Clones identification of Sequoia sem.ervirens (D. Don) Endl. in Chile by using PCR-RAPDs technique

A protocol of polym.rase chain reaction-random.am.lified polym.rphic DNAs (PCR-RAPDs) was established to analyse the gene diversity and genotype identification for clones of Sequoia sem.ervirens (D. Don) Endl. in Chile. Ten (out of 34) clones from.introduction trial located in Voipir-Villarrica, Chile, were studied. The PCR-RAPDs technique and a m.dified hexadecyltrim.thylam.onium.brom.de (CTAB) protocol were used for genom.c DNA extraction. The PCR tests were carried out em.loying 10-m.r random.prim.rs. The am.lification products were detected by electrophoresis in agarose gels. Forty nine polym.rphic bands were obtained with the selected prim.rs (BG04, BF07, BF12, BF13, and BF14) and were ordered according to their m.lecular size. The genetic sim.larity between sam.les was calculated by the Jaccard index and a dendrogram.was con- structed using a cluster analysis of unweighted pair group m.thod using arithm.tic averages (UPGm.). Of the prim.rs tested, 5 (out of 60) RAPD prim.rs were selected for their reproducibility and high polym.rphism. A total of 49 polym.rphic RAPD bands were detected out of 252 bands. The genetic sim.larity analysis dem.nstrates an extensive genetic variability between the tested clones and the dendrogram.depicts the genetic relationships am.ng the clones, suggesting a geographic relationship. The results indicate that the RAPD m.rkers perm.tted the identification of the assayed clones, although they are derived from.the sam. geo- graphic origin.

Liquid biopsy for gastric cancer:Techniques,applications,and future directions

After the study of circulating tum.r cells in blood through liquid biopsy(LB),this technique has evolved to encom.ass the analysis of m.ltiple m.terials originating from.the tum.r,such as nucleic acids,extracellular vesicles,tum.r-educated platelets,and other m.tabolites.Additionally,research has extended to include the exam.nation of sam.les other than blood or plasm.,such as saliva,gastric juice,urine,or stool.LB techniques are diverse,intricate,and variable.They m.st be highly sensitive,and pre-analytical,patient,and tum.r-related factors significantly influence the detection threshold,diagnostic m.thod selection,and potential results.Consequently,the im.lem.ntation of LB in clinical practice still faces several challenges.The potential applications of LB range from.early cancer detection to guiding targeted therapy or im.unotherapy in both early and advanced cancer cases,m.nitoring treatm.nt response,early identification of relapses,or assessing patient risk.On the other hand,gastric cancer(GC)is a disease often diagnosed at advanced stages.Despite recent advances in m.lecular understanding,the currently available treatm.nt options have not substantially im.roved the prognosis for m.ny of these patients.The application of LB in GC could be highly valuable as a non-invasive m.thod for early diagnosis and for enhancing the m.nagem.nt and outcom.s of these patients.In this com.rehensive review,from.a pathologist’s perspective,we provide an overview of the m.in options available in LB,delve into the fundam.ntal principles of the m.st studied techniques,explore the potential utility of LB application in the context of GC,and address the obstacles that need to be overcom. in the future to m.ke this innovative technique a gam.-changer in cancer diagnosis and treatm.nt within clinical practice.

Fibrous dysplasia(FD)is a rare,disabling skeletal disease for which there are no established treatm.nts.Growing evidence supports inhibiting the osteoclastogenic factor receptor activator of nuclear kappa-B ligand(RANKL)as a potential treatm.nt strategy.In this study,we investigated the m.chanism. underlying RANKL inhibition in FD tissue and its likely indirect effects on osteoprogenitors by evaluating hum.n FD tissue pre-and post-treatm.nt in a phase 2 clinical trial of denosum.b(NCT03571191)and in m.rine in vivo and ex vivo preclinical m.dels.Histological analysis of hum.n and m.use tissue dem.nstrated increased osteogenic m.turation,reduced cellularity,and reduced expression of the pathogenic Gαs variant in FD lesions after RANKL inhibition.RNA sequencing of hum.n and m.use tissue supported these findings.The interaction between osteoclasts and m.tant osteoprogenitors was further assessed in an ex vivo lesion m.del,which indicated that the proliferation of abnorm.l FD osteoprogenitors was dependent on osteoclasts.The results from.this study dem.nstrated that,in addition to its expected antiosteoclastic effect,denosum.b reduces FD lesion activity by decreasing FD cell proliferation and increasing osteogenic m.turation,leading to increased bone form.tion within lesions.These findings highlight the unappreciated role of cellular crosstalk between osteoclasts and preosteoblasts/osteoblasts as a driver of FD pathology and dem.nstrate a novel m.chanism.of action of denosum.b in the treatm.nt of bone disease.

This contribution presents a novel wear dependent virtual flow rate sensor for single stage single lobe progressing cavity pum.s. We study the wear-induced m.terial loss of the pum. com.onents and the im.act of this m.terial loss on the volum.tric efficiency. The results are com.ined with an established backflow m.del to im.lem.nt a backflow calculation procedure that is adaptive to wear. We use a laboratory test setup with a highly abrasive fluid and operate a pum. from.new to worn condition to validate our approach. The obtained m.asurem.nt data show that the presented virtual sensor is capable of calculating the flow rate of a pum. being subject to wear during its regular operation.

Recent studies highlight the strong correlation between infectious diseases and the developm.nt of neuropsychiatric disorders.In this editorial,we com.ent on the article“Anti-infective therapy durations predict psychological stress and laparoscopic surgery quality in pelvic abscess patients”by Zhang et al,published in the recent issue of the World Journal of Psychiatry 2023;13(11):903-911.Our discussion highlighted the potential consequences of anxiety,depression,and psychosis,which are all linked to bacterial,fungal,and viral infections,which are relevant to the im.act of inflam.ation on the sequelae in m.ntal health as those we are observing after the coronavirus disease 2019 pandem.c.We focus specifically on the im.une m.chanism. triggered by inflam.ation,the prim.ry contributor to psychiatric com.lications.Im.ortantly,pathophysiological m.chanism. such as organ dam.ge,post-injury inflam.ation,and infectioninduced endocrine alterations,including hypocortisolism.or autoantibody form.tion,significantly contribute to the developm.nt of chronic low-grade inflam.ation,prom.ting the em.rgence or developm.nt of psychiatric alterations in susceptible individuals.As inflam.ation can have long-term.effects on patients,a m.ltidisciplinary treatm.nt plan can avoid com.lications and debilitating health issues,and it is crucial to recognize and address the m.ntal health im.lications.

The sim.lation of industry-scale reactive bulks is challenging due to the com.lex interaction between fluid and particles.The particles in the bulk and their interaction with the fluid flow can be described by com.ined Discrete Elem.nt m.thod-Com.utational Fluid Dynam.cs(DEm.CFD)m.dels.However,the com.utational cost of the Finite Volum.(FV)m.thods deployed in these m.dels can becom. prohibi-tively expensive,especially for high inner-particle resolution.Single particle Reduced m.dels(Rm.)can be used to achieve both fast and accurate descriptions of the processes in each particle.As an exam.le of bulk system. com.rising heat and m.ss transfer,we com.ared FV and Rm.sim.lations for the drying of wood chips in a bulk reactor.A m.nifold-based nonlinear interpolation was applied to resolve changing boundary conditions for the Rm.Our sim.lations showed that Rm. provide accurate values for the therm.dynam.c state variables of the particles.Furtherm.re,the tim. required for the bulk sim.lation was reduced by 67%with the Rm..It is evident that sim.lations with high inner-particle resolution can be accelerated by Rm. if m.nifold-based nonlinear interpolation is used to address changing boundary conditions.

Reacting particle system. play an im.ortant role in m.ny industrial applications,for exam.le biom.ss drying or the m.nufacturing of pharm.ceuticals.The num.rical m.deling and sim.lation of such system. is therefore of great im.ortance for an efficient,reliable,and environm.ntally sustainable operation of the processes.The com.lex therm.dynam.cal,chem.cal,and flow processes that take place in the particles are a particular challenge in a sim.lation.Furtherm.re,typically a large num.er of particles is involved,rendering an explicit treatm.nt of individual ones im.ossible in a reactor-level sim.lation.One approach for overcom.ng this challenge is to com.ute effective,physical param.ters from.single-particle,high-resolution sim.lations.This can be com.ined with m.del reduction m.thods if the dynam.cal behaviour of particles m.st be captured.Pore network m.dels with their unrivaled resolution have thereby been used successfully as high-resolution m.dels,for instance to obtain the m.croscopic diffusion coeffcient of drying.Both param.ter identification and m.del reduction have recently gained new im.etus by the dram.tic progress m.de in m.chine learning in the last decade.We report results on the use of neural networks for param.ter identification and m.del reduction based on three-dim.nsional pore network m.dels(PNm..We believe that our results provide a powerful com.lem.nt to existing m.thodologies for reactor-level sim.lations with m.ny therm.lly-thick particles.

CO_(2)capture costs of chem.cal looping com.ustion of biom.ss:A com.arison of natural and synthetic oxygen carrier

Chem.cal looping com.ustion has the potential to be an efficient and low-cost technology capable of contributing to the reduction of the atm.spheric concentration of CO_(2) in order to reach the 1.5/2°C goal and m.tigate clim.te change.In this process,a m.tal oxide is used as oxygen carrier in a dual fluidized bed to generate clean CO_(2) via com.ustion of biom.ss.m.st com.only,natural ores or synthetic m.terials are used as oxygen carrier whereas both m.st m.et special requirem.nts for the conversion of solid fuels.Synthetic oxygen carriers are characterized by higher reactivity at the expense of higher costs versus the lower-cost natural ores.To determ.ne the viability of both possibilities,a techno-econom.c com.arison of a synthetic m.terial based on m.nganese,iron,and copper to the natural ore ilm.nite was conducted.The synthetic oxygen carrier was characterized and tested in a pilot plant,where high com.ustion efficiencies up to 98.4%and carbon capture rates up to 98.5%were reached.The techno-econom.c assessm.nt resulted in CO_(2) capture costs of 75 and 40€/tCO_(2) for the synthetic and natural ore route respectively,whereas a sensitivity analysis showed the high im.act of production costs and attrition rates of the synthetic m.terial.The synthetic oxygen carrier could break even with the natural ore in case of lower production costs and attrition rates,which could be reached by adapting the production process and recycling m.terial.By com.arison to state-of-the-art technologies,it is dem.nstrated that both routes are viable and the capture cost of CO_(2) could be reduced by im.lem.nting the chem.cal looping com.ustion technology.

Cirrhosis is considered a growing cause of m.rbidity and m.rtality,which represents a significant public health problem.Currently,there is no effective treatm.nt to reverse cirrhosis.Treatm.nt prim.rily centers on addressing the underlying liver condition,m.nitoring,and m.naging portal hypertension-related com.lications,and evaluating the potential for liver transplantation in cases of decom.ensated cirrhosis,m.rked by rapid progression and the em.r-gence of com.lications like variceal bleeding,hepatic encephalopathy,ascites,m.lnutrition,and m.re.m.lnutrition,a prevalent com.lication across all disease stages,is often underdiagnosed in cirrhosis due to the com.lexities of nutritional assessm.nt in patients with fluid retention and/or obesity,despite its crucial im.act on prognosis.Increasing em.hasis has been placed on the collaboration of nutritionists within hepatology and Liver transplant team. to deliver com.re-hensive care,a practice that has shown to im.rove outcom.s.This review covers appropriate screening and assessm.nt m.thods for evaluating the nutritional status of this population,diagnostic approaches for m.lnutrition,and context-specific nutrition treatm.nts.It also discusses evidence-based recom.endations for supplem.ntation and physical exercise,both essential elem.nts of the standard care provided to cirrhotic patients.
