目的观察以6-磷酸甘露糖修饰的白蛋白(M6P26-HSA)作为特异性载体,将甘草次酸(GA) 靶向释放到肝星状细胞治疗肝纤维化的效果。方法用125I记由M6P26-HSA和GA在体外合成新的偶合物GA-HSA-M6P26,观察其在体内的器官分布情况,用双重免疫组织化学的方法观察星状细胞对GA- HSA-M6P26的选择性摄取;选用Sirius红染色观察GA-HSA-M6P26对肝纤维化时胶原沉积的影响,用定量聚合酶链反应检测GA-HSA-M6P26对Ⅰ型前胶原mRNA表达的影响。结果静脉注射后10min,GA-HSA- M6P26选择性地分布于肝脏,摄取高峰可达(5 5.093±5.404)%。双重免疫组织化学染色证实GA-HSA- M6P26主要被星状细胞选择性摄取,GA-HSA-M6P26治疗后肝脏胶原沉积明显减少,Ⅰ型前胶原和α-平滑肌肌动蛋白mRNA表达明显降低。结论GA-HSA-M6P26可以选择性地分布于肝脏星状细胞,有显著的抗肝纤维化作用。

AIM To demonstrate the feasibility of cryopreservation of peripheral blood mononuclear cells(PBMCs) for prognostic circulating tumor cell(CTC) detection in gastroesophageal *** Using 7.5 m L blood samples collected in EDTA tubes from patients with gastroesopheagal adenocarcinoma, CTCs were isolated by epithelial cell adhesion molecule based immunomagnetic capture using the Iso Flux platform. Paired specimens taken during the same blood draw(n = 15) were used to compare number of CTCs isolated from fresh and cryopreserved PBMCs. Blood samples were processed within 24 h to recover the PBMC fraction, with PBMCs used for fresh analysis immediately processed for CTC isolation. Cryopreservation of PBMCs lasted from 2 wk to 25.2 mo(median 14.6 mo). CTCs isolated from pre-treatment cryopreserved PBMCs(n = 43) were examined for associations with clinicopathological variables and survival *** While there was a significant trend to a decrease in CTC numbers associated with cryopreserved specimens(mean number of CTCs 34.4 vs 51.5, P = 0.04), this was predominately in samples with a total CTC count of > 50, with low CTC count samples less affected(P = 0.06). There was no significant association between the duration of cryopreservation and number of CTCs. In cryopreserved PBMCs from patient samples prior to treatment, a high CTC count(> 17) was associated with poorer overall survival(OS)(n = 43, HR = 4.4, 95%CI: 1.7-11.7, P = 0.0013). In multivariate analysis, after controlling for sex, age, stage, ECOG performance status, and primary tumor location, a high CTC count remained significantly associated with a poorer OS(HR = 3.7, 95%CI: 1.2-12.4, P = 0.03). CONCLUSION PBMC cryopreservation for delayed CTC isolation is a valid strategy to assist with sample collection, transporting and processing.

Growing prevalence of diabetes(type 2 as well as type 1) and its related morbidity due to vascular complications creates a large burden on medical care worldwide. Understanding the molecular pathogenesis of chronic micro-, macro- and avascular complications mediated by hyperglycemia is of crucial importance since novel therapeutic targets can be identified and tested. Thiamine(vitamin B1) is an essential cofactor of several enzymes involved in carbohydrate metabolism and published data suggest that thiamine metabolism in diabetes is deficient. This review aims to point out the physiological role of thiamine in metabolism of glucose and amino acids, to present overview of thiamine metabolism and to describe the consequences of thiamine deficiency(either clinically manifest or latent). Furthermore, we want to explain why thiamine demands are increased in diabetes and to summarise data indicating thiamine mishandling in diabetics(by review of the studies mapping the prevalence and the degree ofthiamine deficiency in diabetics). Finally, we would like to summarise the evidence for the beneficial effect of thiamine supplementation in progression of hyperglycemia-related pathology and, therefore, to justify its importance in determining the harmful impact of hyperglycemia in diabetes. Based on the data presented it could be concluded that although experimental studies mostly resulted in beneficial effects, clinical studies of appropriate size and duration focusing on the effect of thiamine supplementation/therapy on hard endpoints are missing at present. Moreover, it is not currently clear which mechanisms contribute to the deficient action of thiamine in diabetes most. Experimental studies on the molecular mechanisms of thiamine deficiency in diabetes are critically needed before clear answer to diabetes community could be given.

Following rapid infrastructure development and industrialization, the problems of water pollution and water shortage have become more severe. Whether there is safe drinking water in cities has attracted wide *** ecological risk management of drinking water project is an important means of ensuring the safety of a drinking water source. Based on ecological risk assessment and management theories, this paper establishes an ecological risk management model and assessment system with the aim of providing theoretical guidance and scientific basis for formulating a policy on the safety and protection of drinking water sources in a city. Kunming is one of the cities plagued by severe water shortage in China. Its ecological risk management of drinking water has attracted the attention of both the local government and the *** Kunming as the case study, this paper conducts a comparative analysis and assessment on three major reservoirs that face ecological risks. It highlights the existing problems and gives helpful suggestions.

*** study the role of Muscone in the IVD degeneration with the focuses on the vasculature, inflammation and apoptosis in the endplate cartilage. Methods. By excising the paraspinal musculature and posterior cervical spinal ligaments of rats, both static and dynamic cervical forces were disrupted. After surgery, some rats were fed with Muscone. Histopathologic and biochemical features of IVD cells, especially chondrocytes in endplage cartilage, were examined with a series of methods including histology, in situ hybridization for TNF-α,ELISA for IL-1α, PGE2, and 6-keto-prostaglandin F1 alpha (6-K-PGF1α), TUNEL staining, and immunostaining of Fas and bcl-2. Results. Degenerative changes in the rat model of surgery-induced cervical instability were characterized by enhanced local inflammation, and increased apoptosis and decreased blood supply in endplate cartilage. Muscone prevents the degenerative process through several ways. Firstly, it improves blood supply to endplate cartilage as shown by increased number of blood vessles and VEGF expression. Secondly, it significantly suppresses local inflammation as demonstrated by decreased concentration of PGE2, 6-K-PGF1a and IL-1α and downregulated TNF-α expression. Thirdly, it delay the apoptotic process as confirmed by decreased number of the TUNEL positive ceils, decreased Fas immunoreactivity and increased immunoreactivity for Bcl-2. Conclusions. Muscone prevents the degenerative changes caused by cervical spine instability by improving blood supply to endplate cartilage, inhibiting local inflammation, and delaying the apoptosis of chondrocyte in endplate cartilage.

In this paper, we describe the fabrication of a new biodegradable porous scaffold composed of polycaprolactone(PCL) and magnesium(Mg)micro-particles. The compressive modulus of PCL porous scaffold was increased to at least 150% by incorporating 29% Mg particles with the porosity of 74% using Micro-CT analysis. Surprisingly, the compressive modulus of this scaffold was further increased to at least 236% when the silane-coupled Mg particles were added. In terms of cell viability, the scaffold modified with Mg particles significantly convinced the attachment and growth of osteoblasts as compared with the pure PCL scaffold. In addition, the hybrid scaffold was able to attract the formation of apatite layer over its surface after 7 days of immersion in normal culture medium, whereas it was not observed on the pure PCL scaffold. This in vitro result indicated the enhanced bioactivity of the modified scaffold. Moreover, enhanced bone forming ability was also observed in the rat model after 3 months of implantation. Though bony in-growth was found in all the implanted scaffolds. High volume of new bone formation could be found in the Mg/PCL hybrid scaffolds when compared to the pure PCL scaffold. Both pure PCL and Mg/PCL hybrid scaffolds were degraded after 3 months. However, no tissue inflammation was observed. In conclusion, these promising results suggested that the incorporation of Mg micro-particles into PCL porous scaffold could significantly enhance its mechanical and biological properties. This modified porous bio-scaffold may potentially apply in the surgical management of large bone defect fixation.
