Molecular and Chemical analyses of Cyanobacterial Blooms in Tropical Lagoons from Southeast Brazil

The genetic diversity and the potential toxicity of bloom-forming cyanobacteria were studied in four lagoons located in the state of Sao Paulo (Campinas, Limeira and Piracicaba cities). Bloom samples were collected on the water surface and cyanobacterial communities were evaluated using DGGE fingerprinting and 16S rDNA clone library. The amplification of genes encoding secondary metabolites such as microcystin (mcy), anatoxin (ana), cylindrospermopsin (cyr), saxitoxin (sxt), cyanopeptolin (mcn) and aeruginosin (aer) was performed and their production analyzed by LC-MS. The comparison of DGGE banding pattern among the different water samples suggested that some operational taxonomic units (OTUs) in these locations were predominant over others. The 16S rDNA clone libraries sequences matched with nine different known cyanobacterial genera available in NCBI, identified as anabaena, Brasilonema, Cylindrospermopsis, Limnococcus, Microcystis, Nostoc, Pseudanabaena, Synechococcus and Woronichinia. The lagoons ESALQ2, Taquaral and Limeira had more than 80% of the cyanobacterial community assigned to the genus Microcystis. Genes encoding aeruginosin, cyanopeptolin and microcystin synthetases and saxitoxin synthase were amplified, and LC-MS/MS confirmed the production of aeruginosin, cyanopeptolin and microcystin. Rapid and sensitive methods for the detection of these secondary metabolites, especially toxins, using chemical and molecular tools together, can be used for a faster diagnostic of toxic cyanobacterial blooms.

Role of Cognitive Behavioral Therapy in Fibromyalgia: A Systematic Review

A literature search of articles from 2002-2019 was performed using Medline, Embase, Cochrane, LILACS, IBECS, CRD, and Epistemonikos databases, to analyze the effects of Cognitive-Behavioral Therapy (CBT) in the treatment of fibromyalgia. Twenty-seven articles were selected in which CBT was performed exclusively by specialist physicians, associated or not with conventional pharmacological treatment and/or physical exercise. In most articles, CBT worked with self-knowledge and cognitive restructuring, attempting to reduce pain perception, and it showed a general improvement in daily activities by decreasing patient’s limitations, such as morning stiffness. The literature showed significant correlations of CBT in pain processing over time. When CBT was compared to conventional pharmacological therapy, a certain superiority of CBT could be observed concerning the quality of life, catastrophizing, and acceptance of pain. However, when they were simultaneously applied, this improvement in quality of life was not observed.

Despite the advances in the hematology field, blood transfusion-related iatrogenesis is still a major issue to be considered during such procedures due to blood antigenic incompatibility. This places pluripotent stem cells as a possible ally in the production of more suitable blood products. The present review article aims to provide a comprehensive summary of the state-of-theart concerning the differentiation of both embryonic stem cells and induced pluripotent stem cells to hematopoietic cell lines. Here, we review the most recently published protocols to achieve the production of blood cells for future application in hemotherapy, cancer therapy and basic research.

Accuracy of gestalt perception of acute chest pain in predicting coronary artery disease

AIM To test accuracy and reproducibility of gestalt to predict obstructive coronary artery disease(CAD)in patients with acute chest *** We studied individuals who were consecutively admitted to our Chest Pain *** admission,investigators performed a standardized interview and recorded14 chest pain *** on these features,a cardiologist who was blind to other clinical characteristics made unstructured judgment of CAD probability,both numerically and *** the reference standard for testing the accuracy of gestalt,angiography was required to rule-in CAD,while either angiography or non-invasive test could be used to *** order to assess reproducibility,a second cardiologist did the same *** In a sample of 330 patients,the prevalence of obstructive CAD was 48%.Gestalt’s numerical probability was associated with CAD,but the area under the curve of0.61(95%CI:0.55-0.67)indicated low level of ***,categorical definition of typical chest pain had a sensitivity of 48%(95%CI:40%-55%)and specificity of 66%(95%CI:59%-73%),yielding a negligible positive likelihood ratio of 1.4(95%CI:0.65-2.0)and negative likelihood ratio of 0.79(95%CI:0.62-1.02).Agreement between the two cardiologists was poor in the numerical classification(95%limits of agreement=-71%to 51%)and categorical definition of typical pain(Kappa=0.29;95%CI:0.21-0.37).CONCLUSION Clinical judgment based on a combination of chest pain features is neither accurate nor reproducible in predicting obstructive CAD in the acute setting.

Resistance of the Eucalyptus Wood in Natura and Torrefied Exposed to the Attack of Cryptotermes brevis

The reduced natural durability and low energy density of eucalyptus wood hampers its use to generate energy. Torrefaction or pre-carbonization, treatment in low oxygenation with temperatures between 200 ℃ and 300℃, accumulates carbon and lignin, decreases hygroscopicity, increases energy efficiency and reduces the wood attractiveness to xylophagous organisms, such as termites. Therefore, this work had as its main aim to study the influence of the roasting temperature on the endurance of the Eucalyptus urophylla wood in natura as well as roasted, both exposed to the attack of dry-wood termites. To the execution of this study, in natura wood chips and torrefied chips (torrefied for 20 min at the following temperatures: 180, 220 and 260 ℃) were submitted to the dry-wood termite resistance test. In this experiment, termites of the Cryptotermes brevis species were used. After 45 d of exposure, it was possible to observe that the torrefied treatment presented a greater resistance that consequently increased the endurance when exposed to the termite’s attack, observing that the control sample, loss five times more than mass than the chips torrefied at 260℃. Besides, in the treatment with in natura chips, was observed less mortality of the termites and greater visual damage, confirming the lower durability of such material compared to torrefied chips.
