Statistical optimization of lignocellulosic waste containing culture medium for enhanced production of cellulase by Bacillus tequilensis G9

The ever increasing energy demands of modern civilization and rapidly dwindling fossil fuels point towards a renewable substitute like ***,higher costs associated with biofuel productions is the major bottleneck for its *** present study demonstrates the use of a statistical approach called response surface methodology(RSm)to investigate the optimum parameters for maximum production of cellulase by Bacillus tequilensis *** Plackett-Bur-man design(PB)of the RSm analysis indicated grass straw(GS)concentration,pH,FeSO4,inoculum,mgSO4,incubation period and NH4Cl as significant variables that influence the cellulase ***,to propose the best medium for the maximum production of cellulase by *** G9,the most influential parameters,namely concentrations of GS as substrate,FeSO4,pH,inoculum size,*** fine-tuned by central composite design(CCD)involving four factors and five *** CCD analysis demonstrated 8%substrate concentration,1.5%of inoculum along with 10 ppm FeSO4 and a pH of 5.5 in media as optimum conditions for highest enzyme *** field emission scanning electron microscopic analysis of the treated GS showed structural alterations depicting significant deconstruction caused by *** *** yield of the partially purified cellulase proteins were found to be 21%revealing molecular mass between 30 and 97 *** enhanced cellulase production by *** G9 demonstrated in our study brands its applications in many industrial processes like biorefinery,biofuels,etc.

Anthelmintic, antimicrobial, antioxidant and cytotoxic activity of Caltha palustris var. alba Kashmir, India

The methanolic extract obtained from the root portion of Caltha palustris var. alba was evaluated for its anthelmintic efficacy against gastrointestinal nematodes of sheep under both in vitro and in vivo conditions using worm motility inhibition(WmI) assay and fecal egg count reduction(FECR) assay, respectively. The extract was subjected to antimicrobial activity using agar-well diffusion method against different bacterial strains. In addition the extract was evaluated for cytotoxic and antioxidant activity against cultured THP-1(Leukemia), A-549(Lung), HCT-15(Colon), Cervix(HeLa) and PC-3(Prostrate) cell lines by SRB and DPPH radical scavenging assays. The extract used resulted in mean %WmI of 94.44%, as observed when the worms were put in lukewarm buffer for 30 min after exposure to different treatments. The mean mortality index of the sample was 0.95. The lethal concentration(LC50) was 0.11 mg·mL-1. Cell lines were exposed to concentration of 100 μg·mL-1 of extract for 48 h, which reduced the viability of these cell lines. The same plant extract also showed 55.58% DPPH radical scavenging activity.

The CCD photometric observations of open star cluster m37(NGC 2099) were carried out up to a limiting magnitude of V ～ 20 in both B and V filters to search for variable stars using a 2k×4k CCD and the 1.3 m telescope at the Vainu Bapu Observatory, Kavalur.A total of 314 stars were in the first observing run, out of which 60 were identified as *** out of the identified 60 variables are classified as W Uma binary *** model fitting, we used PHOEBE based on the W-D code to estimate the physical parameters of these newly detected W Uma binaries that theoretically best match the observed light curves.

Assessment of Genetic Variation Within Indian mustard (Brassica juncea) Germplasm Using Random Amplified Polymorphic DNA markers

Genetic diversity among 45 Indian mustard （Brassica juncea L.） genotypes comprising 37 germplasm collections, five advance breeding lines and three improved cultivars was investigated at the DNA level using the random amplified polymorphic DNA （RAPD） technique. Fifteen primers used generated a total of 92 RAPD fragments, of which 81 （88%） were polymorphic. Of these, 13 were unique to accession ‘Pak85559＇. Each primer produced four to nine amplified products with an average of 6.13 bands per primer. Based on pairwise comparisons of RAPD amplification products, Nei and Li＇s similarity coefficients were calculated to evaluate the relationships among the accessions. Pairwise similarity indices were higher among the oilseed accessions and cultivars showing narrow ranges of 0.77-0.99. An unweighted pair-group method with arithmetic averages cluster analysis based on these genetic similarities placed most of the collections and oilseed cultivars close to each other, showing a low level of polymorphism between the accessions used. However, the clusters formed by oilseed collections and cultivars were comparatively distinct from that of advanced breeding lines. Genetically, all of the accessions were classified into a few major groups and a number of individual accessions. Advanced breeding lines were relatively divergent from the rest of the accessions and formed independent clusters. Clustering of the accessions did not show any pattern of association between the RAPD markers and the collection sites. A low level of genetic variability of oilseed mustard was attributed to the selection for similar traits and horticultural uses. Perhaps close parentage of these accessions further contributed towards their little diversity. The study demonstrated that RAPD is a simple and fast technique to compare the genetic relationship and pattern of variation among the gene pool of this crop.

Binary star detection in the open cluster King 1 field

A rarely studied open cluster,King 1 is observed using the 1.3-m telescope equipped with a 2 k×4 k CCD at Vainu Bappu Observatory,*** analyze the photometric data obtained from CCD observations in both B and V *** of 132 detected stars in the open cluster King 1 field,we have identified four stellar variables,and two among them are reported as newly detected binary *** parallax values from Gaia DR2 suggest that the open cluster King 1 is in the background of these two detected binary systems,falling along the same line of sight,giving rise to different parallax *** analysis was carried out using Phase Dispersion minimization(PDm)and the Lomb-Scargle(LS)method for all the detected *** Of Eclipsing Binari Es(PHOEBE)is extensively employed to model various stellar parameters of both the detected binary *** on the modeling results obtained from this work,one of the binary systems is reported for the first time as an Eclipsing Detached(ED)and the other as an Eclipsing Contact(EC)binary of W-type W Uma.

Aquatic fungi play an important role in litter decomposition in aquatic *** in view the importance of aquatic fungi,diversity of these organisms was studied in two freshwater ecosystems of Gaur River and Khurji Nala Jabalpur madhya Pradesh *** the present study,34 fungal species and 6 chromista species have been recorded on decomposing *** number of fungal genera were recorded in submerged litter samples of Tectona grandis followed by Dendroclamus *** maximum percentage frequencies of various fungal species which colonize litter of different tree species were also *** cluster analysis revealed that different fungal communities on various plant hosts vary significantly.

Using a bidentate ligand, a Schiff base 4-（{[（Z）-（2-hydroxyphenyl）methyldene]amino}methyl）cyclohexanecarboxylic acid, complex I（mL2, where m = Cd（Ⅱ）） was synthesized and characterized by IR spectroscopy and X-ray crystallography. It crystallizes in monoclinic system, space group C2/c, m_r = 685.04, 0.34 mm × 0.26 mm × 0.14 mm, a = 54.050（3）, b = 6.1220（3）, c = 9.5081（5） A, β = 90.229（2）°, Z = 4, C_（30） H_（40） CdN_2 O_9 and V = 3146.2（3） A^3. Complex I was screened for its bactericidal activity against four bacterial strains Chromohalobactersalexigens, Chromohalobacterisraelensis, Halomonashalofila and Halomonassalina and fungicidal activity against Aspergillusnigerand Aspergillusflavus. Antimicrobial activity results showed that the synthesized complex displayed good anti-fungal activity against two fungal strains. It was found to be inactive against four different bacterial strains analyzed and was negative for bactericidal effect.

1,2-Benzothiazine derivatives methyl 3-methoxy-4-oxo-3,4-dihydro-2H-benzo[e] [1,2]thiazine-3-carboxylate 1,1-dioxide（1） and methyl 2-ethyl-3-hydroxy-4-oxo-3,4-dihydro-2Hbenzo[e][1,2]thiazine-3-carboxylate 1,1-dioxide（2） were synthesized, and characterized by spectroscopic techniques; 1H-NmR and infrared（IR） spectroscopy. Crystals of 1 and 2 were grown by slow evaporation of methanol and ethyl acetate, respectively and their crystal structures were investigated by single-crystal X-ray diffraction analysis. Geometric properties were calculated by the B3 LYP method of density functional theory（DFT） at the 6-31G＋（d） basis set to compare with the experimental data. Simulated properties were found in strong agreement with the experimental ones. Intermolecular forces have also been modeled in order to investigate the strength of packing and strong hydrogen bonding was observed in both compounds 1 and 2. Electronic properties such as Ionization Potential（IP）, Electron Affinities（EA） and coefficients of the highest occupied molecular orbital（HOmO） and the lowest unoccupied molecular orbital（LUmO） of com- pounds 1 and 2 were simulated for the first time.
