Recombineering is an essential tool for molecular biologists,allowing for the facile and efficient manipulation of bacterial genomes directly in cells without the need for costly and laborious in vitro manipulations involving restriction *** main workhorses behind recombineering are bacteriophage proteins that promote the single-strand annealing(SSA)homologous recombination pathway to repair double-stranded DNA *** there have been several reviews examining recombineering methods and applications,comparatively few have focused on the mechanisms of the proteins that are the key players in the SSA pathway:a 5′→3′exonuclease and a single-strand annealing protein(SSAP or“annealase”).This review dives into the structures and functions of the two SSA recombination systems that were the first to be developed for recombineering in ***:the RecET system from *** Rac prophage and the𝜆Red system from bacteriophageλ.By comparing the structures of the RecT and Red𝛽annealases,and the RecE and𝜆Exo exonucleases,we provide new insights into how the structures of these proteins dictate their *** the sequence conservation of the𝜆λExo and RecE exonucleases gives more profound insights into their critical functional ***,as recombineering accelerates and evolves in the laboratory,a better understanding of the mechanisms of the proteins behind this powerful technique will drive the development of improved and expanded capabilities in the future.

Establishment of pregnancy in pigs involves maintaining progesterone secretion from the corpora lutea in addition to regulating a sensitive interplay between the maternal immune system and attachment of the rapidly expanding trophoblast for nutrient absorption. The peri-implantation period of rapid trophoblastic elongation followed by attachment to the maternal uterine endometrium is critical for establishing a sufficient placental-uterine interface for subsequent nutrient transport for fetal survival to term, but is also marked by the required conceptus release of factors involved with stimulating uterine secretion of histotroph and modulation of the maternal immune system. Many endometrial genes activated by the conceptus secretory factors stimulate a tightly controlled proinflammatory response within the uterus. A number of the cytokines released by the elongating conceptuses stimulate inducible transcription factors such as nuclear factor kappa B （NFKB） potentially regulating the maternal uterine proinflammatory and immune response. This review will establish the current knowledge for the role of conceptus cytokine production and release in early development and establishment of pregnancy in the pig.

Communication after cancellations in orthopaedics: The patient perspective

AIM: To examine patients' perceptions on communication surrounding the cancellation of orthopaedic operations and to identify areas for improvement in ***: A prospective survey was undertaken at a university teaching hospital within the department of Trauma and Orthopaedics. Patients admitted to an acute orthopaedic unit, whose operations were cancelled, were surveyed to assess patient satisfaction and preferences for notification of cancellation of their operations. Patients with an abbreviated mental test score of < 9, patients unable to complete the survey independently, those under 16 years of age, and any patient notified of the cancellation by any of the authors were excluded from this study. Patients were surveyed themorning after their operation had been cancelled thus ensuring that every opportunity was given for the medical staff to discuss the cancellation with the patient. The survey included questions on whether or not patients were notified of the cancellation of their surgery, the qualifications of the person discussing the cancellation, and patient preferences on the process. Satisfaction was assessed via 5-point Likert scale ***: Sixty-five consecutive patients had their operations cancelled on 75 occasions. Fifty-four point seven percent of the patients who had cancellations were notified by a nurse and 32% by a doctor. No formal communication occurred for 13.3% cancellations and no explanation was provided for a further 16%. Patients reported that they were dissatisfied with the explanation provided for 36 of the 75(48%) cancellations. Of those patients who were dissatisfied, 25(69.4%) were notified by a nurse. Twenty-three of the 24(96%) patients notified by a doctor were satisfied with the explanation and that communication. Of those patients who were notified by a nurse 83% patients reported that they would have preferred it if a doctor had discussed the cancellation with them. There was a significant difference

This paper introduces the online Sociometrics Social, Behavioral, and Health Sciences Library, an exciting new science-based resource for HIV/AIDS researchers, health educators, and clinicians. The over 400 products in the Sociometrics Library supplement the online publications—journal articles, books, reports, monographs—that have been the focus of scientific research libraries and publishers to date, both printed and online. Examples of the innovative science-based products that serve as the library’s content include: Evidence-based interventions and programs (EBIs/EBPs) that evaluation research has shown to be effective in preventing HIV or its risky social and behavioral antecedents;primary research data and survey instruments;and interactive, multimedia training tools and courses to build HIV professionals’ capacity to implement EBPs with fidelity and to cooperate with evaluators in the assessment of their effectiveness. A Scientist Expert Panel has guided and will continue to guide product selection and acquisition, ensuring the collection’s continuing technical merit, research utility, and relevance for practice and policy. The Sociometrics Library aims to become the dominant online source of behavioral and social science-based HIV research by-products, operationally sustainable and able to stay up-to-date both from a technological and scientific perspective.

Tropical cyclones are devastating hazards and have been a major problem for the coastal population of B *** the advancements in atmospheric and oceanic prediction,accurate forecasting of storm surges is of specific interest due to their great potential to inflict loss of life and *** decades,the numerical model based storm surge prediction systems have been an important tool to reduce the loss of human lives and property *** order to improve the accuracy in predicting storm surge and coastal inundation,recent model development efforts tended to include more modeling components,such as meteorology model and surface wave model in storm surge *** this study,we used the outputs of an atmospheric model to force the ocean model for simulating storm surges in the Bay of Bengal with particular focus on the Bangladesh *** ability of the modeling system was investigated simulating water levels in the Bangladesh coast of two tropical cyclones Sidr(2007)and Aila(2009).The effectiveness of the model was verified through comparing the obtained computational outputs against tide gauge *** cyclone tracks and intensities reproduced by the atmospheric model were reasonable,though the model had a tendency to overestimate the cyclone intensity during peaks and also close to *** water levels are reproduced fairly well by the ocean model,although errors still *** root mean square errors in water level at different gauges range from 0.277 to 0.419 m with coefficient of correlation(R^2)between 0.64 and 0.97 in case of Sidr and0.209-0.581 m with R^20.62 to 0.98 for *** overall coupled modeling system is found to be useful with reasonable accuracy and precision,though there are spaces for ***-resolution modeling approaches are recommended to gain more skills.

Parenteral nutrition-associated liver disease(PNALD)is a liver dysfunction caused by various risk factors presented in patients receiving total parenteral nutrition(TPN).Omega-6 rich Intralipid?and omega-3 rich Omegaven?are two intravenous lipid emulsions used in *** could affect the hepatic expression of genes in anti-oxidative stress,but it’s unknown whether TPN affects genes in drug *** this study,either Intralipid?-or Omegaven?-based TPN was administered to mice and the expression of a cohort of genes involved in anti-oxidative stress or drug metabolism was analyzed,glutathione(GSH)levels were measured,and protein levels for two key drug metabolism geneswere ***,the expression of most genes was downregulated by Intralipid?-based TPN(Gstpl,Gstml,3,6,Nqol,Ho-1,Mt-1,Gclc,Gclm,Cyp2d9,2f2,2b 10,and 3a11).Omegaven?showed similar results as Intralipid?except for preserving the expression of Gstml and Cyp3a11,and increasing *** GSH levels were decreased by Intralipid?,but increased by Omegaven?.CYP3A11 protein levels were increased by Omegaven?.In conclusion,TPN reduced the expression of many genes involved in anti-oxidative stress and drug metabolism in ***,Omegaven?preserved expression of Cyp3a11,suggesting another beneficial effect of Omegaven?in protecting liver functions.

The liver plays a central role in the pharmacokinetics of drugs through drug metabolizing enzymes and ***-alcoholic steatohepatitis(NASH)causes disease-specific alterations to the absorption,distribution,metabolism,and excretion(ADME)processes,including a decrease in protein expression of basolateral uptake transporters,an increase in efflux transporters,and modifications to enzyme *** can result in increased drug exposure and adverse drug reactions(ADRs).Our goal was to predict drugs that pose increased risks for ADRs in NASH *** research identified 71 drugs with reported ADRs in patients with liver disease,mainly non-alcoholic fatty liver disease(NAFLD),54 of which are known substrates of transporters and/or metabolizing *** NASH is the progressive form of NAFLD but is most frequently undiagnosed,we identified other drugs at risk based on NASH-specific alterations to ADME ***,we present another list of 71 drugs at risk of pharmacokinetic disruption in NASH,based on their transport and/or metabolism *** encompasses drugs from various pharmacological classes for which ADRs may occur when used in NASH patients,especially when eliminated through multiple pathways altered by the ***,these results may inform clinicians regarding the selection of drugs for use in NASH patients.

The cultural universe is sometimes confusing,surprising and murky,so many cultural maps get drawn,discussed and envisioned.A study was undertaken around Mabira Forest Reserve in central Uganda to identify the trees and shrubs culturally managed on-farm,assess the cultural practices of forest and tree system management and determine the relationship between farmer gender and forest and tree system *** engaged 203 farmers in focus group discussions and semi-structured interviews to collect *** data were jointly evaluated with farmers;quantitative data were analyzed in SPSS *** results showed a high likelihood for involvement of local people in tree or forest management for economic gain,as timber and fast-growing species were highly *** and medicinal species were also regarded as important,suggesting high prospects of integrating them into the local farming system or protecting them in the *** cultural practices(including rituals,trenching,bark slashing,ring barking,spot weeding and use of organic manure and pesticides)of forest and tree system management were ***,their knowledge was mixed and unclear about distinct cultural and supportive arrangements for natural forest and tree *** gender was not a significant cultural attribute for knowledge of the forest and allied tree system management,age substantially affected farmer propensity for various timber *** farmer’s family size influenced the collection of tree wildings and *** encourage considering gender disparities and livelihood needs including income,during selection of cultural practices for forest and tree restoration.
