How Sugars Might Coordinate Chloroplast and Nuclear Gene Expression during Acclimation to High Light Intensities

The concept of retrograde control of nuclear gene expression assumes the generation of signals inside the chloroplasts, which are either released from or sensed inside of the organelle. In both cases, downstream signaling path- ways lead eventually to a differential regulation of nuclear gene expression and the production of proteins required in the chloroplast. This concept appears reasonable as the majority of the over 3000 predicted plastidial proteins are encoded by nuclear genes. Hence, the nucleus needs information on the status of the chloroplasts, such as during acclimation responses, which trigger massive changes in the protein composition of the thylakoid membrane and in the stroma. Here, we propose an additional control mechanism of nuclear- and plastome-encoded photosynthesis genes, taking advantage of pathways involved in sugar- or hormonal signaling. Sugars are major end products of photosynthesis and their con- tents respond very sensitively to changes in light intensities. Based on recent findings, we ask the question as to whether the carbohydrate status outside the chloroplast can be directly sensed within the chloroplast stroma. Sugars might syn- chronize the responsiveness of both genomes and thereby help to coordinate the expression of piastome- and nuclear- encoded photosynthesis genes in concert with other, more specific retrograde signals.

More than 200 research papers on the molecular phylogeny and phylogenetic biogeography of bryophytes have been published since the beginning of this millenium.These papers corroborated assumptions of a complex genetic structure of morphologically circumscribed bryophytes,and raised reservations against many morphologically justified species concepts,especially within the mosses.However,many molecular studies allowed for corrections and modifications of morphological classification schemes.Several studies reported that the phylogenetic structure of disjunctly distributed bryophyte species reflects their geographical ranges rather than morphological disparities.Molecular data led to new appraisals of distribution ranges and allowed for the reconstruction of refugia and migration routes.Intercontinental ranges of bryophytes are often caused by dispersal rather than geographical vicariance.Many distribution patterns of disjunct bryophytes are likely formed by processes such as short distance dispersal,rare long distance dispersal events,extinction,recolonization and diversification.

Natural forests and stands subj ected to little to moderate human impact are continuously declining worldwide and with these,their biodiversity and ecosystem services.Many Nothofagus forests in the south of the South American continent are in a pristine state or only moderately impacted by humans.Forest grazing by livestock,in the past and still today often practiced in a non-sustainable way is,however,increasingly under discussion to meet current environmental and socio-economic challenges.Accordingly,we investigate the regeneration of Nothofagus dombeyi,a keystone species of the Patagonian Andes,in secondary forests in Argentinian northern Patagonia,particularly addressing the role of disturbance through grazing by livestock at various intensities.We test the hypothesis that the regeneration of this tree species is favored by grazing impact and,thus,disturbance of the herb layer and soil surface.In support of our hypothesis,Nothofagus dombeyi regeneration was significantly higher in terms of individuals and height classes in moderately grazed forests.Multivariate analysis shows significant positive effects of moderate grazing pressure,herb layer cover,and the occurrence of bare soil on the regeneration of Nothofagus dombeyi.Our results show that an integration of livestock grazing and forest regeneration is possible and that agroforestry systems can be an adequate management option for stakeholders in the region.A grazing management can also be part of a forest fire prevention strategy.However,the regeneration success and grazing pressure should be continuously monitored.

Killing of p53-deficient hepatoma cells by parvovirus H-1 and chemotherapeutics requires promyelocytic leukemia protein

AIM: To evaluate the synergistic targeting and killing of human hepatocellular carcinoma (HCC) cells lacking p53 by the oncolytic autonomous parvovirus (PV) H-1 and chemotherapeutic agents and its dependence on functional promyelocytic leukemia protein (PML). METHODS: The role of p53 and PML in regulating cy-totoxicity and gene transfer mediated by wild-type (wt) PV H-1 were explored in two pairs of isogenic human hepatoma cell lines with different p53 status. Further-more,H-1 PV infection was combined with cytostatic drug treatment. RESULTS: While the HCC cells with different p53 status studied were all susceptible to H-1 PV-induced apoptosis,the cytotoxicity of H-1 PV was morepronounced in p53-negative than in p53-positive cells. Apoptosis rates in p53-negative cell lines treated by genotoxic drugs were further enhanced by a treatment with H-1 PV. In flow cytometric analyses,H-1 PV infection resulted in a reduction of the mitochondrial transmembrane potential. In addition,H-1 PV cells showed a significant increase in PML expression. Knocking down PML expression resulted in a striking reduction of the level of H-1 PV infected tumor cell death. CONCLUSION: H-1 PV is a suitable agent to circumvent the resistance of p53-negative HCC cells to genotoxic agents,and it enhances the apoptotic process which is dependent on functional PML. Thus,H-1 PV and its oncolytic vector derivatives may be considered as therapeutic options for HCC,particularly for p53-negative tumors.

Development of a Multi-Plex Electrochemiluminescent Assay for the Detection of Serum Antibody Responses to Meningococcal Conjugate Vaccines

Neisseria meningitidis is a gram negative diplococcal bacterium. Worldwide, N. meningitidis is the leading cause of bacterial meningitis and sepsis, with five serogroups (A, B, C, Y, and W-135) responsible for the majority of the disease. Multivalent (A, C, Y, and W-135) polysaccharide and conjugate vaccines have been licensed in the United States and elsewhere and are widely available. We have developed a multi-plexed electrochemiluminescent assay to quantitate serum antibody responses to meningococcal polysaccharides A, C, Y, and W-135 to allow for rapid evaluation of li- censed and investigational vaccines. A 96-well plate containing a carbon electrode arrayed with polysaccharides A, C, Y, and W-135 on separate spots within each well has been developed for simultaneous detection of polysaccharidespecific antibodies in serum samples from vaccinated individuals. The assay conditions were optimized using the anti-meningococcal serogroup A/C reference serum pool, CDC 1992 (NIBSC 99/706), through evaluation of plate types, coating polysaccharide concentrations, and blocking and serum diluent buffers. Comparison of single and multiplex assays demonstrated the sensitivity, specificity, and speed of the multi-plex format for the quantification of serum antibody responses to N. meningitidis polysaccharides A, C, Y and W-135.

Validation of Automated Dilution and Transfer Methods for Rapid Assessment of Functional Immune Responses to Meningococcal Vaccines

A high-throughput Serum Bactericidal Assay (SBA) was developed to monitor the functional antibody responses to capsular polysaccharide antigens from multiple serotypes of Neisseria meningitidis. This assay measures the ability of an antibody, aided by complement, to mediate killing of bacteria. Functional assays of this type are increasingly used in the quality control arena as potency release tests. Consequently, there has been an enhanced requirement for reproducibility in the performance of this type of assay. Assay validation is, therefore, important to facilitate understanding of the significance of values obtained, and to enable appropriate and informed use of the assay. This study involved the evaluation of the high-throughput serum bactericidal assay including the effects of different dilution and transfer techniques on assay performance. The results presented here demonstrate the repeatability, and the precision and ruggedness of the assay.

Vegetables, Herbs and Spices: The Importance of Family and Tasting

Objective: To explore why and how consumers utilize vegetables, herbs and spices. Methods: Focus groups were conducted with adult participants (n = 54) to determine attitudes, self-efficacy, and behaviors related to liking and usage of vegetables, herbs and spices as well as strategies to increase vegetable intake. Results: Seasonal availability, and family-centered memories and practices were motivation for vegetable intake. A diverse number of vegetables (n = 39) were listed as favorites based on taste and versatility, but seasonings were not commonly included in responses regarding preparation. Participants felt that vegetable dish names should reflect ingredients explicitly;that seasonings enhanced vegetable flavors;and that salt and pepper were most commonly used seasonings. The preferred strategy to increase vegetable consumption was through tasting or sampling opportunities;seasoning use was not mentioned. However, participants perceived that suggestions or demonstrations on how to prepare vegetables at home would be helpful. Personal gardens were mentioned frequently, but were not connected to strategies for increasing intake. Conclusions and Implications: Vegetable intake has a family focus that should be emphasized to increase intake. Efforts to increase intake could emphasize flavor and versatile preparation methods through tasting opportunities or educational demonstrations.
