“Stem cells is what stem cells does” not Forrest Gump In the present day Stem cells are increasingly becoming popularized as the potential “ultimate” cure for the most challenging maladies… the “Daddy of medical intervention”. Forefront Sc research on human induced pluripotent stem cells (iPScs) and other sub-disciplines, is quickly revolutionizing healthcare towards “Regenerative Medicine”, as beautifully exemplified by the use of iPScs in treating and possibly curing osteoarthritis, discussed at the end of this publication. This review documents and reflects on the most topical discoveries in Sc research, and the challenges researchers in this field nowadays face. Major Findings: 1) In 2006 Yamanaka et al. generated the first iPScs from mouse fibroblasts, using retroviral transmission of c-Myc, Oct3/4, Klf4 and SOX2 transcription factors. Later, they successfully generated iPScs from human fibroblasts (2007). 2) contemporary cultivation methods carry high risks of iPSc genome disruption, possibly leading to tumorigenesis, teratoma formation and reducing iPSc induction efficacy. 3) Many studies on preserving genome integrity and decreasing malignancy in iPScs, suggest using valiproic acid and protecting tumour suppressor genes. 4) In many malignant tumours only a small minority of cells, called cancer Stem cells, metastasise and hyper-proliferate. 5) Not all mature cell sources yield the same [undifferentiated iPScs: lineage-committed] ratio as others. Feb 2014: Obokata et al. claimed to have generated iPScs by exposing mature cells to a 25 min, pH 5.7 bath. These iPScs were termed “Stimulus-triggered Acquisition Pluripotency cells” (STAP). However by July 2014 this study had been revoked, as the results could not be replicated. conclusion: Stem cells have enormous potential to offer, especially iPScs. Although currently not

This paper investigates the stabilization effect on compressed earth blocks (cEB) produced from quartz-kaolinite rich earthen material stabilized with 0% - 25% calcium carbide residue (ccR). The paper evaluated various physico-thermal properties of the stabilized cEB and thermal comfort in the model building made of cEB masonry. The optical properties of cEB were evaluated from the mineral composition of the earthen material and ccR and apparent density of the cEB. A simulation was carried out on naturally ventilated model building whose masonry is made of ccR stabilized cEB comparing to the so-called conventional cementitious materials such as cement blocks and concrete. The results showed a decrease of the apparent density of the cEB from 2100 kg·m–3 for unstabilized cEB (0% ccR) to 1600 kg·m–3 for 25% ccR stabilized cEB. The thermal conductivity and depth of penetration of the heat flux on a 24 hours period of cEB respectively decreased from 1 W·m–1·K–1 and 12.7 cm for 0% ccR-cEB to 0.5 W·m–1·K–1 and 10.2 cm for 25% ccR-cEB. The emissivity, solar absorptivity and visible absorptivity of the cEB respectively decreased from 0.82, 0.82 and 0.82 for 0% ccR-cEB to 0.80, 0.64 and 0.64 for 25% ccR-cEB. The number of hours of warm and humid thermal discomfort was impacted for stabilized cEB based masonry in comparison with cement based masonry. The warm discomfort in building made of 20% ccR-cEB masonry was 400 hours lesser than that in building made of hollow cement blocks masonry. If air conditioning system is used to keep the indoor temperature below 28°c, the economy of 310,000 cFA francs (535 USD) is made every year on energy consumption for cooling in the model building made of 20% ccR-cEB masonry, corresponding to 9.6% less, with respect to that made of hollow cement blocks masonry.

Dynamics of ^(14)c-labelled Gluc.se and NH_4^+ in a Regularly Flooded Extremely Alkaline Saline Soil

Flooding an extremely alkaline(pH 10.6) saline soil of the former Lake Texcoco to reduce salinity will affect the soil carbon(c)and nitrogen(N) dynamics.A laboratory incubation experiment was done to investigate how decreasing soil salt content affected dynamics of c and N in an extremely alkaline saline *** soil with electrical conductivity(Ec) of 59.2 dS m^(-1) was packed in columns,and then flooded with tap water,drained freely and conditioned aerobically at 50%water holding capacity for a *** process of flooding-drainage-conditioning was repeated eight *** original soil and the soil that had undergone one,two,four and eight flooding-drainage-conditioning cycles were amended with 1000 mg gluc.se-^(14)c kg^(-1) soil and 200 mg NH_4^+-N kg^(-1)soil,and then incubated for 28 *** cO_2 emissions,soil microbial biomass,and soil ammonium(NE_4^+),nitrite(NO_2^-) and nitrate(NO_3^-) were monitored in the aerobic incubation of 28 *** soil Ec decreased from 59.2 to 1.0 dS m^(_1) after eight floodings,and soil pH decreased from 10.6 to *** the added ^(14)c-labelled gluc.se,only 8%was mineralized in the original soil,while 24%in the soil flooded eight times during the 28-d *** priming effect was on average 278 mg c kg^(-1) soil after the 28-d *** microbial biomass c(mean 66 mg c kg^(-1) soil) did not change with flooding times in the unamended soil,and increased 1.4 times in the gluc.se-NH_4^+-amended *** immobilization and NO_2^- concentration in the aerobically incubated soil decreased with increasing flooding times,while NO_3^- concentration *** was found that flooding the Texcoco soil decreased the Ec sharply,increased mineralization of gluc.se,stimulated nitrification,and reduced immobilization of inorganic N,but did not affect soil microbial biomass c.

A Method for Detection of Trace concentrations of Underivatized Amino Acid in Hydrothermal Fluids by Ion-Pairing Reversed-Phase UPLc-ESI-QTOF-MS

Investigation of amino acids in hydrothermal systems is of prime importance for the understanding of geochemistry and microbiology of hydrothermal vents and plumes, for carbon and metals global cycles, for metabolism of some hydrothermal microorganisms and for the origin of life issue. Extensive theoretical and experimental work on amino acids behaviour in hydrothermal fluids has been done, conversely only few data exist on natural samples. Because each hydrothermal vent is unique, the more data we collect the better we will be able to address each of these questions. Usually amino acids in hydrothermal fluids have been measured by HPLc-FLD. The chromatographic separation was at least 26 min and up to 135 min and the required derivatization step may be time consuming, may use harmful chemicals and may be source of contamination. Alternatively, we describe here a method combining quickness (4.5 min), high resolution (10,000), very low LOD (sub-ppb) and without derivatization. characterisation and separation of 10 relevant proteinogenic underivatized amino acids was achieved by ion-pairing reversed-phase Ultra-high Performance Liquid chromatography-Electrospray Ionisation-Quadrupole Time of Flight-Mass Spectrometry (UPLc-ESI-QTOF-MS). Excellent linearity in the response was obtained for all amino acids with correlation coefficients > 0.9921. This method was successfully applied to natural hydrothermal fluid samples from ultramafic-hosted vents of the Mid-Atlantic Ridge region. Results are consistent with the only 2 other studies published on ultramafic-hosted vents and complete the few available data.

Digestate, the product obtained after anaerobic digestion of organic waste for biogas production, is rich in plant nutrients and might be used to fertilize crops. Wheat （Triticum spp. L.） was fertilized with digestate, urea, or left unfertilized and cultivated in the greenhouse for 120 d. Emissions of greenhouse gasses （carbon dioxide （cO2）, methane （cH4）, and nitrous oxide （N20）） were monitored and plant growth characteristics were determined at harvest. The digestate was characterized for heavy metals, pathogens, and c and N mineralization potential in an aerobic incubation experiment. No Salmonella spp., Shigella spp., or viable eggs of helminths were detected in the digested pig slurry, but the number of faecal coliforms was as high as 3.6 ~ 104 colony-forming units （cFU） g-1 dry digestate. The concentrations of heavy metals did not surpass the upper limits established by US Environmental Protection Agency （EPA）. After 28 d, 17% of the organic c （436 g kg-1 dry digestate） and 8% of the organic N （6.92 g kg-1 dry digestate） were mineralized. Emissions of cO2 and cH4 were not significantly affected by fertilization in the wheat-cultivated soil, but digestate significantly increased the cumulative N20 emission by 5 times compared to the urea-amended soil and 63 times compared to the uncultivated unfertilized soil. It could be concluded that digestate was nutrient rich and low in heavy metals and pathogens, and did not affect emissions of cH4 and cO2 when applied to a soil cultivated with wheat, but increased emission of N20. Going beyond 1000000 resolved points in a Brillouin distributed fiber sensor: theoretical analysis and experimental demonstration

Distributed fiber sensing possesses the unique ability to measure the distributed profile of an environmental quantity along many tens of kilometers with spatial resolutions in the meter or even centimeter *** feature enables distributed sensors to provide a large number of resolved points using a single optical ***,in current systems,this number has remained constrained to a few hundreds of thousands due to the finite signal-to-noise ratio(SNR)of the measurements,which imposes significant challenges in the development of more performing ***,we propose and experimentally demonstrate an ultimately optimized distributed fiber sensor capable of resolving 2100000 independent points,which corresponds to a one-orderof-magnitude improvement compared to the *** a Brillouin distributed fiber sensor based on phase-modulation correlation-domain analysis combined with temporal gating of the pump and time-domain acquisition,a spatial resolution of 8.3 mm is demonstrated over a distance of 17.5 *** sensor design addresses the most relevant factors impacting the SNR and the performance of medium-to-long range sensors as well as of sub-meter spatial resolution *** step record in the number of resolved points could be reached due to two theoretical models proposed and experimentally validated in this study:one model describes the spatial resolution of the system and its relation with the sampling interval,and the other describes the amplitude response of the sensor,providing an accurate estimation of the SNR of the measurements.

Objective: This study aims to evaluate the vasodilatory effect of chenopodium ambrosioides on the isolated rat aorta, and to explore its mechanism of ***: The vasorelaxant effect and the mode of action of various extracts from the leaves of c. ambrosioides were evaluated on thoracic aortic rings isolated from Wistar rats. In addition, ethyl acetate and methanol fractions were analyzed, using thin-layer chromatography and high-performance liquid chromatography techniques, for their polyphenolic ***: The various active extracts of c. ambrosioides at four concentrations(10^(-3), 10^(-2), 10^(-1) and 1 mg/mL) relaxed the contraction elicited by phenylephrine, in a concentration-dependent *** effect seems to be endothelium-dependent, since the vasodilatory effect was entirely absent in denuded aortic rings. The vasorelaxant effect of the methanol fraction(MF) of c. ambrosioides at 1 mg/mL was also inhibited by atropine and tetraethylammonium. This effect remained unchanged by Nx-nitro-L-arginine methyl ester hydrochloride and glibenclamide. The preliminary phytochemical analysis showed that the leaves of c. ambrosioides are rich in phenolic and flavonoid ***: These results suggest that the MF of c. ambrosioides produces an endothelium-dependent relaxation of the isolated rat aorta, which is thought to be mediated mainly through stimulation of the muscarinic receptors, and probably involving the opening of ca^(2+)-activated potassium channels.

Introduction: Perinatal mortality linked to fetal anemia of red cell alloimmunization in côte d’Ivoire as in many developing countries can be explained by a lack of knowledge of Rhesus D feto-maternal alloimmunization, hence the obvious importance of carrying out a study on the problem of prevention of alloimmunization in rhesus negative births. This study takes stock of the management of this pathology in a reference hospital. Results and Discussion: Rh-negative mothers account for 6% of births, 42.7% (70/164) of women had a history of risk, about 61% of women had not had proper prophylaxis during previous pregnancies;and as many had not had follow-up of the coombs test during the current pregnancy;only 4.9% of patients had systematic prophylaxis with anti D serum at 28 weeks of pregnancy. This low rate of prevention is related to the financial difficulties of the patients, but also to a lack of knowledge of the pathology by the nursing staff. conclusion: Good management of rhesus negative women during their pregnancy allows their incompatible child to benefit from all current treatments ensuring a healthy birth. It is therefore important for medical personnel to know how to deal with this rare disease in a small proportion of pregnant women.
