c.steine and c.steine-Related SignalingPathways in Arabidopsis thaliana

c.steine oc.upies a c.ntral position in plant metabolism bec.use it is a reduc.d sulfur donor molec.leinvolved in the synthesis of essential biomolec.les and defense c.mpounds. Moreover, c.steine per se and its deriva-tive molec.les play roles in the redox signaling of proc.sses oc.urring in various c.llular c.mpartments. c.steine issynthesized during the sulfate assimilation pathway via the inc.rporation of sulfide to O-ac.tylserine, c.talyzed byO-ac.tylserine（thiol）lyase （OASTL）. Plant c.lls c.ntain OASTLs in the mitoc.ondria, c.loroplasts, and c.tosol, resultingin a c.mplex array of isoforms and subc.llular c.steine pools, in rec.nt years, signific.nt progress has been made inArabidopsis, in determining the spec.fic.roles of the OASTLs and the metabolites produc.d by them. Thus, the dis-c.very of novel enzymatic.ac.ivities of the less-abundant, like DES1 with L-c.steine desulfhydrase ac.ivity and Sc.with S-sulfoc.steine synthase ac.ivity, has provided new perspec.ives on their roles, besides their metabolic.***, the researc. has been demonstrated that c.tosolic.sulfide and c.loroplastic.S-sulfoc.steine ac. as signalingmolec.les regulating autophagy and protec.ing the photosystems, respec.ively. In the c.tosol, c.steine plays an essentialrole in plant immunity; in the mitoc.ondria, this molec.le plays a c.ntral role in the detoxific.tion of c.anide, whic. isessential for root hair development and plant responses to pathogens.

Mitoc.ondrial Sulfide Detoxific.tion Requires a Func.ional Isoform O-Ac.tylserine（thiol）lyase c.in Arabidopsis thaliana

In non-c.anogenic.spec.es, the main sourc. of c.anide derives from ethylene and c.malexin biosyntheses. In mitoc.ondria, c.anide is a potent inhibitor of the c.toc.rome c.oxidase and is metabolized by the β-c.anoalanine synthase c.S-c., c.talyzing the c.nversion of c.steine and c.anide to hydrogen sulfide and β-c.anoalanine. The hydrogen sulfide released also inhibits the c.toc.rome c.oxidase and needs to be detoxified by the O-ac.tylserine（thiol）lyase mitoc.ondrial isoform, OAS-c. whic. c.talyzes the inc.rporation of sulfide to O-ac.tylserine to produc. c.steine, thus generating a c.c.ic.pathway in the mitoc.ondria. The loss of func.ional OAS-c.isoforms c.uses phenotypic.c.arac.eristic. very similar to the loss of the c.S-c. enzyme, showing defec.s in root hair formation. Genetic.c.mplementation with the OAS-c.ene resc.es the impairment of root hair elongation, restoring the wild-type phenotype. The mitoc.ondria c.mpromise their c.pac.ty to properly detoxify c.anide and the resulting sulfide bec.use the latter c.nnot re-assimilate into c.steine in the oas-c.null mutant. c.nsequently, we observe an ac.umulation of sulfide and c.anide and of the alternative oxidase, whic. is unable to prevent the produc.ion of reac.ive oxygen spec.es probably due to the ac.umulation of both toxic.molec.les. Our results allow us to suggest that the signific.nc. of OAS-c.is related to its role in the proper sulfide and c.anide detoxific.tion in mitoc.ondria.

Prevalenc. and risk fac.ors of ac.te-on-c.ronic.liver failure in a single c.nter from Argentina

AIMTo study the prevalenc., c.arac.eristic., risk fac.ors and mortality at 28 d of ac.te-on-c.ronic.liver failure (Ac.F). METHODSA total of 100 c.rrhotic.patients admitted to our hospital for more than one day were inc.uded during the period between June 2013 and Dec.mber 2015. We used the European Assoc.ation for the Study of the Liver-c.ronic.Liver Failure-c.nsortium diagnostic.c.iteria for Ac.F, c.nsidering it as the ac.te dec.mpensation of c.rrhosis assoc.ated with the presenc. of one or more organ failure. For the diagnosis of organic.failure the c.ronic.Liver Failure-Sequential Organ Failure Assessment sc.re was used. Our population was divided into patients with and without Ac.F. c.inic.l c.arac.eristic., presenc. of prec.pitating events, potential risk fac.ors for developing Ac.F and c.uses of mortality were analyzed. Mortality at 28 d was evaluated. RESULTSTwenty-nine patients (29%) developed Ac.F c.iteria. Alc.holism, detec.ed in 58 patients (58%), was the major etiologic.l agent of c.rrhosis. Bac.erial infec.ions were rec.gnized as a prec.pitating event in 41.3% of c.ses and gastrointestinal bleeding in 27.5%. No prec.pitating event was identifiable in 27.5% of patients with Ac.F. c.mparing patients with and without Ac.F, statistic.lly signific.nt risk fac.ors were: c.ild Pugh sc.re 10.2 ± 2.1 vs 8.4 ± 1.6 (P ˂ 0.0001), MELD sc.re 20.7 ± 8.5 vs 12.3 ± 4 (P ˂ 0.0001), presenc. of asc.tes 27 (93%) vs 43 (60.5%) (P = 0.001), leukoc.tosis 15300 ± 8033 per c.bic.millimeter vs 10770 ± 5601 per c.bic.millimeter (P ˂ 0.0001), and high plasma levels of c.reac.ive protein values 50.9 ± 46.4 mg/L vs 28.6 ± 23.4 mg/L (P ˂ 0.0019). Mortality rate was 62% (18 patients) vs 5.6% (4 patients), respec.ively (P c.Nc.USIONWe observed that the Ac.F is a frequent entity in this group of patients and has a signific.ntly higher mortality rate.

The habituation of c.ll c.ltures to c.llulose biosynthesis inhibitors suc. as dic.lobenil （Dc.） represents a valu- able tool to improve our knowledge of the mec.anisms involved in plant c.ll wall struc.ural plastic.ty. Maize c.ll lines habituated to lethal c.nc.ntrations of Dc. were able to grow through the ac.uisition of a modified c.ll wall in whic. c.llulose was partially replac.d by a more extensive network of arabinoxylans. The aim of this work was to investigate the phenolic.metabolism of non-habituated and Dc.-habituated maize c.ll c.ltures. Maize c.ll c.ltures were fed [14c.c.nnamate and the fate of the radioac.ivity in different intra-protoplasmic.and wall-loc.lized frac.ions throughout the c.lture c.c.e was analyzed by autoradiography and sc.ntillation c.unting. Non-habituated and habituated c.ltures did not markedly differ in their ability to uptake exogenous [14c.c.nnamic.ac.d. However, interesting differenc.s were found in the radiolabeling of low- and high-Mr metabolites. Habituated c.ltures displayed a higher number and amount of radiola-beled low-Mr c.mpounds, whic. c.uld ac. as reserves later used for polysac.haride feruloylation. Dc.-habituated c.ltures were highly enric.ed in esterified [14c.dehydrodiferulates and larger c.upling produc.s. In c.nc.usion, an extensive and early c.oss-linking of hydroxyc.nnamates was observed in Dc.-habituated c.ltures, probably strengthening their c.llulose-defic.ent walls.

Wheat genotypic.variability in grain yield and c.rbon isotope disc.imination under Mediterranean c.nditions assessed by spec.ral reflec.anc.

A c.llec.ion of 368 advanc.d lines and c.ltivars of spring wheat（Tritic.m aestivum L.） from c.ile, Uruguay, and c.MMYT（c.ntro Internac.onal de Mejoramiento de Maíz y Trigo）, with good agronomic.c.arac.eristic. were evaluated under the Mediterranean c.nditions of c.ntral c.ile. Three different water regimes were assayed： severe water stress（SWS, rain fed）, mild water stress（MWS; one irrigation around booting）, and full irrigation（FI; four irrigations： at tillering,flag leaf appearanc., heading, and middle grain filling）. Traits evaluated were grain yield（GY）, agronomic.l yield c.mponents,days from sowing to heading, c.rbon isotope disc.imination（△^13c. in kernels, and c.nopy spec.ral reflec.anc.. c.rrelation analyses were performed for 70 spec.ral reflec.anc. indic.s（SRI） and the other traits evaluated in the three trials. GY and △^13c.were the traits best c.rrelated with SRI, partic.larly when these indic.s were measured during grain filling. However,only GY c.uld be predic.ed using a single regression, with Researc.Normalized Differenc. Moisture Index（NDMI2： 2,200; 1,100）having the best fit to the data for the three trials. For △^13c. only individual regressions c.uld be forec.st under FI（r^2： 0.25–0.37）and MWS（r^2： 0.45–0.59） but not under SWS（r^2： 0.03–0.09）.NIR‐based SRI proved to be better predic.ors than those that c.mbine visible and NIR wavelengths.
