Objective:Bone metastasis is a clinically important outcome of prostate carcinoma(PC).We focused on the phenotypic and functional characterization of a particularly aggressive phenotype within the androgen-independent bone metastasis-derived PC3 cell *** cells,originated from the spontaneous conversion of a Cd44-negative subpopulation,stably express the Cd44 v8-10 isoform(Cd44 v8-10 pos)and display stem cell-like features and a marked invasive phenotype in vitro that is lost upon Cd44 v8-10 ***:Flow cytometry,enzyme-linked immunoassay,immunofluorescence,and Western blot were used for phenotypic and immunologic ***-time quantitative polymerase chain reaction and functional assays were used to assess ***:Analysis of epithelial–mesenchymal transition markers showed that Cd44 v8-10 pos PC3 cells surprisingly display epithelial phenotype and can undergo osteomimicry,acquiring bone cell phenotypic and behavioral *** of specific si RNA evidenced the ability of Cd44 v8-10 variant to confer osteomimetic features,hence the potential to form bone-specific ***,the ability of tumors to activate immunosuppressive mechanisms which counteract effective immune responses is a sign of the aggressiveness of a *** we report that Cd44 v8-10 pos cells express programmed death ligand 1,a negative regulator of anticancer immunity,and secrete exceptionally high amounts of interleukin-6,favoring osteoclastogenesis and immunosuppression in bone ***,we identified a novel pathway activated by Cd44 v8-10,involving tafazzin(TAZ)and likely the Wnt/TAZ axis,known to play a role in upregulating osteomimetic ***:Cd44 v8-10 could represent a marker of a more aggressive bone metastatic PC population exerting a driver role in osteomimicry in bone.A novel link between TAZ and Cd44 v8-10 is also shown.

Minimal crystallographic descriptors of sorption properties in hypothetical MOFs and role in sequential learning optimization

We focus on gas sorption within metal-organic frameworks(MOFs)for energy applications and identify the minimal set of crystallographic descriptors underpinning the most important properties of MOFs for CO_(2)and H_(2)O.A comprehensive comparison of several sequential learning algorithms for MOFs properties optimization is performed and the role played by those descriptors is *** energy transformations,thermodynamic limits of important figures of merit crucially depend on equilibrium properties in a wide range of sorbate coverage values,which is often only partially accessible,hence possibly preventing the computation of desired objective *** propose a fast procedure for optimizing specific energy in a closed sorption energy storage system with only access to a single water Henry coefficient value and to the specific surface *** are thus able to identify hypothetical candidate MOFs that are predicted to outperform state-of-the-art water-sorbent pairs for thermal energy storage applications.

Mass-forming pancreatitis: Value of contrast-enhanced ultrasonography

AIM： To assess the utility of contrast-enhanced ultrasonography （CEUS） with a second-generation contrast medium in the differential diagnosis between mass-forming pancreatitis and pancreatic carcinoma. METHOdS： From our radio-pathology database, we retrieved all the patients affected by mass-forming pancreatitis or pancreatic carcinoma who underwent CEUS. We evaluated the results of CEUS in the study of the 173 pancreatic masses considering the possibilities of a differential diagnosis between mass-forming pancreatitis and pancreatic tumor by identifying the ＂parenchymographic＂ enhancement during the dynamic phase of CEUS, which was considered diagnostic for mass-forming pancreatitis. RESULTS： At CEUS, 94% of the mass-forming pancreatitis showed intralesional parenchymography. CEUS allowed diagnosis of mass-forming pancreatitis with sensitivity of 88.6%, specificity of 97.8%, positive predictive value of 91.2%, negative predictive value of 97.1%, and overall accuracy of 96%. CEUS significantly increased the diagnostic confidence in the differential diagnosis between mass-forming pancreatitis and pancreatic carcinoma, with receiver operating characteristic curve areas from 0.557 （P = 0.1608） for baseline US to 0.956 （P 〈 0.0001） for CEUS. CONCLUSION： CEUS allowed diagnosis of massforming pancreatitis with diagnostic accuracy of 96%. CEUS significantly increases the diagnostic confidence with respect to basal US in discerning mass-forming pancreatitis from pancreatic neoplasm.

Heat Transfer in Internal Channel of a Blade:Effects of Rotation in a Trailing Edge Cooling System

The aerothermal performance of a trailing edge （TE） internal cooling system of a high pressure gas turbine blade was evaluated under stationary and rotating conditions. The investigated geometry consists of a 30：1 scaled model reproducing a typical wedge shaped discharge duct with one row of enlarged pedestals. The airflow pattern inside the device simulates a highly loaded rotor blade cooling scheme with a 90 [deg] turning flow from the radial hub inlet to the tangential TE outlet. Two different tip configurations were tested, the first one with a completely closed section, the second one with a 5 holes outlet surfaces discharging at ambient pressure. In order to assess rotation effects, a rotating test rig, composed of a rotating ann holding both the PMMA TE model and the instru mentation, was purposely developed and manufactured. A thin Inconel heating foil and wide band Thermochromic Liquid Crystals are used to perform steady state heat transfer measurements on the blade pres sure side. A rotary joint ensures the pneumatic connection between the blower and the rotating apparatus; more over several slip rings are used for both instrumentation power supply and thermocouple connection. A parallel CFd analysis involving steadystate RANS modeling was conducted to allow an insight of the flow field inside the redirecting channel and the interpedestal ducts to better interpret the developing vortical structures. LowReynolds grid clustering permits to integrate up to the wall both the momentum and the thermal boundary layer. Calculations were performed by means of an inhouse developed pressure based solver exploiting the kco SST turbulence model implemented in the framework of the opensource finite volume discretization toolbox OpenFOAM~. Analyzed flow conditions correspond to Reynolds number of 20000 in the hub inlet section and angular speed varies to obtain rotation numbers in the range from 0 to 0.3. A Comprehensive Overview on the Latest Progress in the Additive Manufacturing of Metal Matrix Composites: Potential, Challenges, and Feasible Solutions

Nowadays, as an emerging technology, additive manufacturing(AM) has received numerous attentions from researchers around the world. The method comprises layer-by-layer manufacturing of products according to the 3d CAd models of the objects. Among other things, AM is capable of producing metal matrix composites(MMCs). Hence, plenty of works in the literature are dedicated to developing different types of MMCs through AM processes. Hence, this paper provides a comprehensive overview on the latest research that has been carried out on the development of the powder-based AM manufactured MMCs from a scientific and technological viewpoint, aimed at highlighting the opportunities and challenges of this innovative manufacturing process. For instance, it is documented that AM is not only able to resolve the reinforcement/matrix bonding issues usually faced with during conventional manufacturing of MMCs, but also it is capable of producing functionally graded composites and geometrically complex objects. Furthermore, it provides the opportunity for a uniform distribution of the reinforcing phase in the metallic matrix and is able to produce composites using refractory metals thanks to the local heat source employed in the method. despite the aforementioned advantages, there are still some challenges needing more attention from the researchers. Rapid cooling nature of the process, significantly different coe fficient of expansion of the matrix and reinforcement, processability, and the lack of suitable parameters and standards for the production of defect-free AM MMCs seem to be among the most important issues to deal with in future works.
