A salient feature of metastasis is the ability of different tu- mor types to colonize the same or different organ sites. Many genetic and epigenetic events have been identified that con- tributing to the metastatic cascades, including migration, lo- cal invasion, intravasation, survival in the circulation, ex- travasation and colonization, and some key molecules might have dual functions promoting metastasis by providing both local advantage from malignant progression in the primary tu- mor and a distal advantage from infiltration of distant organs. Nevertheless, the metastatic process remains poorly under- stood and the insight into the molecular events that initiate and/or sustain this process remains incomplete.

Objective:Epigenetic aberration plays an important role in the development and progression of hepatocellular carcinoma(HCC).However,the alteration of RNA N6-methyladenosine(m6A)modifications and its role in HCC progression remain *** therefore aimed to provide evidence using bioinformatics ***:We comprehensively analyzed the m6A regulator modification patterns of 605 HCC samples and correlated them with metabolic alteration *** elucidated 390 gene-based m6A-related signatures and defined an m6Ascore to quantify m6A *** then assessed their values for predicting prognoses and therapeutic responses in HCC ***:We identified 3 distinct m6A modification patterns in HCC,and each pattern had distinct metabolic *** evaluation of m6A modification patterns using m6Ascores could predict the prognoses,tumor stages,and responses to sorafenib treatments of HCC patients.A nomogram based on m6Ascores showed high accuracy in predicting the overall survival of *** area under the receiver operating characteristic curve of predictions of 1,3,and 5-year overall survivals were 0.71,0.69,and 0.70 in the training cohort,and in the test cohort it was 0.74,0.75,and 0.71,respectively.M6Acluster C1,which corresponded to hypoactive mRNA methylation,lower expression of m6A regulators,and a lower m6Ascore,was characterized by metabolic hyperactivity,lower tumor stage,better prognosis,and lower response to sorafenib *** contrast,m6Acluster C3 was distinct in its hyperactive mRNA methylations,higher expression of m6A regulators,and higher m6Ascores,and was characterized by hypoactive metabolism,advanced tumor stage,poorer prognosis,and a better response to *** m6Acluster,C2,was intermediate between C1 and ***:HCCs harbored distinct m6A regulator modification patterns that contributed to the metabolic heterogeneity and diversity of *** of m6A gene signatures and the m6Ascore

Hepatocellular carcinoma(HCC)represents an extremely poor prognostic cancer,which is mainly due to the high frequency of metastasis/recurrence after surgical *** the molecular mechanisms involved in HCC metastasis could be helpful in the pre-diction and early diagnosis of HCC recurrence and could also provide new therapeutic targets for HCC *** the recent decade,we analyzed the genomic aberrations of the clinical specimens,as well as the metastatic models and cell lines of human HCC to identify the genetic mar-kers related to HCC metastasis and to verify their clinical values in the prediction and control of metastasis of *** the comparative genomic hybridization(CGH)technique,we compared the differences of chromosomal aberrations between primary HCC tumors and their matched metastatic lesions,and found that chromosome 8p deletions might contribute to HCC *** novel finding was further confirmed by comparison between nude mice models of HCC with different meta-static *** the more sensitive genome-wide microsatellite analysis,8p deletion was defined to 8p23.3 and 8p11.2,which are two likely regions harboring meta-stasis-related genes of ***‘8p-specific’microar-rays,two novel metastatic suppressors(HTPAP and MRSA)were identified,and were proven to suppress in vitro invasion and in vivo metastasis of *** studies indicate that 8p deletion detected in HCC or cir-culating plasma DNA of patients is a useful predictor for metastatic recurrence and prognosis,even for patients with early stage *** novel findings are regarded as important advances in the study of the molecular mechanisms of HCC metastasis,which provide not only a holistic view on the molecular cytogenetic bases of HCC metastasis,but also candidate regions for further study to identify metastatic suppressor genes.

Recently, we have analyzed expressional transcriptome and proteome for a number of cancer cell lines, including 8 hepatocellular carcinoma （HCC） ones [1,2]. These 8 HCC cell lines consist of 6 metastatic （all with mutant p53） and 2 non-metastatic （with a mutant and a lost p53） ones, being all HBV traceable for relatively close genetic-backgrounds. We have analyzed the related dataset and obtained transcrip- tome （16353 genes） and proteome （7861 proteins） of these 8 HCC cell lines, and explored a group of up-regulated and down-regulated genes [3].

Cancer metastasis is the major cause of cancer-related deaths and accounts for poor therapeutic outcomes.A metastatic cas-cade is a series of complicated biological processes.N6-methyladenosine(m^(6)A)is the most abundant and conserved epi-transcriptomic modification in eukaryotic cells,which has great impacts on RNA production and metabolism,including RNA splicing,processing,degradation and *** evidence demonstrates that m^(6)A plays a critical role in regulating cancer ***,there is a lack of studies that review the recent advances of m^(6)A in cancer ***,we systematically retrieved the functions and mechanisms of how the m^(6)A axis regulates metastasis,and especially summarized the organ-specific liver,lung and brain metastasis mediated by m^(6)A in various ***,we discussed the potential application of m^(6)A modification in cancer diagnosis and therapy,as well as the present limitations and future perspectives of m^(6)A in cancer *** review provides a comprehensive knowledge on the m^(6)A-mediated regulation of gene expression,which is helpful to extensively understand the complexity of cancer metastasis from a new epitranscriptomic point of view and shed light on the developing novel strategies to anti-metastasis based on m^(6)A alteration.

Rapamycin enhances the anti-tumor activity of cabozantinib in cMet inhibitor-resistant hepatocellular carcinoma

Cabozantinib,mainly targeting cMet and vascular endothelial growth factor receptor 2,is the second-line treatment for patients with advanced hepatocellular carcinoma(HCC).However,the lower response rate and resistance limit its enduring clinical *** this study,we found that cMet-low HCC cells showed primary resistance to cMet inhibitors,and the combination of cabozantinib and mammalian target of rapamycin(mTOR)inhibitor,rapamycin,exhibited a synergistic inhibitory effect on the in vitro cell proliferation and in vivo tumor growth of these ***,the combination of rapamycin with cabozantinib resulted in the remarkable inhibition of AKT,extracellular signal-regulated protein kinases,mTOR,and common downstream signal molecules of receptor tyrosine kinases;decreased cyclin D1 expression;and induced cell cycle ***,rapamycin enhanced the inhibitory effects of cabozantinib on the migration and tubule formation of human umbilical vascular endothelial cells and human growth factor-induced invasion of cMet inhibitor-resistant HCC cells under hypoxia *** effects were further validated in xenograft *** conclusion,our findings uncover a potential combination therapy of cabozantinib and rapamycin to combat cabozantinib-resistant HCC.

通过近20年的努力，肝癌转移复发研究取得诸多进展，主要包括：（1）发现了包括骨桥蛋白在内的一组转移复发相关分子，建立并优化了多分子转移预测模型。（2）发现肿瘤微环境免疫反应状态失衡是促进转移的重要方面，可用于预测转移复发。（3）发现并证实干扰索可预防术后复发，并且miR-26a低表达者更获益。（4）液体活检和免疫治疗的突破为HCC转移复发的预测与防治带来光明前景。但是，这些预测模型尚需多中心研究进一步验证；辅助性肝动脉化学疗法栓塞术和索拉菲尼靶向治疗的作用尚需进一步评估。

Intercellular cross-talk plays important roles in cancer progression and *** how these cancer cells interact with each other is still largely *** released by tumor cells have been proved to be effective cell-to-cell signal *** explored the functional roles of exosomes in metastasis and the potential prognostic values for hepatocellular carcinoma(HCC).Exosomes were extracted from HCC cells of different metastatic *** metastatic effects of exosomes derived from highly metastatic HCC cells(HMH)were evaluated both in vitro and in *** proteins were identified with iTRAQ mass spectrum and verified in cell lines,xenograft tumor samples,and functional *** released by HMH significantly enhanced the in vitro invasion and in vivo metastasis of low metastatic HCC cells(LMH).S100 calcium-binding protein A4(S100A4)was identified as a functional factor in exosomes derived from HMH.S100A4r,ch exosomes significantly promoted tumor metastasis both in vitro and in vivo compared with S100A4^(rich) exosomes or ***,exosomal S100A4 could induce expression of osteopontin(OPN),along with other tumor metastasis/stemness-related *** S100A4 activated OPN transcription via STAT3 *** patients with high exosomal S100A4 in plasma also had a poorer *** conclusion,exosomes from HMH could promote the metastatic potential of LMH,and exosomal S100A4 is a key enhancer for HCC metastasis,activating STAT3 phosphorylation and up-regulating OPN *** suggested exosomal S100A4 to be a novel prognostic marker and therapeutic target for HCC metastasis.
