Nf.κB Controls Resistance of.Human Salivary Gland (HSG) Cells to Apoptosis in an <i>in Vitro</i>Model of.Sjogren’s Syndrome

Aim: To elucidate the anti-apoptotic properties of.nuclear f.ctor kappa light-chain-enhancer of.activated B cells (Nf.κB) and f.edback regulation of.Nf.κB by nuclear f.ctor of.kappa light-chain-enhancer of.activated B-cells inhibitor alpha (IκBα). Methods: We developed an in vitro model of.Sjogren’s syndrome by transf.cting human salivary gland (HSG) and acinar cells (NS-SV-AC) with a plasmid-encoding IκBαM (pCMV-IκBαM), a degradation-resistant IκBα (nuclear f.ctor of.kappa light polypeptide gene enhancer in B-cells inhibitor, alpha)-mutant, and examined TNf.induced apoptosis and anti-apoptotic properties of.Nf.κB. Apoptosis and induction of.pro-apoptotic and anti-apoptotic genes were investigated by cDNA arrays, RT-PCR, electrophoretic mobility shif. assays, and western blot. Results: In the presence of.Nf.κB inhibitors, TNf.induced apoptosis was markedly increased in both salivary gland and acinar cells. Increased caspase-3 activity was present in both HSG and NS-SV-AC cells. IκBαM-transf.cted salivary gland cells were more sensitive to TNf.induced apoptosis than IκBαM-transf.cted acinar cells. Transcription of.pro-apoptotic genes was conf.rmed in both HSG and NS-SV-AC cells that were transf.cted with IκBαM. Results f.om caspase-3 activity assay conf.rmed previous experiments showing an apoptotic role f.r Nf.κB. Conclusion: Data f.om gene expression arrays suggest that dif.erent mechanisms may operate during TNf.induced apoptosis in salivary gland ductal and acinar cells.

Optimal Design of.Hybrid Microgrid in Isolated Communities of.Ecuador

In rural territories, the communities use energy sources based on f.ssil f.els to supply themselves with electricity, which may address two main problems: greenhouse gas emissions and high f.el prices. Hence, there is an opportunity to include renewable resources in the energy mix. This paper develops an optimization model to determine the optimal sizing, the total annual investment cost in renewable generation, and other operating costs of.the components of.a hybrid microgrid. By running a k-means clustering algorithm on a meteorological dataset of.the community under study, the hourly representative values become input parameters in the proposed optimization model. The method f.r the optimal design of.hybrid microgrid is analyzed in six operating scenarios considering:(1) 24-hour continuous power supply;(2) load shedding percentage;(3) diesel power generator(genset) curtailment;(4) the worst meteorological conditions;(5) the use of.renewable energy sources including battery energy storage systems(BESSs);and(6) the use of.genset. A mathematical programming language(AMPL) tool is used to f.nd solutions of.the proposed optimization model. Results show that the total costs of.microgrid in the scenarios that cover 100% of.the load demand(without considering the scenario with 100% renewables) increase by over 16% compared with the scenario with genset operation limitation. f.r the designs with power supply restrictions, the total cost of.microgrid in the scenario with load shedding is reduced by over 27% compared with that without load shedding.

Use of.two-surf.ctants mixtures to attain specif.c HLB values f.r assisted TPH-diesel biodegradation

In a surf.ctant assisted biodegradation process, the choice of.surf.ctant(s) is of.crucial importance. The question is: does the type of.surf.ctant(i.e. chemical f.mily) af.ect the biodegradation process at f.xed hidrophillic-lypof.llic balance(HLB) values? Microcosm assessments were developed using contaminated soil, with around of.5000 mg/kg of.hydrocarbons as TPH-diesel. Mixtures of.three nonionic surf.ctants were employed to get a wide range of.specif.c HLB values. Tween20 and Span20 were mixed in the appropriate proportions to get HLB values between 8.6 and 16.7. Tween/Span60 mixtures reached HLB values between 4.7 and 14.9. f.nally, Tween/Span80 combinations yielded HLB values between 4.3 and 15. TPH-diesel biodegradation was measured at the beginning, and af.er 8 weeks, as well as the f.U/gr soil, as a measure of.microorganisms′ development during the biodegradation period. A second aim of.this work was to assess the use of.guar gum as a biodegradation enhancer instead of.synthetic products. The conclusions of.this work are that surf.ctant chemical f.mily, and not only the HLB value clearly af.ects the assisted biodegradation rate. Surf.ctant's synergism was clearly observed. Regarding the use of.guar gum, no biodegradation enhancement was observed f.r the three assessed concentrations, i.e., 2, 20, and 200 mg/kg, respectively. On the contrary, TPH-diesel removal was lower as the gum concentration increased. It is quite possible that guar gum was used as a microbial substrate.

In simulations of.f.uidized beds using computational f.uid dynamics （Cf.）, the description of.gas-solid f.ow hydrodynamics relies on a drag model to account f.r the momentum transf.r between gas and solid phases. Although several studies of.drag models have been published, there have been f.w investigations of.the application of.lattice Boltzmann method （LBM）-based drag models to bubbling f.uidized bed simu- lations. In the present study, a comprehensive comparison of.empirical and LBM-based drag models was carried out to assess the perf.rmance of.these models during simulations of.gas-solid f.ow hydrodynam- ics in a bubbling f.uidized bed. A Cf. model using the Mf.X code based on the Eulerian-Eulerian approach and the kinetic theory of.granular f.ow was used to simulate a 2D bubbling f.uidized bed with Geldart B particles. The simulation results were validated by comparison with experimental data. Statistical anal- ysis of.the results shows that LBM-based drag models can reliably model gas-solid f.ow hydrodynamics in a bubbling f.uidized bed.

Currently,specif.c nutrient concentration,metabolizable energy(ME)and digestible amino acids are used as f.ed f.rmulation criteria.A balanced nutrient density(BND)concept based on 2 criteria of.nutrient density and balanced amino acids-to-ME ratio may of.er more f.exibility in optimisation of.prof.t in f.rmulation of.diets compared with current f.rmulation based on set values per unit of.f.ed mass.A total of.672 one-d-old of.-sex male Ross 308 broiler chickens were used across two 42-d perf.rmance trials in a 3×2 f.ctorial arrangement of.treatments with each diet replicated 8 times(14 birds per replicate).The experimental f.ctors were 2 nutrient density levels(low[LD]and high[HD])and 3 digestible lysine-to-ME ratios(DLYS:ME;low,medium,and high).Low density diets had ME of.2,876 and 3,023 kcal/kg f.r starter and f.nisher,respectively,while values f.r HD diets were 3,169 and 3,315 kcal/kg with proportionally higher non-nitrogenated *** digestibility and apparent metaboliz-able energy(AME)assays were conducted at d 21 and *** assays at d 7 were conducted on birds used f.r perf.rmance *** of.the diet density,birds f.d low DLYS:ME had a lower(Pf.ed intake(d 0 to 42)than medium and high DLYS:*** interaction,birds f.d low and medium DLYS:ME had a similar body weight gain being the heaviest while birds low DLYS:ME were the *** an interaction(Pf.R value was observed f.r birds f.d LD×low DLYS:ME and improved linearly when DLYS:ME increased to the highest level reaching a limit f.r birds f.d HD×medium DLYS:*** conversion linearly decreased(Pf.ected on d 21 and 42 but not on d 7 of.*** the independence of.response on BW and f.ed consumption,the use of.BND as a f.exible system in diet f.rmulations has the potential to enable more accurate f.rmulation f.r optimisation of.growth perf.rmance

Photocatalytic perf.rmance of.K2Ti6O13 whiskers to H2 evolution and CO2 photo-reduction

K2Ti6O13 whiskers were synthesized by conventional sol-gel method, sono-chemical assisted and microwave assisted sol-gel method in order to obtain catalysts with dif.erent particle sizes and to modif. their optical, textural and electrochemical properties. These modif.cations improved their photocatalytic activity f.r H2 evolution and CO2 photo-reduction. Long K2Ti6O13 whiskers prepared by ultrasound assisted sol-gel method are the most active photocatalysts f.r the hydrogen evolution reaction using pure water as reactant (U-SG, 10,065 μmol g^-1). In contrast, an opposite behavior was observed using a mixture of.ethanol-water, where the highest activity was achieved by the shortest and less crystalline K2Ti6O13 whiskers (C-SG, 3,2871 μmol g^-1). In case of.CO2 photo-reduction, long whiskers that were also prepared by the sono-chemical assisted sol-gel method were the most active to transf.rm CO2 to f.rmaldehyde, methane, methanol and hydrogen. The Ef. value of.this catalyst is located very close to the potential f.r f.rmaldehyde production and f.vors the selectivity to this organic product.

BACKGROUND Cholesterol is related to improvements in the rate of.sustained virological response and a robust immune response against the hepatitis C virus(HCV).APOE gene polymorphisms regulate cholesterol levels modif.ing the course of.the HCV *** relationship between cholesterol,APOE alleles,and the outcome of.HCV inf.ction has not been evaluated in the admixed population of.*** To investigate the role of.APOE-ε2,-ε3,and-ε4 alleles and the metabolic prof.le in the outcome of.HCV *** A total of.299 treatment-na?ve HCV patients were included in this retrospective *** were stratif.ed in chronic hepatitis C(CHC)(n=206)and spontaneous clearance(SC)(n=93).A clinical record was *** tests were assessed by dry chemistry *** genotypes were determined using a Real-Time polymerase chain reaction *** Total cholesterol,low-density lipoprotein cholesterol(LDL-c),triglycerides,and hypercholesterolemia were higher in SC than CHC patients as well as the f.equency of.the APOEε4 allele(12.4%vs 7.3%).SC patients were overweight(54.8%).Theε4 allele was associated with SC(OR=0.55,95%CI:0.31-0.98,P=0.042)and mild f.brosis(f.-f.)in CHC patients(OR 0.091,95%CI 0.01-0.75,P=0.020).LDL-c≥101.5 mg/dL(OR=0.20,95%CI:0.10-0.41,Pf.rred a protective ef.ect in the course of.the HCV inf.ction in the context of.excess body weight.

Extracting the light trapped in a waveguide,or the opposite ef.ect of.trapping light in a thin region and guiding it perpendicular to its incident propagation direction,is essential f.r optimal energetic perf.rmance in illumination,display or light harvesting *** we demonstrate that the paradoxical goal of.letting as much light in or out while maintaining the wave ef.ectively trapped can be achieved with a periodic array of.interpenetrated f.bers f.rming a photonic f.ber *** entering perpendicular to that plate may be trapped in an intermittent chaotic trajectory,leading to an optically ergodic *** f.bricated such a photonic f.ber plate and showed that f.r a solar cell incorporated on one of.the plate surf.ces,light absorption is greatly *** this,we f.und the unexpected result that a more chaotic photon trajectory reduces the production of.photon scattering entropy.
