Hepatocyte nuclear factor 1 alpha(HNF1A),hepatocyte nuclear factor 4 alpha(HNF4A),and forkhead box protein A2(FOXA2)are key transcription factors that regulate a complex gene network in the liver,cre-ating a regulatory transcriptional *** Encode and ChIP-Atlas databases identify the recognition sites of these transcription factors in many glycosyltransferase *** in silico analysis of HNF1A,*** FOXA2 binding to the ten candidate glyco-genes studied in this work confirms a significant enrich-ment of these transcription factors specifically in the *** previous studies identified HNF1A as a master regulator of fucosylation,glycan branching,and galactosylation of plasma ***,we aimed to functionally validate the role of the three transcription factors on downstream glyco-gene transcriptional expression and the possible effect on glycan *** used the state-of-the-art clus-tered regularly interspaced short palindromic repeats/dead Cas9(CRISPR/dCas9)molecular tool for the downregulation of the HNF1A,HNF4A,and FOXA2 genes in HepG2 cells-a human liver cancer cell *** results show that the downregulation of all three genes individually and in pairs affects the transcrip-tional activity of many glyco-genes,although downregulation of glyco-genes was not always followed by an unambiguous change in the corresponding glycan *** effect is better seen as an overall change in the total HepG2 N-glycome,primarily due to the extension of biantennary *** propose an alternative way to evaluate the N-glycome composition via estimating the overall complexity of the glycome by quantifying the number of monomers in each glycan *** also propose a model showing feedback loops with the mutual activation of HNF1A-FOXA2 and HNF4A-FOXA2 affecting glyco-genes and protein glycosylation in HepG2 cells.

The benefits of regular physical activity are well ***,few studies have examined the effectiveness of integrating physical activity(PA)into curricula within a post-secondary *** investigate the incorporation of PA into medical curriculum,we developed a series of optional exercise-based review sessions designed to reinforce musculoskeletal(mSK)anatomy course *** synchronous sessions were co-taught by a group fitness instructor and an anatomy *** fitness instructor would lead students through both strength and yoga style exercises,while the anatomy instructor asked questions about relevant anatomical structures related to course material previously *** the sessions,participants were asked to evaluate the classes on their self-reported exam preparedness in improving mSK anatomy knowledge,PA levels,and mental *** participants completed surveys;a majority agreed that the classes increased understanding of mSK concepts(90.0%)and activity levels(97.7%).many(70.0%)felt that the classes helped reduce *** majority of respondents(90.0%)agreed that the classes contributed to increased feelings of social ***,medical students saw benefit in PA based interventions to supplement mSK course *** with increasing activity levels and promoting health behaviours,integrating PA into medical curriculum may improve comprehension of learning material,alleviate stress and foster social connectivity among medical students.

Superspreaders(individuals with a high propensity for disease spread)have played a pivotal role in recent emerging and re-emerging *** disease outbreak studies,host heterogeneity based on demographic(***,sex,vaccination status)and environmental(***,urban/rural residence,clinics)factors are critical for the spread of infectious diseases,such as Ebola and middle East Respiratory Syndrome(mERS).Transmission rates can vary as demographic and environmental factors are altered naturally or due to modified behaviors in response to the implementation of public health *** this work,we develop stochastic models to explore the effects of demographic and environmental variability on human-to-human disease transmission rates among superspreaders in the case of Ebola and *** show that the addition of environmental variability results in reduced probability of outbreak occurrence,however the severity of outbreaks that do occur *** observations have implications for public health strategies that aim to control environmental variables.

Spinal cord injury remains a major cause of disability in young adults,and beyond acute decompression and rehabilitation,there are no pharmacological treatments to limit the progression of injury and optimize recovery in this *** the thorough investigation of the complement system in triggering and propagating cerebral neuroinflammation,a similar role for complement in spinal neuroinflammation is a focus of ongoing *** this work,we survey the current literature investigating the role of complement in spinal cord injury including the sources of complement proteins,triggers of complement activation,and role of effector functions in the *** study relevant data demonstrating the different triggers of complement activation after spinal cord injury including direct binding to cellular debris,and or activation via antibody binding to damage-associated molecular *** effector functions of complement have been implicated in spinal cord injury,and we critically evaluate recent studies on the dual role of complement anaphylatoxins in spinal cord injury while emphasizing the lack of pathophysiological understanding of the role of opsonins in spinal cord *** this pathophysiological review,we systematically review the different translational approaches used in preclinical models of spinal cord injury and discuss the challenges for future translation into human *** review emphasizes the need for future studies to dissect the roles of different complement pathways in the pathology of spinal cord injury,to evaluate the phases of involvement of opsonins and anaphylatoxins,and to study the role of complement in white matter degeneration and regeneration using translational strategies to supplement genetic models.

Introduction: Sexual risk behaviour among adolescents is a public health problem worldwide. Widely studied in the Western countries, this problem has only been addressed partially in Brazzaville. Thus, 12 years later, we conducted this study with the aim of analysing the sexual behaviour of adolescents in Congo. Patients and Methods: This was a cross-sectional and analytical study from January to August 2019 (8 months). It involved 2000 adolescents residing in the departments of Brazzaville and Pointe-Noire constituting the urban population, and those of Cuvette and Bouenza, the rural population. Results: A total of 1167 adolescents reported being sexually active. The mean age at first sexual intercourse differed between the two sexes (p 0.0005). Of the adolescents surveyed 96.1% (n = 1122) claimed to have ever heard of HIV/AIDS infection and 8.7% (n = 101) mily:"">mily:Verdana;">to have ever contracted sexually transmitted infections. The main channel of information was school in 57.7%. Condoms were the only means of protection (85.5%). The factors associated with risky sexual behaviour were, on the one hand, those related to the adolescent (lack of schooling, orphan status, alcohol and tobacco consumption, paid employment and possession of a mobile phone). On the other hand, those related to parents (low level of education, mother’s employment status and single-parent household structure).
