Modulation of a Specific Pattern of microRNAs, Including miR-29a, miR-30a and miR-34a, in Cultured Human Skin Fibroblasts, in Response to the Application of a Biofunctional Ingredient that Protects against Cellular Senescence <i>in Vitro</i>

Skin aging is a process of structural and compositional remodeling that can be manifested by wrinkling and sagging. Remarkably, the dermis plays a dominant role in the aging process. Recent studies suggest that microRNAs are implicated in the regulation of gene expression during aging. However, studies about age-related microRNAs and how they modulate skin aging remain limited. In the present work, a complex of hydrolyzed natural yeast proteins (Saccharomyces cerevisiae) and hydrolyzed natural soya bean was developed and showed the ability to modulate the expression of telomere-binding protein TRF2, which is a key factor for telomere protection and to prevent cellular senescence in vitro and DNA damage. The aim of the study was to identify microRNAs specifically modulated after application of the ingredient complex to cultured fibroblasts, and their possible involvement in remodeling of the human extracellular matrix and fibroblast senescence. Consequently, human skin fibroblasts were cultured and treated with 1% of the ingredient complex for 48 h before analyzing microRNA modulation by RT-qPCR. The use of bioinformatics allowed us to predict the target genes for modulated microRNAs. Results show that the ingredient complex modulated a pattern of microRNAs including the down-regulation of miR-29a-3p, miR-30a-5p and miR-34a-5p, which are associated with fibroblast senescence and remodeling of the human dermal extracellular matrix. In conclusion, our results indicate that miR-29a-3p, miR-30a-5p and miR-34a-5p possibly represent key microRNAs that impact human fibroblast senescence and remodeling of the dermal extracellular matrix.

A multi-channel chemical sensor and its application in detecting hydrothermal vents

There are well-established chemical and turbidity anomalies in the plumes occurring vicinity of hydrothermal vents, which are used to indicate their existence and locations. We here develop a small, accurate multi-channel chemical sensor to detect such anomalies which can be used in deep-sea at depths of more than 4 000 m. The design allowed five all-solid-state electrodes to be mounted on it and each (apart from one reference electrode) could be changed according to chemicals to be measured. Two experiments were conducted using the chemical sensors. The first was a shallow-sea trial which included sample measurements and in situ monitoring. pH, Eh, CO3^2- and SO4^2- electrodes were utilized to demonstrate that the chemical sensor was accurate and stable outside the laboratory. In the second experiment, the chemical sensor was integrated with pH, Eh, CO3^2- and H2S electrodes, and was used in 29 scans of the seabed along the Southwest Indian Ridge (SWIR) to detect hydrothermal vents, from which 27 sets of valid data were obtained. Hydrothermal vents were identified by analyzing the chemical anomalies, the primary judging criteria were decreasing voltages of Eh and H2S, matched by increasing voltages of pH and CO3^2- . We proposed that simultaneous detection of changes in these parameters will indicate a hydrothermal vent. Amongst the 27 valid sets of data, five potential hydrothermal vents were targeted using the proposed method. We suggest that our sensors could be widely employed by marine scientists.

The paper comprises new analytical data on the nature and occurrence of gold in solid pyrobitumen,closely associated with the main gold-bearing sulfide arsenic ores of the Bakyrchik gold deposit(Kazakhstan),related to post-collisional magmatic-hydrothermal *** mineralization of the deposit occurs mainly in the form of an“invisible”type of gold in the structures of arsenian pyrite and arsenopyrite,and the form of gold-organic compounds of pyrobitumen in carbonaceous-terrigenous sequences of Carboniferous *** and electron microscopic analysis,Raman and FT-Infrared analysis,mineralogical and three-step sequential extraction analysis(NH2OHHCl,H2O2,HNO3?HCl)has been carried out using 9 ore samples(from 3 different types of ores)for a comprehensive study of pyrobitumen and sulfide arsenic ores focusing mainly on organic *** sequentially extracted precious metal content of pyrobitumen reaches up to 7 ppm gold and other metals like Ag 4 ppm,Pt 31 ppb,and Pd 26 ppb,forming metal–organic compounds,while arsenic sulfide minerals incorporate 11 ppm gold,39 ppm Ag,0.49 ppm *** enrichment of gold associating with organic matter and sulfide ore minerals was confirmed in this *** matter was active in the migration of gold and the capture of gold by ***,the reductive organic matter agent released gold,most likely for the sulfide arsenic ore *** was a decisive factor in the concentration,transportation,and preservation of gold in the deposit.

A Semi-Field Approach to Testing Botanical Insecticides. Effects of Natural and Analogues Annonaceous Acetogenins on <i>Spodoptera frugiperda</i>Smith (Lepidoptera: Noctuidae)

Annonaceous acetogenins enclose a large number of biological activities, among which the insecticidal stands out. Spodoptera frugiperda is a pest that affects corn crops among others and has a great capacity to develop resistance to traditional insecticides, which represents sufficient reasons for the search for new alternatives for their control. The objective of this study was to determine the appropriate concentration and screening new natural insecticides against second instar larvae of S. frugiperda, under glasshouse conditions on Zea mays L. Natural products such as annonaceous acetogenins and some acetylated and methoxy methylated ACG derivatives and the commercial product Lambda-cyhalotrin (LC) were evaluated. The percent mortalities of S. frugiperda larvae in glasshouse conditions were recorded after 24, 48 and 72 h of treatment application. The results showed that the acetogenins tested were significantly (P 5) (100 μg/mL) + squamocin (6) (100 μg/mL) + LC (50 μg/mL) that proved the most effective and gave (100%), while rolliniastatin-2 (5) at 100 μg/mL alone, gave lowest percent mortality (65%), followed by squamocin (6) at 100 μg/mL (55%) and LC at 50 μg/mL (30%). The acetylated and methoxy methylated ACGs derivatives caused very low mortality (25% - 35%). It is recommended the mixture as a management option of S. frugiperda as a component of integrated pest management. The results allow us to infer that the addition of natural ACGs synergizes the insecticidal activity of the commercial product. Finding a new ecological alternative for insect control.
