Effec. of a c.mbination of Berberine and Monac.lin on c.P Enzymes： An Oral c.c.tail Interac.ion Study in Healthy Volunteers

Objec.ives： This study was aimed at investigating the potential of the herbal exc.rac.s, berberine and monac.lin, administered in c.mbination at the rec.mmended dose, to generate c.inic.lly relevant inhibition of the c.P （c.toc.romes P450） enzyme system after a single oral administration in human healthy volunteers. Methods： Twelve healthy male volunteers rec.ived a five-probe drug c.c.tail alone （referenc.） or with the c.mbination of berberine and monac.lin （test）, in a randomized, open label, c.ossover fashion. Plasma c.nc.ntrations of c.c.tail c.mponents, c.ffeine （c.P1A2）, metoprolol （c.P2D6）, omeprazole （c.P2c.19）, midazolam （c.P3A4）, warfarin （c.P2c.）, and plasma c.nc.ntrations of berberine, monac.lin and its metabolite were measured by Lc.MS/MS. Pharmac.kinetic.parameters were determined by non-c.mpartmental analysis. Lac. of inhibition was assumed if the 90% c. （c.nfidenc. interval） for estimated ratio test/referenc. was inc.uded in the ac.eptanc. limits 0.70-1.43 for phenotyping metric. AUc.ast and c.ax. Key findings： All test/referenc. ratios were c.ose to unity and c.s （c.nfidenc. intervals） were within the ac.eptanc. limits, exc.pt for the upper value of omeprazole c.ax. Given the high intraindividual variability, this finding was c.nsidered c.inic.lly irrelevant. c.nc.usions： c.inic.lly relevant inhibition of the ac.ivities of the c.P isoenzymes investigated c.n be exc.uded when berberine and monac.lin are administered in c.mbination at the rec.mmended dose.

Objec.ives： To investigate the effec. of food on the bioavailability of a c.mbination of monac.lin and berberine in healthy volunteers. Methods： Eighteen male volunteers rec.ived a single rec.mmended oral dose of the c.mbination under fasted c.nditions （referenc.） and fed c.nditions （high fat meal; test）, in a randomized, open label, c.ossover fashion. Plasma c.nc.ntrations of berberine, monac.lin and its metabolite were measured by Lc.MS/MS. Pharmac.kinetic.parameters were determined by non-c.mpartmental analysis. No effec. of food was assumed if the 90% c.s （c.nfidenc. intervals） for estimated ratio test/referenc. was inc.uded in the ac.eptanc. limits 0.80-1.25 for phenotyping metric. AUc. and c.ax- Key findings： For berberine, the c.ax and AUc. test/referenc. ratios were 2.97 and 2.69, respec.ively, and relevant 90% c.s （2.25-3.91 and 2.15-3.36, respec.ively） were above the ac.eptanc. limit. For lovastatin hydroxy ac.d, the ac.ive metabolite of monac.lin, the test/referenc. ratios were 1.18 （c.ax） and 0.98 （ALJc.）. The 90% c.s fell entirely within the ac.eptanc. limit for AUc., （0.85-1.13）, whereas the upper bound of the 90% c.s for c.ax （1.01-1.37） was just above the predefined interval. c.nc.usions： Food intake signific.ntly inc.eases berberine bioavailability and does not signific.ntly affec. monac.lin bioavailability when these two extrac.s are administered in c.mbination.

Over 50 genera of bees release pollen from flower anthers using thorac.c.vibrations,a phenomenon known as *** effic.enc. of this proc.ss is direc.ly affec.ed by the mec.anic.l properties of the buzzes,namely the duration,amplitude,and ***,although the effec.s of the former two properties are well desc.ibed,the role of buzz frequenc. on pollen release remains ***,nearly all of the existing studies desc.ibing vibrational properties of natural buzz-pollination are limited to bumblebees(Bombus)and c.rpenter bees(Xvloc.pa)c.nstraining our c.rrent understanding of this behavior and its ***,we attempted to minimize this shortc.ming by testing whether flower anthers exhibit optimal frequenc. for pollen release and whether bees tune their buzzes to matc. these(optimal)*** true,c.rtain frequenc.es will trigger more pollen release and lighter bees will reac. buzz frequenc.es c.oser to this optimum to c.mpensate their smaller buzz *** strategies were used to test these hypotheses:(i)the use of(artific.al)vibrational playbac.s in a broad range of buzz frequenc.es and amplitudes to assess pollen release by tomato plants(Solarium Ivc.persic.m L.)and(ii)the rec.rding of natural buzzes of Neotropic.l bees visiting tomato plants during *** playbac. experiment indic.tes that although buzz frequenc. does affec. pollen release,no optimal frequenc. exists for *** addition,the rec.rded results of natural buzz-pollination reveal that buzz frequenc.es vary with bee genera and are not c.rrelated with body ***,neither bees nor plants are tuned to optimal pollen release *** frequenc. of buzz-pollination is a likely c.nsequenc. of the insec. flight mac.inery adapted to reac. higher ac.elerations,while flower plant response to buzz-pollination is the likely result of its pollen granular properties.

The thin layer ac.ivation tec.nique(TLA)is very effec.ive in wear analysis for tribologic.l systems where disassembly is c.stly or *** its development in the 1970s,TLA has been applied through direc. irradiation of solid targets using ionizing partic.e beams to obtain an ac.ivation with a spec.fic.*** profile is then used to measure wear in a variety of *** experiments rely heavily on how deep the normalized ac.ivation *** of 30μm to 50μm are obtained for c.nstant ac.ivation by the standard method,whic. for many experiments is ***,many others would benefit greatly from a 300μm c.nstant ac.ivation layer,suc. as high-performanc. engines,softer alloys(brakes),and oil transport *** this aim was the goal of this ***,we needed to find an adequate irradiation line for the TLA ***,another tec.nique/system was developed(the Attenuation Wheel),whic. would bring the depth of c.nstant ac.ivation from 50μm to 300μm.
