Immunogenicity and mechanisms impairing the response to vaccines in inflammatory bowel disease

Inflammatory bowel disease(IBD) is an immunological disorder that is usually treated with immunosuppressive therapy,potentially leading to increases in vulnerability to infections. Although many infections can be pre-vented by vaccination,vaccination coverage in these patients in clinical practice is insufficient. Therefore,the seroprotection condition should be verified,even for routine vaccines,such as hepatitis B or pneu-mococcus. Response to vaccines in IBD patients is thought to be impaired due to the immunological alterations generated by the disease and to the immunomodulatory treatments. The immunogenicity of hepatitis B,influenza,and pneumococcal vaccines is impaired in IBD patients,whereas the response to papillomavirus vaccine seems similar to that observed in the healthy population. On the other hand,data on the immunogenicity of tetanus vaccine in IBD patients are conflicting. Studies assessing the response to measles-mumps-rubella,varicella,and herpes zoster vaccines in IBD patients are scarce. The cellular and molecular mechanisms responsible for the impairment of the response to vaccination in IBD patients are poorly understood. Studies aiming to assess the response to vaccines in IBD patients and to identify the mechanisms involved in their immunogenicity are warranted. A better understanding of the immune response,specifically to vaccines,in patients with immune-mediated diseases(such as IBD),is crucial when developing vaccines that trigger more potent immunologic responses.

Error induced by the estimation of the corneal power and the effective lens position with a rotationally asymmetric refractive multifocal intraocular lens

AIm : To evaluate the prediction error in intraocular lens(IOL) power calculation for a rotationally asymmetric refractive multifocal IOL and the impact on this error of the optimization of the keratometric estimation of the corneal power and the prediction of the effective lens position(ELP).mETHODS: Retrospective study including a total of 25 eyes of 13 patients(age, 50 to 83y) with previous cataract surgery with implantation of the Lentis mplus LS-312 IOL(Oculentis Gmb H, Germany). In all cases, an adjusted IOL power(P IOLadj) was calculated based on Gaussian optics using a variable keratometric index value(n kadj) for the estimation of the corneal power(P kadj) and on a new value for ELP(ELP adj) obtained by multiple regression *** P IOLadj was compared with the IOL power implanted(P IOLReal) and the value proposed by three conventional formulas(Haigis, Hoffer Q and Holladay Ⅰ).RESULTS: P IOLReal was not significantly different than P IOLadj and Holladay IOL power(P >0.05). In the Bland and Altman analysis, P IOLadj showed lower mean difference(-0.07 D) and limits of agreement(of 1.47 and-1.61 D)when compared to P IOLReal than the IOL power value obtained with the Holladay formula. Furthermore, ELP adj was significantly lower than ELP calculated with other conventional formulas(P nd was found to be dependent on axial length, anterior chamber depth and P kadj. COnCLUSIOn: Refractive outcomes after cataract surgery with implantation of the multifocal IOL Lentis mplus LS-312 can be optimized by minimizing thekeratometric error and by estimating ELP using a mathematical expression dependent on anatomical factors.

Understanding variability in seed germination among populations is essential for planning an effective germplasm collection for restoration and conservation *** knowledge of germination and dormancy patterns among populations of desert grasses is crucial for determining the potential of the species and populations to be used for restoration and conservation as well as forage *** in seed germination of Panicum turgidum Forssk and Pennisetum divisum(Gmel.)*** the desert of Kuwait was evaluated in different populations in may *** of seed germination(25 seeds and 4 replicates)was conducted for each population at night/day temperatures of 15℃/20℃and 20℃/30℃under the following light condition:continuous darkness or 12 h/12 h light/*** showed that seed masses of both species strongly varied according to their seed provenances,and both species produced heavier seeds in population with a higher soil electrical *** germination percentage considerably varied between two species,and the variation in *** was greater(17%–49%)than that of ***(72%–93%).Germination percentage in *** was greater at high temperature(20℃/30℃)than at low temperature(15℃/20℃).However,temperature regimes had no effect on germination percentage of *** *** germination time of both species exhibited significant inter-population *** result is especially relevant to assure the selection of the best population of each species and the regeneration success of the *** this,inter-population variability also provides valuable information for enhancing our understanding of the mechanisms that regulate seed germination and how they might be related to seed provenance.

Bacterial diversity in Fez tanneries and morocco's Binlamdoune River,using 16S RnA gene based fingerprinting

Tannery wastewater causes serious ecological and sanitary damage. Chemical analysis of water from Binlamdoune River of the medina of Fez was conducted and the results revealed the presence of toxic elements from tanneries and other industrial activities, which strongly affected water quality. To determine the effectiveness of bioremediation for depollution, we studied the abundance and diversity of bacteria residing in these polluted environments. Conducting denaturing gradient gel electrophoresis （PCR-DGGE） of the 16S rDnA area using primers related to bacteria showed a bacterial community belonging to eubacterial groups, that is, Epsilonproteobacteria, Clostridia, Lactobacillales, Bacteroidetes, Gammaproteobacteria, and Alphaproteobacteria. In addition, cloning displayed the presence of clones belonging to the Firmicutes group. moreover, scanning electron microscopy revealed a significant heterogeneity of microorganism forms and structures. These endogenous microbes could have a significant role in the purification of Binlamdoune River and Fez tannery wastewater.

Association of Congenital Heart Defects (CHD) with Factors Related to maternal Health and Pregnancy in newborns in Puerto Rico

Background:Given the pervasive issues of obesity and diabetes both in Puerto Rico and the broader United States,there is a compelling need to investigate the intricate interplay among body mass index(BmI),pregesta-tional,and gestational maternal diabetes,and their potential impact on the occurrence of congenital heart defects(CHD)during neonatal ***:Using the comprehensive System of Vigilance and Surveillance of Congenital Defects in Puerto Rico,we conducted a focused analysis on neonates diagnosed with CHD between 2016 and *** assessment encompassed a range of variables,including maternal age,gestational age,BmI,pregestational diabetes,gestational diabetes,hypertension,history of abortion,and presence of ***:A cohort of 673 patients was included in our *** average maternal age was 26 years,within a range of 22 to 32 *** mean gestational age measured 39 weeks,with a median span of 38 to 39 *** the 673 patients,274(41%)mothers gave birth to neonates diagnosed with *** this group,22 cases were linked to pre-gestational diabetes,while 202 were not;20 instances were associated with gestational diabetes,compared to 200 without;and 148 cases exhibited an overweight or obese BmI,whereas 126 displayed a normal ***:We identiﬁed a statistically signiﬁcant correlation between pre-gestational diabetes mellitus and the occurrence of ***,our analysis did not show a statistically signiﬁcant association between maternal BmI and the likelihood of *** results may aid in developing effective strategies to prevent and manage CHD in neonates.

Antioxidant role of heme oxygenase-1 in prehepatic portal hypertensive rats

AIm： To study the effect of bilirubin on the oxidative liver status and the activity and expression of heine oxygenase-1 （HO-1） in rat liver injury induced by prehepatic portal hypertension. mETHODS： Wistar male rats, weighing 200-250 g, were divided at random into two groups： one group with prehepatic portal hypertension （PH） induced by regulated prehepatic portal vein ligation （PPVL） and the other group corresponded to sham operated rats. Portal pressure, oxidative stress parameters, antioxidant enzymes, HO-1 activity and expression and hepatic sinusoidal vasodilatation were measured. RESULTS： In PPVL rats oxidative stress was evidenced by a marked increase in thiobarbituric acid reactive substances （TBARS） content and a decrease in reduced glutathione （GSH） levels. The activities of liver antioxidant enzymes, superoxide dismutase （SOD）, catalase （CAT） and glutathione peroxidase （GSH-Px） were also diminished while activity and expression of HO-1 were enhanced. Administration of bilirubin （5μmol/kg body weight） 24 h before the end of the experiment entirely prevented all these effects. Pretreatment with Sn-protoporphyrin IX （Sn-PPIX）（100 μg/kg body weight, i.p.）, a potent inhibitor of HO, completely abolished the oxidative stress and provoked a slight decrease in liver GSH levels as well as an increase in lipid peroxidation. Besides, carbon monoxide, another heme catabolic product, induced a significant increase in sinusoidal hepatic areas in PPVL group. Pretreatment of PPVL rats with Sn-PPIX totally prevented this effect COnCLUSIOn： These results suggest a beneficial role of HO-1 overexpression in prehepatic portal hypertensive rats.
