AMP-activated kinase in human spermatozoa： identification, intracellular localization, and key function in the reRulation of sperm motility

AMP-activated kinase （AMPK）, a protein that regulates energy balance and metabolism, has recently been identified in boar spermatozoa where regulates key functional sperm processes essential for fertilization. This work＇s aims are AMPK identification, intracellular localization, and their role in human spermatozoa function. Semen was obtained from healthy human donors. Sperm AMPK and phospho-Thr172-AMPK were analyzed by Western blotting and indirect immunofluorescence. High- and low-quality sperm populations were separated by a 40%-80% density gradient. Human spermatozoa motility was evaluated by an Integrated Semen Analysis System （ISAS） in the presence or absence of the AMPK inhibitor compound C （CC）. AMPK is localized along the human spermatozoa, at the entire acrosome, midpiece and tail with variable intensity, whereas its active form, phospho-Thr172-AMPK, shows a prominent staining at the acrosome and sperm tail with a weaker staining in the midpiece and the postacrosomal region. Interestingly, spermatozoa bearing an excess residual cytoplasm show strong AMPK staining in this subcellular compartment. Both AMPK and phospho-Thr172-AMPK human spermatozoa contents exhibit important individual variations. Moreover, active AMPK is predominant in the high motility sperm population, where shows a stronger intensity compared with the low motility sperm population. Inhibition of AMPK activity in human spermatozoa by CC treatment leads to a significant reduction in any sperm motility parameter analyzed： percent of motile sperm, sperm velocities, progressivity, and other motility coefficients. This work identifies and points out AMPK as a new molecular mechanism involved in human spermatozoa motility. Further AMPK implications in the clinical efficiency of assisted reproduction and in other reproductive areas need to be studied.

The international scientific literature presents still incipient results regarding the management of cancer symptom clusters by oncology nursing,especially in pediatric *** is a promising field of investigation for clinical nurses and researchers,and when it is subsidized by medium-range theories,they co-rroborate the diagnoses and interventions of nursing in oncology,enhancing the science of nursing *** minireview article aims to discuss the utilizing the hospital clowns as a complementary therapy,to enhance quality of life and reduce stress and fatigue in pediatric cancer ***,the evidence presented so far pointed out that complementary therapy might help improve the quality of life of pediatric cancer patients,and that complementary therapy usage should be part of a health comprehensive care model,delivering therapeutic approaches that might enhance the mind-body during a pediatric cancer patients’life *** results of scientific investigations by nurses,particularly those linked to the basic sciences,play a critical role in advancing personalized care in pediatric integrative oncology.

Contrasting climate drivers of seasonal growth in western vs. eastern Mexican mountain conifer forests

Hydroclimate affects the radial growth responses of trees, but the drivers of their spatial and population variability are not sufficiently understood. We addressed this issue by sampling several conifer populations located at the same latitude, but at different longitude and elevation in western(W) and eastern(E) Mexican regions. We used dendroecology to disentangle how earlywood width(EW), latewood width(LW) and adjusted LW(LWadj),i.e. the residuals after removing EW influences on LW, responded to climate variables(temperature and precipitation), climate indices(Southern Oscillation Index, SOI, Nino 3.4, Pacific Decadal Oscillation, PDO) and a drought index(Standardised Precipitation-Evapotranspiration Index, SPEI). The W species(Pinus herrerae Martinez, Pinus durangensis Martinez, Abies durangensis Martínez and Cupressus lusitanica Mill.) showed lower growth rates than the E species(Pinus hartwegii Lindl., Picea mexicana Martinez, Pseudotsuga menziesii(Mirb.) Franco and Abies vejari Martinez). Growth in W benefits mostly from high precipitation in the prior winter and current spring and it is limited by high temperatures in spring, whereas growth in the E showed similar but weaker ***, positive(negative) correlations were found in radial growth with the Nino 3.4(SOI) and the PDO from the prior to current autumns, which were again stronger in absolute terms in the W than in the E regions,excepting SOI in summer. In the W, P. durangensis and C. lusitanica were the least and most responsive species to spring drought, respectively;whilst P. menziesii and A. vejari were very responsive to spring drought compared to P. hartwegii in the E. Our results suggest greater responsiveness to hydroclimate and atmospheric patterns in the W than in the E region. The renin-angiotensin system(RAS)was classically considered a circulating hormonal system that regulates blood ***,different tissues and organs,including the brain,have a local paracrine *** regulation between the dopaminergic system and RAS has been observed in several *** of these interactions leads to renal and cardiovascular diseases,as well as progression of dopaminergic neuron degeneration in a major brain center of dopamine/angiotensin interaction such as the nigrostriatal system.A decrease in the dopaminergic function induces upregulation of the angiotensin type-1(AT1)receptor activity,leading to recovery of dopamine ***,AT1 receptor overactivity in dopaminergic neurons and microglial cells upregulates the cellular NADPH-oxidase-superoxide axis and Ca^(2+)release,which mediate several key events in oxidative stress,neuroinflammation,andα-synuclein aggregation,involved in Parkinson’s disease(PD)*** intraneuronal antioxidative/anti-inflammatory RAS counteracts the effects of the pro-oxidative AT1 receptor *** with this,an imbalance in RAS activity towards the pro-oxidative/pro-inflammatory AT1 receptor axis has been observed in the substantia nigra and striatum of several animal models of high vulnerability to dopaminergic ***,autoantibodies against angiotensin-converting enzyme 2 and AT1 receptors are increased in PD models and PD patients and contribute to blood-brain barrier(BBB)dysregulation and nigrostriatal pro-inflammatory RAS *** strategies addressed to the modulation of brain RAS,by AT1 receptor blockers(ARBs)and/or activation of the antioxidative axis(AT2,Mas receptors),may be neuroprotective for individuals with a high risk of developing PD or in prodromal stages of PD to reduce progression of the disease.

Risk factors and classifications of hilar cholangiocarcinoma

Cholangiocarcinoma is the second most common primary malignant tumor of the *** cholangiocarcinoma or Klatskin tumor represents more than 50% of all biliary tract cholangiocarcinomas.A wide range of risk factors have been identified among patients with Perihilar cholangiocarcinoma including advanced age,male gender,primary sclerosing cholangitis,choledochal cysts,cholelithiasis,cholecystitis,parasitic infection(Opisthorchis viverrini and Clonorchis sinensis),inflammatory bowel disease,alcoholic cirrhosis,nonalcoholic cirrhosis,chronic pancreatitis and metabolic *** classifications have been used to describe the pathologic and radiologic appearance of *** three systems most commonly used to evaluate Perihilar cholangiocarcinoma are the Bismuth-Corlette(BC) system,the Memorial Sloan-Kettering Cancer Center and the TNM *** BC classification provides preoperative assessment of local *** Memorial Sloan-Kettering cancer center proposes a staging system according to three factors related to local tumor extent:the location and extent of bile duct involvement,the presence or absence of portal venous invasion,and the presence or absence of hepatic lobar *** TNM classification,besides the usual descriptors,tumor,node and metastases,provides additional information concerning the possibility for the residual tumor(R) and the histological grade(G).Recently,in 2011,a new consensus classification for the Perihilar cholangiocarcinoma had been *** consensus was organised by the European Hepato-PancreatoBiliary Association which identified the need for a new staging system for this type of *** classification includes information concerning biliary or vascular(portal or arterial) involvement,lymph node status or metastases,but also other essential aspects related to the surgical risk,such as remnant hepatic volume or the possibility of underlying disease.

Effects of a 3-month vigorous physical activity intervention on eating behaviors and body composition in overweight and obese boys and girls

Purpose: This study aims to analyze the effects of a 3-month vigorous physical activity(VPA) intervention on eating behavior and body composition in overweight and obese children and ***: Forty-seven participants(7–16 years) took part in the study: 28 were assigned to the intervention group(IG)(10 boys and 18 girls) and19 in a control group(CG)(8 boys and 11 girls). Body composition(dual-energy X-ray absorptiometry), anthropometrics(body mass, height, and body mass index(BMI)), and eating behavior traits(Three-Factor Eating Questionnaire-R21 C) were determined before and after the VPA ***: A decrease in the percentage of body fat and BMI(-2.8% and-1.8%, respectively), and an increase in most lean mass variables were found in the IG(all p ≤ 0.05). In relation to the eating behavior traits, IG subjects showed a 14% reduction in the Emotional Eating score(p = 0.04),while Cognitive Restraint score did not change after the VPA intervention. The baseline factors of the questionnaire predicted changes in body mass and fat mass variables only in the ***: A 3-month VPA intervention influenced eating behaviors of overweight or obese young, especially the Emotional Eating factor, in the presence of favorable body composition changes.

Background:Among urban stimuli,anthropogenic noise has been identified to be one of the behavioral drivers of species that rely on acoustic signals for *** have shown both species‑specific and assemblage responses to urban noise,ranging from the modulation of their acoustic frequencies and spatiotemporal adjustments to declines in species *** this study,we assessed the citywide relationship between two anthropogenic noise variables(noise levels recorded during bird surveys and daily average noise levels)and vegetation cover with bird species ***:This study was conducted in the city of Xalapa(Mexico)through a 114 citywide point‑count *** recorded bird communities at each sampling *** measured noise levels using a sound level meter while performing point‑***,we generated a map of average daily noise of the city using an array of 61 autonomous recording units distributed across the city of Xalapa and calculated daily noise levels for the 114 *** ran a linear model(LM)to assess potential relationships between both point‑count and daily(24 h)noise values and vegetation cover with bird ***:Results from the LM show:(1)a negative relationship between maximum point‑count noise and avian species richness,(2)no relationship between 24 h noise and bird species richness,and(3)a positive relationship between vegetation cover and bird species ***:Results provide evidence that decreases in urban bird species richness do not necessarily imply the permanent absence of species,suggesting that birds can temporarily fly away from or avoid sites when noisy,become cryptic while noisy events are occurring,or be undetected due to our inability to record them in the field during noisy events.

Cetuximab （CTX） is a potent chimeric mouse/human monoclonal antibody （mAb） approved worldwide for treatment of metastatic colorectal cancer. Among the various biological and physical analyses per- formed for full study on this biopharmaceutic, the determination of the concentration preparations throughout manufacturing and subsequent handling in hospital is particularly relevant. In the present work, the study and validation of a method for quantifying intact CTX by reverse-phase high-perfor- mance liquid chromatography with diode array detection （（RP）HPLC/DAD） is presented. With that end, we checked the performance of a chromatographic method for quantifying CTX and conducted a study to validate the method as stability-indicating in accordance with the International Conference on Harmo- nization guidelines （ICH） for biotechnological drugs; therefore, we evaluated linearity, accuracy, preci- sion, detection and quantification limits, robustness and system suitability. The specificity of the method and the robustness of the mAb formulation against external stress factors were estimated by compre- hensive chromatographic analysis by subjecting CTX to several informative stress conditions. As de- monstrated, the method is rapid, accurate, and reproducible for CTX quantification. It was also suc- cessfully used to quantify CTX in a long-term stability study performed under hospital conditions.
