Colorectal cancer(CRC)is the second leading cause of cancer-related death *** five-year survival rate of CRC patients depends on the stage at diagnosis,being higher than 80%when CRC is diagnosed in the early stages but lower than 10%when CRC is diagnosed in advanced *** against specific CRC autoantigens(tumor-associated antigens(TAAs))in the sera of patients have been widely demonstrated to aid in early ***,we herein aim to identify autoantigens target of autoantibodies specific to CRC that possess a significant ability to discriminate between CRC patients and healthy individuals by means of liquid *** that end,we examined the protein content of the exosomes released by five CRC cell lines and tissue samples from CRC patients by means of immunoprecipitation coupled with mass spectrometry analysis.A total of 103 proteins were identified as potential autoantigens specific to *** bioinformatics and meta-analysis,we selected 15 proteins that are more likely to be actual CRC autoantigens in order to evaluate their role in CRC prognosis by Western blot(WB)and immunohistochemistry(IHC).We found dysregulation at the protein level for 11 of these proteins in both tissue and plasma exosome samples from patients,along with an association of nine of these proteins with CRC *** validation,all but one showed a statistically significant high diagnostic ability to distinguish CRC patients and individuals with premalignant lesions from healthy individuals,either by luminescence Halotag-based beads,or by a multiplexed biosensing platform involving the use of magnetic microcarriers as solid support modified with covalently immobilized Halotag fusion proteins constructed for CRC *** together,our results highlight the usefulness of the approach defined here to identify the TAAs specific to chronic diseases;they also demonstrate that the measurement of autoantibody levels in plasma against the TAAs identified here

A Di-D-Fructose Dianhydride-Enriched Caramel modulates Pig Fecal microbiota Composition

A correlation has been previously described between bifidobacteria counts before and after the use of a dietary additive in human studies. However, to our knowledge no information on this topic has yet been reported in animals, and no information exists either on similar possible correlations of bacterial groups other than bifidobacteria. The potential prebiotic effects of di-D-fructose dianhydride (DFA)-enriched caramels have been previously reported in laboratory animals, poultry and pigs. In the present work, twelve growing male castrated pigs (41.8 ± 1.9 kg mean BW) were fed in succession on a control (no additive) or DFA-enriched caramel (20 g/kg) containing diets. Another group of 10 pigs (38.0 ± 3.7 kg mean BW) fed on a control diet without any additive was used as negative control. Bacterial log10 number of copies of the 16S rRNA gene was determined in fecal samples by using qPCR. Increased (P10 number of copies were determined in fecal samples of pigs fed on the caramel containing diet compared with non-caramel controls. In addition, for all bacterial groups studied microbiological values co-variated with initial counts and, except for enterobacteria, variations in the fecal bacterial numbers after caramel supplementation correlated (Pmbers before supplementation. In conclusion, the supplementation of pig diets with DFA-enriched caramels induced significant increases in the fecal number of copies of bacterial groups regarded as beneficial, and variations in the fecal number of copies correlated with the initial fecal number of copies.

Biodrying under Greenhouse Conditions as Pretreatment for Horticultural Waste

The biodrying process was studied as an alternative technology to reduce the mass and volume of horticultural waste. Four static piles were prepared inside a greenhouse: two containing whole waste and two consisting of shredded waste. All the piles were compared with a test pile containing whole waste and placed outside the greenhouse. In two cases, ventilation ducts were installed to improve aeration. Each greenhouse was 2.0 m wide, 3.5 m long and 1.16 m high. The air temperature and relative humidity were monitored both inside and outside the greenhouse. mass, humidity, organic matter and total nitrogen in the waste were measured. Piles inside the greenhouse showed decreases of 80% and 75% in weight and volume, respectively, during the first 16 days. The data obtained in this work suggest that biodrying could improve the handling and transport of horticultural waste while also minimizing the impact of pollutants.

Effect of Brefeldin A on the Development of <i>Sargassum cymosum</i>Zygotes (Phaeophyceae, Fucales): Structural and Ultrasctructural Analysis

To evaluate the effects of brefeldin A (BFA) on Golgi bodies and indirectly on the polarized development of S. cymosum zygotes, zygotes were cultured at concentrations of 4 μm, 8 μm, 16 μm and 32 μm of BFA, prepared in sterilized seawater. After 12 hours, the samples were fixed and processed for transmission electron microscopy (TEm), confocal laser scanning microscopy, light microscopy and cell counting. For recovery analysis, after treatment, the samples were again placed in sterile seawater for over 12 hours. In the control, after 12 hours, the embryos were adhered and with advanced stage of development, presenting up to 6 cell divisions. In the treated samples, the embryos did not adhere and only in the lower concentrations were they visualized with cellular divisions, is that in the highest concentration, the majority of the zygotes were dead. After recovery, embryos showed development only in the lowest concentrations. By confocal microscopy, the treated samples had an irregular distribution of chloroplasts and physodes. Also through TEm, it was possible to observe the loss of organization of Golgi bodies, in turn leading to the formation of vesicles and fusion of physodes. We can conclude that Golgi bodies are responsible for the production and secretion of molecules related to the adhesion and formation of cell wall membrane, also aiding in the polarization and orientation of physodes. The fused physodes occupied much of the cytoplasm, preventing other cytoplasmic processes, thus directly affecting the development of this alga.

Development of the Gusto Facial Reflex in Perinatally Undernourished Rats

In the rat, the taste system plays a critical role in motivating the animal to consume nutrients and avoid toxic substances. In neonatal rat the orofacial movements can be modified by the application of sucrose and quinine in the mouth, producing ingestion or rejection responses, respectively, but there is no information available on the gusto facial reflexes (GFR) under perinatal fasting. The aim of the present study was to determine how undernutrition can affect the development of orofacial responses to sucrose, quinine, and NaCl during early development. Pregnant dams were undernourished by being given 50% of a balanced diet from G6 to G12, 60% from G13-G18, and 100% from G19-G21. On postpartum days 0 - 9, prenatally underfed (UG) pups continued the undernourishment by remaining for 12 h with a foster dam and 12 h with a nipple-ligated mother. Stimuli were presented as a single droplet of sucrose (S), sodium chloride (NaCl), quinine hydrochloride (Q), and water (W) onto the lips at 1, 3, 5, 7, and 9 days of age, and mouth-opening frequency (mO) and lip-licking frequency (LL) were analyzed. The results show consistent effects associated with age, but not with diet, except in the cases of 0.01 m Q, which provoked in the UG subjects increased mO and the reduction of LL, and 0.1 m Q, which consistently reduced both responses. moreover, S provoked increases in both mO and LL at 0.1 m while NaCl and water elicited minimal effects on GFR. These data suggest that perinatal undernutrition affects the basic components of the gustatory system necessary to produce GFR in the first postnatal days by possibly interfering with the integration of taste input for food learning and the hedonic aspects of gustatory cues.

Retrieval of Pups by Female Rats Undernourished during the Pre- and Neonatal Period

Early mother-litter bonds alterations elicited by pre- and neonatal undernutrition are a source of long-lasting brain and functional disorders such as exploratory and emotional deficits and maternal unresponsiveness to the newborn. Here, we examined the prolonged effects of gestational and neonatal food restriction on the retrieval of pups and nest building activity by primiparous adult Wistar rats on postnatal days (PDs) 4, 8, and 12. In early undernourished dams, the latency to retrieve the first or the fifth pups in the litter was extended;and nest building activity was significantly reduced. Additionally, early underfed dams retrieved the pups in a rough manner, eliciting distress cries and grasping the pups by unusual body areas. The current findings suggest that pre- and neonatal food restriction may affect the anatomical and functional forebrain structures, modulating the cognitive and motor components underlying maternal responsiveness.

Oncomodulin and macrophages derived factors in pancreas injury and development paradigms

Prior studies in the optic nerve injury paradigm showed con?icting data regarding production and signi?cance of the Ca2+-binding protein oncomodulin (OCm). Some have shown its potentaxon-regenerative or-growth attribute, where other showed little to no effect. We show here that pancreas inflammation lead to macrophages infiltration that produce OCm and inflamed tissues that express OCm receptorsin vivo. In culture OCm has a cytostatic effect on embryonic pancreas explants. Secretory products of zymosan activated macrophages are cytotoxic and factors derived from non-activated macrophages seem to promote pancreas development. It is our view that OCm is involved in protective injury response that allows through metabolism slowing sublethally injured cells to undergo recovery.

Immunotherapy and immunoescape in colorectal cancer

Immunotherapy encompasses a variety of interventions and techniques with the common goal of eliciting tumor cell destructive immune responses. Colorectal carcinoma often presents as metastatic disease that impedes curative surgery. Novel strategies such as active immunization with dendritic cells (DCs), gene transfer of cytokines into tumor cells or administration of immunostimulatory monoclonal antibodies (such as anti-CD137 or anti-CTLA-4) have been assessed in preclinical studies and are at an early clinical development stage. Importantly, there is accumulating evidence that chemotherapy and immunotherapy can be combined in the treatment of some cases with colorectal cancer, with synergistic potentiation as a result of antigens cross-presented by dendritic cells and/or elimination of competitor or suppressive T lymphocyte populations (regulatory T-cells). However, genetic and epigenetic unstable carcinoma cells frequently evolve mechanisms of immunoevasion that are the result of either loss of antigen presentation, or an active expression of immunosuppressive substances. Some of these actively immunosuppressive mechanisms are inducible by cytokines that signify the arrival of an effector immune response. For example, induction of 2, 3 indoleamine dioxygenase (IDO) by IFNγ in colorectal carcinoma cells. Combinational and balanced strategies fostering antigen presentation, T-cell costimulation and interference with immune regulatory mechanisms will probably take the stage in translational research in the treatment of colorectal carcinoma.
