BACKGROUND The influence of Helicobacter-pylori(***)infection and the characteristics of gastric cancer(GC)on tumor-infiltrating lymphocyte(TIL)levels has not been extensively *** of infiltrating-immune-cell subtypes as well as survival is necessary to obtain comprehensive *** To determine the rates of deficient mismatch-repair(dMMR),HER2-status and *** infection and their association with TIL levels in *** Samples from 503 resected GC tumors were included and TIL levels were evaluated following the international-TILs-working-group recommendations with assessment of the intratumoral(IT),stromal(ST)and invasive-border(IB)*** density of CD3,CD8 and CD163 immune cells,and dMMR and HER2-status were determined by immunohistochemistry(IHC).*** infection was evaluated by routine histology and quantitative PCR(qPCR)in a subset of *** dMMR was found in 34.4%,HER2+in 5%and ***-positive in 55.7%of *** IT-TIL was associated with grade-3(P=0.038),while ST-TIL with grade-1(P<0.001),intestinal-histology(P<0.001)and no-recurrence(P=0.003).dMMR was associated with high TIL levels in the ST(P=0.019)and IB(P=0.01)compartments,and STCD3(P=0.049)and ST-CD8(P=0.05)***2-was associated with high IT-CD8(P=0.009).*** was associated with high IT-TIL levels(P=0.009)when assessed by routine-histology,and with high TIL levels in the 3 compartments(P=0.002-0.047)and CD8 density in the IT and ST compartments(P=0.001)when assessed by qPCR.A longer overall survival was associated with low IT-CD163(P=0.003)and CD8/CD3(P=0.001 in IT and P=0.002 in ST)and high IT-CD3(P=0.021),ST-CD3(P=0.003)and CD3/CD163(P=0.002).CONCLUSION TIL levels were related to dMMR and *** CD8/CD3 and high CD163/CD3 were associated with lower recurrence and longer survival.

Fibrous dysplasia(FD)is a rare,disabling skeletal disease for which there are no established *** evidence supports inhibiting the osteoclastogenic factor receptor activator of nuclear kappa-B ligand(RANKL)as a potential treatment *** this study,we investigated the mechanisms underlying RANKL inhibition in FD tissue and its likely indirect effects on osteoprogenitors by evaluating human FD tissue pre-and post-treatment in a phase 2 clinical trial of denosumab(NCT03571191)and in murine in vivo and ex vivo preclinical *** analysis of human and mouse tissue demonstrated increased osteogenic maturation,reduced cellularity,and reduced expression of the pathogenic Gαs variant in FD lesions after RANKL *** sequencing of human and mouse tissue supported these *** interaction between osteoclasts and mutant osteoprogenitors was further assessed in an ex vivo lesion model,which indicated that the proliferation of abnormal FD osteoprogenitors was dependent on *** results from this study demonstrated that,in addition to its expected antiosteoclastic effect,denosumab reduces FD lesion activity by decreasing FD cell proliferation and increasing osteogenic maturation,leading to increased bone formation within *** findings highlight the unappreciated role of cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of FD pathology and demonstrate a novel mechanism of action of denosumab in the treatment of bone disease.

在光电子学应用中,器件性能主要取决于半导体纳米材料中的光生载流子动力学过程.但是,受反应速率、材料表面积、材料组成等多种因素影响,描述其中的动力学过程非常具有挑战性.模拟光生载流子动力学过程可以通过绝热分子动力学方法实现,即求解包含非绝热耦合项的含时薛定谔方程.在众多绝热分子动力学方法中,面跳跃方法出色地平衡了计算精度和计算成本,因而成为描述半导体纳米材料中不同非绝热过程间竞争的有力工具,已被用来模拟材料中的超快动力学过程和其他复杂效应,如Janus过渡金属二硫族化合物范德华异质结中的电荷分离.本综述通过介绍该领域代表性的理论及实验工作,阐述了光生载流子对半导体纳米材料性能的重要影响,以及面跳跃方法在描述其动力学行为中的重要作用.山于日趋复杂的材料体系对理论工作提出了巨大的挑战,本综述重点介绍了最近用于模拟这些复杂材料的一些创性的新方法,包括高精度的电子结构方法和与之相结合的绝热分子动力学方法.

How reliable is the ReaxFF potential for describing the structure of alkanethiols on gold?A molecular dynamics study

The structures of self-assembled monolayers(SAMs)of short(methyl)and long(hexyl)chain alkyl thiols on the clean gold(111)surface were modeled using for the Au–S interactions either the reactive ReaxFF potential or the well-known nonreactive Morse potential,while for the Au–Au interactions either the ReaxFF potential or an embeddedatom method(EAM).Analysis of the molecular dynamics(md)trajectories of possible SAM structures suggests that disordering of interfacial Au atoms is definitely driven by the gold–sulphur *** md results reveal a novel structure where two methanethiol molecules are bound to a gold adatom that has been lifted from the surface at 300 K,and the same kind of RS–Au–SR motif was also observed for hexanethiol at 600 K but not at 300 *** is more,the above motif is only observed for the reactive ReaxFF ***,these results are in clear agreement with recent experiments and more costly first principles-based md *** findings strongly support the use of reactive potentials such as ReaxFF for gathering an accurate description of Au–S interactions in inexpensive classical md simulations.

Erythropoietin modulates bone marrow stromal cell differentiation

Erythropoietin is essential for bone marrow erythropoiesis and erythropoietin receptor on non-erythroid cells including bone marrow stromal cells suggests systemic effects of erythropoietin. Tg6 mice with chronic erythropoietin overexpression have a high hematocrit, reduced trabecular and cortical bone and bone marrow adipocytes, and decreased bone morphogenic protein 2 driven ectopic bone and adipocyte formation. Erythropoietin treatment(1 200 IU·kg–1) for 10 days similarly exhibit increased hematocrit,reduced bone and bone marrow adipocytes without increased osteoclasts, and reduced bone morphogenic protein signaling in the bone marrow. Interestingly, endogenous erythropoietin is required for normal differentiation of bone marrow stromal cells to osteoblasts and bone marrow adipocytes.ΔEpoR_E mice with erythroid restricted erythropoietin receptor exhibit reduced trabecular bone, increased bone marrow adipocytes, and decreased bone morphogenic protein 2 ectopic bone formation. Erythropoietin treated ΔEpoR_E mice achieved hematocrit similar to wild-type mice without reduced bone, suggesting that bone reduction with erythropoietin treatment is associated with non-erythropoietic erythropoietin response. Bone marrow stromal cells from wild-type,Tg6, and ΔEpoR_E-mice were transplanted into immunodeficient mice to assess development into a bone/marrow organ. Like endogenous bone formation, Tg6 bone marrow cells exhibited reduced differentiation to bone and adipocytes indicating that high erythropoietin inhibits osteogenesis and adipogenesis, while ΔEpoR_E bone marrow cells formed ectopic bones with reduced trabecular regions and increased adipocytes, indicating that loss of erythropoietin signaling favors adipogenesis at the expense of osteogenesis. In summary, endogenous erythropoietin signaling regulates bone marrow stromal cell fate and aberrant erythropoietin levels result in their impaired differentiation.

Hyperthermic intraperitoneal chemotherapy:Rationale and technique
