[Objectives]This study was conducted to establish a qualitative method for rapid detection of pesticide residues in feed.[Methods]A rapid screening method for 82 pesticide residues in feed was established using ultra liquid chromatography coupled with quadrupole-time of flight mass spectrometry(UPLC-Q-TOF-MS)combined with optimized pretreatment methods.[Results]Feed samples were extracted with 0.2%formic acid-acetonitrile solution,shaken,centrifuged,and re-centrifuged,and the supernatant was filtered with a 0.22μm filter membrane for *** samples were separated on a Waters ACQUITY UPLC HSS T3 chromatographic column(2.1 mm×50 mm,1.8μm),and subjected to gradient elution with methanol-water solution as the mobile phase,and scanning was performed in ESI+mode.82 compounds could be accurately qualitatively detected in 13 min.[Conclusions]This method is simple to operate,time-saving,and can be applied to rapid screening of large-scale *** study provides a theoretical basis for the development of new methods in the future.

[Objectives]This study was conducted to investigate the main factors affecting the aerobial plate count in raw milk.[Methods]Drinking water,medicated baths and raw milk under different storage and transportation conditions were detected for the values of aerobial plate count to analyze their effects on the aerobial plate count in raw milk.[Results]Disinfection of drinking water tanks could significantly reduce the aerobial plate count in *** use of medicated baths before and after milking could effectively reduce the aerobial plate count and had a significant bactericidal *** growth of microorganisms in raw milk stored below 4℃was relatively *** disinfecting drinking water tanks and disinfecting nipples before and after milking could reduce the aerobial plate count in the tanks and *** raw milk was extruded,the temperature should decrease to 0-4℃within 2 h,and the storage time should not exceed 48 h,which could effectively control the aerobial plate count in raw milk.[Conclusions]This study provides a reference for scientific control of the aerobial plate count in raw milk.

[Objectives]This study was conducted to study the effects of whole-plant corn silage and whole-plant wheat silage on growth performance and economic benefits of beef cattle.[Methods]Thirty Simmental crossbred cattle were selected and randomly divided based on their weight into two groups,15 cows in each group,with an average weight of 398 *** group was fed the same concentrated feed,and the whole-plant corn silage and wheat silage were fed as roughage feed,*** experimental period was 90 d.[Results](1)There was no significant difference in the initial weight of beef cattle between test group 1 and test group 2(P>0.05).Compared with group 2,group 1 had a higher final weight,average daily gain,and dry matter intake of beef cattle,without significant differences(P>0.05).Group 1 was 0.58%,1.99%,and 1.43%higher than group 2,*** feed-to-gain ratio of test group 1 decreased by 0.54%compared with test group 2(P>0.05),with no significant difference therebetween.(2)The economic benefit of group 1 was 11.45 yuan/(day·capita),while the economic benefit of group 2 was 2.41 yuan/(day·capita),and group 1 was 9.04 yuan/(day·capita)higher than group 2.[Conclusions]Whole-plant corn silage for feeding beef cattle has the advantages of good weight gain and low breeding cost,and can be popularized in beef cattle fattening.

[目的]研究饲粮中不同添加比例的全株玉米青贮饲喂西门塔尔杂交肉牛效果。[方法]选取体重378 kg左右的西门塔尔杂交牛50头,随机分为5组,饲喂相同的精饲料,粗饲料中分别添加20%、40%、60%、80%和100%的全株玉米青贮作为试验Ⅰ、Ⅱ、Ⅲ、Ⅳ、Ⅴ组,试验期70 d。[结果]随着全株玉米青贮添加比例增加,试验牛末重逐渐增加;饲粮中全株玉米青贮添加比例对肉牛平均日增重有极显著差异,Ⅴ组平均日增重极显著高于Ⅰ组14.40%;各组间料重比存在显著性差异。试验Ⅴ组增重收入和养殖效益最高,分别为35.75元/(d·头)和9.47元/(d·头),试验Ⅴ组养殖效益分别高出Ⅰ、Ⅱ、Ⅲ、Ⅳ组7.34、5.34、3.42和1.05元/(d·头)。[结论]随着饲粮中全株玉米青贮比例增加,肉牛增重和养殖收益均呈上升趋势,当全株玉米青贮比例为80%~100%时,肉牛增重效果较好,养殖效益较佳。

旨在研究饲粮能量水平对育成牛养分表观消化率、生长性能和瘤胃发酵的影响。本研究选取8月龄中国荷斯坦育成奶牛45头,采用单因素完全随机设计,分为3组,每组15头牛。Ⅰ组饲喂低能日粮(NEL5.64 MJ·kg-1),Ⅱ组饲喂中能日粮(NEL5.94MJ·kg-1),Ⅲ组饲喂高能日粮(NEL6.21 MJ·kg-1),各试验组的饲粮蛋白质水平基本相同(CP14.04%)。试验期110d。测定每组育成牛的生长性能指标(体重、体高、体长、胸围、管围、乳头长度、乳头间距);采集粪样用于测定营养物质表观消化率;采集瘤胃液样品用于测定瘤胃发酵指标(pH、微生物蛋白、氨态氮、挥发性脂肪酸)。结果表明:1)与试验Ⅰ、Ⅱ组相比,试验Ⅲ组中性洗涤纤维(NDF)和酸性洗涤纤维(ADF)表观消化率显著降低(P0.05)。2)Ⅰ、Ⅱ、Ⅲ组育成牛平均日增重(ADG)分别为0.70、0.80、0.91kg·d-1,与Ⅰ组相比,Ⅱ组和Ⅲ组分别显著提高了14.28%和30.00%(P0.05)。3)与Ⅰ组和Ⅲ组相比,Ⅱ组牛前乳头平均长度分别高出4.15%(P>0.05)和9.06%(P0.05);氨态氮(NH3-N)含量以Ⅱ组最低,较Ⅰ组显著降低了3.56%(P0.05);Ⅲ组丙酸含量分别比Ⅰ、Ⅱ组高4.99%(P0.05)。综上所述,在本试验中,8～11月龄育成牛日粮适宜产奶净能水平为5.94MJ·kg-1,粗蛋白质水平为14.03%。

[Objectives]A rapid screening and analysis method for 26 organophosphorus agrochemicals in fresh milk was established using ultra performance liquid chromatography coupled with quadrupole-time of flight mass spectrometry.[Methods]Raw milk was extracted with acetonitrile solution containing 0.2%formic acid by volume,and purified with a Dikma ProElut QuECHERS solid phase extraction *** compounds were separated on a Waters ACQUITY UPLC HSS T3 chromatographic column(2.1 mm×50 mm,1.8μm)with methanol-water solution as a mobile phase for gradient elution,and through scanning with an electrospray ion source in positive ion mode,26 kinds of organophosphorus agrochemicals could be accurately qualitatively determined within 10 min.[Results]When using formic acid acetonitrile with a volume fraction of 0.2%,there were more types of detected compounds and a greater recovery;and using B cartridge could effectively eliminate the interference of non-polar substances such as phospholipids,achieve higher number of detected compounds than those of A and C,and well separate the 26 kinds of agrochemical residues.[Conclusions]This study provides a reference method for the rapid screening of agrochemical residues in dairy cows in the future.

Liquid chromatography-high resolution time-of-flight mass spectrometry has high-resolution and high-throughput detection functions, and has gradually begun to be applied in the detection of milk and dairy products. This paper summarized the application of milk and dairy products in liquid chromatography-high resolution time-of-flight mass spectrometry, laying a foundation for the development of new methods.

为了解唐山地区生鲜乳体细胞(SCC)变化规律,笔者对唐山地区的生鲜乳进行了调查研究。分别于2013年-2018年的5月及11月每次采集50个生鲜乳样品,每年共采集100个样品,比较分析了生鲜乳SCC的变化规律。结果表明,2013年-2018年间,唐山地区生鲜乳中SCC呈现出下降的趋势,全年均值由2013年的70.95×104/m L下降到2018年的17.95×104/m L,在2016年生鲜乳SCC值达到最低(14.71×104/m L)。2013年-2018年,唐山地区生鲜乳中SCC小于40×104/m L的比例呈上升趋势,由2013年的5%增长到2018年的98%,但每年度生鲜乳SCC数值的标准差和变异系数均较大。2013年-2018年,唐山地区生鲜乳体细胞评分(SCS)大体呈现出好转的趋势,各年度生鲜乳SCS之间差异极显著(P<0.01)。

为了研究谷草与玉米秸秆黄贮不同配比饲粮对肉牛屠宰性能、肉品质与经济效益的影响,试验选取体重相近[(440.89±38.49)kg]、体况良好的西门塔尔杂交牛45头,随机分为3组(试验Ⅰ组、试验Ⅱ组、试验Ⅲ组),每组15头,分别饲喂谷草与玉米秸秆黄贮不同配比(1∶9,2∶8,3∶7)的日粮(精料补充料及精粗比相同),饲养100 d(预试期10 d,正试期90 d),试验结束后每组随机选择接近平均体重的5头试验牛屠宰,测定屠宰指标、肉品质指标、肉常规营养指标、脂肪酸组成及含量。结果表明:各组试验牛胴体重、屠宰率、净肉率差异不显著(P>0.05),但试验Ⅱ组牛各项屠宰指标与Ⅰ组、Ⅲ组相比均有所提高。随着谷草比例的增加,肉色中红度值逐渐降低,Ⅲ组比Ⅰ组、Ⅱ组分别降低13.10%和9.77%(P0.05)。各试验组牛背最长肌中饱和脂肪酸(SFA)、单不饱和脂肪酸(MUFA)、多不饱和脂肪酸(PUFA)之间无显著差异(P>0.05)。随着谷草比例的升高,SFA、PUFA有下降的趋势,MUFA有上升的趋势。从养殖效益来分析,以Ⅱ组牛最高,为3 471.83元/头,比Ⅰ组高879.55元/头,比Ⅲ组高999.11元/头。说明可以用20%的谷草来替代玉米秸秆黄贮,且肉牛屠宰性能与肉品质有所改善,经济效益最好。

本文研究了用液相色谱串联飞行时间质谱建立生乳中29种兽药残留的快速检测方法。生乳样品采用0.2%甲酸乙腈溶液提取上清液,采用Dikma ProElut QuECHERS净化管净化后测定。目标化合物通过Waters ACQUITY UPLC HSS T3色谱柱分离,以乙腈和1.0%甲酸水溶液为洗脱剂进行程序洗脱,正离子扫描模式检测。结果表明,通过对数据进行靶向分析,20.0 min内生乳中29种兽药残留能够同时被检测出来,其中Mass Error在±2.0 mg/kg之内,Library Score得分均在90分以上,准确性高,操作简便。
