evaluation of Different Parasitological Techniques for Diagnosing Intestinal Parasites in Dogs

Intestinal nematodes are among the most common parasites of dogs, being Toxocara spp. and Ancylostoma spp. threats to public health due to their zoonotic potential. In this study, three parasitological diagnostic techniques for detecting the eggs/oocysts of canine intestinal parasites were evaluated. Fecal samples (n = 285) were collected from the environment (i.e. public streets, squares and homes) in Garanhuns, Pernambuco, Brazil. Samples were analyzed using the Willis-Mollay simple flotation technique, spontaneous sedimentation (Hoffman, Pons and Janer) and modified centrifugal flotation. Among the samples analyzed, 56.49% (161/285) showed eggs and/or oocysts of intestinal parasites. In particular, 44.21% (126/285) were detected by the Willis technique, 45.14% (121/268) by centrifugal flotation and 31.57% (90/285) by the Hoffmann technique. The kappa index indicated good concordance between the centrifugal flotation and Willis techniques (κ = 0.625), whereas fair concordance was observed between Hoffmann and Willis (κ = 0.480), and between Hoffman and centrifugal flotation (κ = 0.433). In conclusion, the centrifugal flotation technique showed high sensitivity towards detecting Ancylostoma spp. and Toxocara spp. (the most common parasites in this study) and could be used in association with the Willis technique to obtain safe diagnoses within routine laboratory analysis.

Rapid Differentiation of Closely Related Citrus Genotypes by Fluorescence Spectroscopy

The differentiation of closely related Citrus genotypes is a meticulous, laborious, and time-consuming task that involves the assessment of complex traits such as growth, tolerance to stress, photosynthetic efficiency, yield, and many others. Such a task is generally accomplished either by analyzing specific features of adult plants or by applying molecular markers to young trees. On one hand, only after plants start yielding can distinct genotypes be differentiated by comparing their fruit sizes, shapes, taste, and the number of seeds. On the other hand, molecular markers are expensive, and demand expertise and time for the analysis of a larger number of plants. For these reasons, the development of techniques that could assist in an early, quick and accurate differen-tiation of closely related Citrus varieties is of utmost importance. In this context, laser-induced fluorescence spectroscopy (LIFS) is a promising technique, since it is rapid, highly sensitive, and inexpensive. Previous studies showed that LIFS can differentiate a variety of sweet orange. However, this new study aimed to determine LIFS accuracy in the differentiation and grouping of very closely varieties of four Sunki mandarin selections: Comum, Florida, Tropical, and Maravilha. Furthermore we compared the results with ISSR and SSR molecular markers for the same varieties. LIFS technique distinguished the four selections with accuracy greater than 70%. Only with molecular markers was possible distinguishing clearly Tropical from Maravilha, but not Comum from Florida selections. In this way the results suggest that LIFS may be a sound tool for helping the identification of closely Citrus varieties.

The coconut mite, Aceria guerreronis Keifer (Acari: eriophyidae), is a major pest of coconut plantations (Cocos nucifera L.) worldwide. Here, we conducted a bioguided phytochemical approach using toxicity and repellency bioassays of nonpolar extract and its fractions of Vitex gardneriana Schauer (Lamiaceae) leaves to this pest. Nonpolar crude extract was fractionated by column chromatography using solvents with increased polarity and binary mixtures, resulting in five semipurified groups. The biomonitoring bioassay provided active fractions and led to the isolation and characterization of the bioactive compound squalene, a biosynthetic precursor of 20-hydroxyecdysone, which plays an important role in plant defense against arthropods. The LC50 of the crude extract of V. gardneriana for A. guerreronis was estimated to be 0.185 mg·mL-1 and LC80 = 4.123 mg·mL-1. Also, the extract was highly repellent to this pest for up to 24 h. The fractions of V. gardneriana, and also squalene, caused mortality to A. guerreronis. The potential of V. gardneriana fractions/squalene as biopesticides for controlling A. guerreronis in coconut plantations is discussed herein.

Comparison of Two Protocols of Agar Gel Immunodiffusion(AGID)Used to Diagnose of equine Infectious Anemia(eIA)

The aim of the present study was to compare two protocols of Agar Gel Immunodiffusion (AGID) that can be used to diagnose equine Infectious Anemia (eIA). Different volumes of reagents and support for agar gel were used, as recommended by the Ministry of Agriculture, Livestock and Food Supply (MAPA) and the World Organization for Animal Health (OIe). equidae sera samples (n = 1000) were tested using a national commercial kit and different volumes (i.e. 25 μL—MAPA and 50 μL—OIe) of reagents and standard sera. Antigen was used as recommended by MAPA. The results were assessed by two operators, 48 hours after beginning the test. At the end of the experiment, a Kappa index of 1 was obtained by the operators, for the interpretation of the results and for the different volumes of reagents used. Similar results of agreement (Kappa index = 1) were obtained when using slides or Petri dishes as support for the agar gel. In both cases, an almost perfect agreement was achieved, suggesting that the use of different supports does not affect the results. These findings indicate that both protocols (MAPA and OIe) can be used to diagnose eIA.

Background: Dengue is a Neglected tropical disease (NTDs) with high incidence in Brazil. This disease is caused by Dengue virus and is transmitted by Aedes aegypti mosquito. The search for new approaches for controlling of this disease is the subject of numerous studies. The aaNAT is a key enzyme in the metabolism of A. aegypti and is crucial in the sclerotization process, as well as regulation of circadian rhythm and inactivation of neurotransmitters. Computational techniques applied to studies of biological systems become an effective weapon in the mapping and management of 3D data structures, giving direction and guidance of potential ligands that can form stable complexes with targets of interest, using a Molecular Docking approach. The present study was conducted by a virtual screening, followed by docking calculations, in order to find molecules that could inhibit aaNAT. In this study, we used available compounds in SAM database (Bioinformatics and Medicinal Chemistry Laboratory—Southwest Bahia State University, Jequié-Bahia, Brazil), PubChem and ZINC. Results: The result of dockings with selected ligands showed good energy affinities, presenting potential inhibitory interactions with the enzyme active site. Conclusions: The Coa-S-acetyl-tryptamine and 3-indoleacriloil-coenzyme-A showed the same binding energies -8.9 Kcal/Mol and were described as possible inhibitors of aaNAT.

Clique Approach for Networks:Applications for Coauthorship Networks

Coauthorship networks consist of links among groups of mutually connected authors that form a clique. Classical approaches using Social Network Analysis indices do not account for this characteristic. We propose two new cohesion indices based on a clique approach, and we redefine the network density using an index of variance of density. We have applied these indices to two coauthorship networks, one comprising researchers that published in Mathematics education journals and the other comprising researchers from a Computational Modeling Graduate Program. A contextualized and comparative analysis was performed to show the applicability and potential of the indices for analyzing social networks data.

Contextualized Analysis of Social Networks:Collaboration in Scientific Communities

Currently, the collaboration in scientific communities has been studied in order to explain, among other things, the knowledge diffusion. The quality of Graduate Programmes is often associated with the scientific collaboration. This paper discusses how scientific collaboration processes can be identified and characterized through social and complex networks. For this purpose, collaboration networks of bibliographic production, research projects, and committees of PhD theses and Masters’ dissertations by researchers from a graduate program in computational modeling were studied. The data were obtained from CAPeS’ reports of the period from 2001 to 2009. Among the studied indices, centrality indices indicate the presence of prominent researchers who influence others and promptly interact with other researchers in the network. The indices of complex networks reveal the presence of the small-world (i.e. these networks are favorable to increase coordination between researchers) phenomenon and indicate a behavior of scale-free degree distribution (i.e. some researchers promote clustering more than others) for one of the studied networks.
