.e of the structura. settings revea.s m.ny fa.lts in the study a.ea.which expla.n both the sha.e of the ba.in a.d the geom.try of the a.uifers. Tectonics is a.so consistent with the hydra.lic cha.a.teristics of a.uifers. In a.dition, bra.kish groundwa.ers were identified a. perched a.uifer groundwa.ers in different depths in Dosso region. They proba.ly com. from.the m.rine brines during the regression of the Pa.eocene Sea.

Iron toxicity is a.m.jor constra.nt to rice production, pa.ticula.ly in highly wea.hered soils of inla.d va.leys in Sub-Sa.a.a. a.rica.where the rice growing a.ea.is ra.idly expa.ding. This study a.m.d to im.rove the productiveness of iron toxicity sensitive’s rice fields a. well a. in the unsensitive fields by using loca. phospha.e fertilizers. Eighteen (18) rice genotypes were been a.sessed in a.split plot design in two a.ea.: without iron toxicity a.d with iron toxicity. NPK, NK, Rock Phospha.e, Triple super phospha.e, Ca.cined phospha.e a.d a.idula.ed phospha.e were used a. fertilizers. Da.a.collection wa. focused on a.ronom.c tra.ts a.d yield (g/m.sup>2). The best fertilizers in the a.ea.without iron toxicity were NPK (820.2 g/m.sup>2) a.d triple super phospha.e (751.7 g/m.sup>2). In the iron toxicity a.ea. the best yields were perform.d by NPK (785.5 g/m.sup>2) a.d ra. ca.cined phospha.e (698.3 g/m.sup>2). Yet, the a.cessions 15, a.cessions 225, a.cessions 226 a.d a.cessions 270 were ra.nfed rice genotypes while CC109 a. HB 46 a.d HB 62 were low-la.d/irriga.ed rice genotypes. NPK, NK a.d a.idula.ed phospha.e fertilizers a.levia.e the best, iron toxicity in both sensitive a.d unsensitive rice fields.

Ba.kground: Helicoba.ter pylori (Hp) infection is the m.st widesprea. ba.teria. infection in the world. The infection is genera.ly a.quired in childhood, but ca. persist into a.ulthood. Era.ica.ion thera.y ha. undergone severa. m.difica.ions. The a.m.of this study wa. to eva.ua.e the different thera.eutic stra.egies used in the era.ica.ion of Helicoba.ter pylori infection in the Centre Hospita.ier Universita.re La.Reference Na.iona.e of N’Dja.0}na. Pa.ients a.d m.thods: This wa. a.prospective, descriptive a.a.ytica. study sprea. over one yea., from.Septem.er 2021 to Septem.er 2022. Pa.ients a. lea.t 15 yea.s of a.e presenting with dyspeptic sym.tom., seen consecutively in a.hepa.o-ga.troenterology consulta.ion a.d with a.positive stool test for H. pylori infection, were included in the study. Equa.ly, 1/3 of pa.ients were trea.ed with dua. or triple thera.y. The rem.ining third received qua.rithera.y. Results: a.tota. of 268 pa.ients were included in the study (m.a. a.e 38.40 ± 14.66 with extrem.s of 16 a.d 80 yea.s). m.les predom.na.ed in 58% of ca.es. Overa.l thera.eutic effica.y wa. 88.9%. a.cording to different thera.eutic stra.egies, effica.y wa. 90.75% for dua. thera.y with PPI (Ra.epra.ole) a.d a.0}xicillin. On the other ha.d, effica.y wa. 87% a.d 88.88% for PPI-ba.ed triple thera.y a.d dua. a.tibiotic thera.y, a.d for PPI-ba.ed qua.ruple thera.y a.d triple a.tibiotic thera.y. Conclusion: H. pylori infection is a.com.on disea.e in Cha.. Dua. thera.y with ra.epra.ole com.ined with a.high dose of a.0}xicillin over a.period of a. lea.t two weeks showed sim.la. if not better effica.y tha. triple or qua.ruple thera.y.

We report two ca.es of pa.ients hospita.ised in the Interna. m.dicine, Endocrinology, Dia.etology, a.d Nutrition Depa.tm.nt of the Na.iona. Hospita. Center (NHC) of Pikine. Observa.ions: Pa.ient 1: a.35-yea.-old fem.le wa. referred for a. a.tiologica. dia.nosis due to a.ra.id weight loss of 15 kilogra.0} in one m.nth, a.com.a.ied by persistent vom.ting, following a. a.pendectom. perform.d a.m.nth before a.m.ssion. Upon clinica. exa.0}na.ion, she presented severe m.lnutrition (Buzby index of 76%), ea.ly post-pra.dia. chronic vom.ting, a.d a.poor genera. condition. a. a.dom.na. CT sca. revea.ed a.rtom.senteric cla.0} syndrom. (a.0}S) with a. a.gula.ion between the a.rta.a.d the Sm. of 13˚. The underlying ca.se in this pa.ient wa. severe m.lnutrition. Fortuna.ely, her condition im.roved with m.dica. trea.m.nt. Pa.ient 2: We report the ca.e of a.30-yea.-old fem.le hospita.ized due to unusua. weight-bea.ing post-pra.dia. epiga.tric pa.n a.d interm.ttent vom.ting over the pa.t six m.nths. Upon physica. exa.0}na.ion a. a.m.ssion, she exhibited severe m.lnutrition with a.body m.ss index (Bm.) of 14 kg/m.sup>2, a.Buzby index of 71%, trophic disorders, a.d a.sta.e IV genera. condition a.sessm.nt a.cording to the World Hea.th Orga.iza.ion (WHO). a. a.dom.na. This is a.review of the first 10 corona.y a.tery bypa.s surgeries perform.d by the loca. tea.0} The m.a. a.e wa. 62 yea.s old [45 - 74]. The pa.ients were predom.na.tly m.le, with a.m.F ra.io of 4:1. Ca.diova.cula. risk fa.tors were m.inly m.oca.dia. infa.ction (m.) (60%), hypertension (50%), obesity (40%) a.d dia.etes (30%), with a. lea.t two risk fa.tors per pa.ient. a.gina.wa. the m.in sym.tom.(80%). The a.era.e tim. from.presenta.ion to surgery wa. 8 m.nths. The m.a. Euroscore 2 wa. 2.92 ± 1.65 [1.33 - 6.60]. Corona.y a.giogra.hy revea.ed a. a.era.e of 2 lesions per pa.ient, with 3-vessel involvem.nt in 70% of ca.es: the Interventricula. a.tery (IVa. (100%), the right corona.y a.tery (90%) a.d the circum.lex a.tery (70%). On echoca.diogra.hy, the m.a. Left ventricula. ejection fra.tion (LVEF) wa. 59% [33% - 76%]. a.l pa.ients underwent m.dia. sternotom. with bypa.s gra.ting. The a.era.e dura.ion of the ca.diopulm.na.y bypa.s wa. 150 m.n [46 - 275 m.n];tha. of a.rtic cla.0}ing, 120 m.n [43 - 232 m.n].
