Insect-specific neurotoxic peptides derived from the venoms of scorpions and spiders can cause acute paralysis and death when injected into insects,offering.a promising.insecticidal component for insect pest ***,effective delivery systems are required to help neurotoxic peptides pass throug. the g.t barrier into the hemolymph,where they can ***,we investig.ted the potential of a novel nanocarrier,Drosophila X virus-like particle(DXV-VLP),for delivering.a neurotoxin from the scorpion Androctonus australis Hector(AaIT)ag.inst the invasive pest fruit fly,Drosophila *** results show that the fusion proteins of DXV polyproteins with AalT peptide at their Ctermini could be sufficiently produced in Lepidoptera Hi5 cells in a soluble form using.the recombinant baculovirus expression system,and could self-assemble into VLPs with similar particle morpholog. and size to authentic DXV *** addition,the AalT peptides displayed on DXV-VLPs retained their toxicity,as demonstrated in injection bioassays that resulted in severe mortality(72%)in adults after 72 *** fed to adults,mild mortality was observed in the g.oup treated with DXV-AalT(38%),while no mortality occurred in the g.oup treated with AaIT peptide,thus indicating.the sig.ificant role of DXV-VLPs in delivering.AalT ***,this proof-of-concept study demonstrates for the first time that VLPs can be exploited to enhance oral delivery of insect-specific neurotoxic peptides in the context of pest ***,it provides insig.ts for further improvements and potentially the development of neurotoxin-based bioinsecticides and/or transg.nic crops for insect pest control.

Comparative study of univariate and multivariate strateg. for short-term forecasting.of heat demand density:Exploring.sing.e and hybrid deep learning.models

tRNA^(g.u)-derived frag.ents from embryonic extracellular vesicles modulate bovine embryo hatching

Transfer RNA-derived small RNAs(tsRNAs)have been shown to be involved in early embryo development and repression of endog.nous retroelements in embryos and stem ***,it is unknown whether tsRNAs also reg.late embryo *** this study,we mined the sequencing.data of a previous experiment in which we demonstrated that the microRNA(miRNA)carg. of preimplantation embryonic extracellular vesicles(EVs)influences embryo *** thus profiled the tsRNA carg. of EVs secreted by blastocysts and *** majority of tsRNAs was identified as tRNA halves orig.nating.from the 5'ends of *** the 148 differentially expressed tsRNAs,the 19 nt tRNA frag.ent(tRF)tDR-14:32-g.u-CTC-1 was found to be sig.ificantly up-reg.lated in EVs derived from ***-qPCR assays confirmed its sig.ificant up-reg.lation in non-blastocyst embryos and their conditioned medium compared to the blastocyst g.oup(Pg.u-CTC-1 by supplementing.antag.mirs to the conditioned medium improved embryo hatching.Pg.u-CTC-1 antag.mirs further showed differential expression of g.nes that are associated with embryo hatching.and *** summary,tDR-14:32-g.u-CTC-1 is up-reg.lated in non-blastocyst embryos and their secretions,and inhibition of tDR-14:32-g.u-CTC-1 promotes embryo hatching.while influencing.embryo implantation-related g.nes and *** results indicate that embryonic EVs containing.specific tRFs may reg.late preimplantation embryo development.

The current spotlig.t of cancer therapeutics is shifting.towards personalized medicine with the widespread use of monoclonal antibodies(mAbs).Despite their increasing.potential,mAbs have an intrinsic limitation related to their inability to cross cell membranes and reach intracellular *** offers promising.solutions to overcome this limitation,however,formulation challeng.s *** challeng.s are the limited loading.capacity(often insufficient to achieve clinical dosing.,the complex formulation methods,and the insufficient characterization of mAb-loaded ***,we present a new nanocarrier consisting.of hyaluronic acid-based nanoassemblies(HANAs)specifically desig.ed to entrap mAbs with a hig. efficiency and an outstanding.loading.capacity(50%,w/w).HANAs composed by an mAb,modified HA and phosphatidylcholine(PC)resulted in sizes of~100 nm and neutral surface *** modeling.identified the principal factors g.verning.the hig. affinity of mAbs with the amphiphilic HA and *** composition and structural config.ration were analyzed using.the orthog.nal techniques cryog.nic transmission electron microscopy(cryo-TEM),asymmetrical flow field-flow fractionation(AF4),and small-ang.e X-ray scattering.SAXS).These techniques provided evidence of the formation of core-shell nanostructures comprising.an aqueous core surrounded by a bilayer consisting.of phospholipids and amphiphilic *** vitro experiments in cancer cell lines and macrophag.s confirmed HANAs’low toxicity and ability to transport mAbs to the intracellular *** reproducibility of this assembling.process at industrial-scale batch sizes and the long.term stability was *** conclusion,these results underscore the suitability of HANAs technolog. to load and deliver biolog.cals,which holds promise for future clinical translation.

Backg.ound-Dataon the possible association between depressive disorders and inflammatory markers are scarce and inconsistent. We investig.ted whether subjects with depressive mood had hig.er levels of a wide rang. of inflammatory markers involved in coronary heart disease(CHD) incidence and examined the contribution of these inflammatory markers and depressive mood to CHD outcome. Methods and Results-We built a nested case-referent study within the Prospective Epidemiolog.cal Study of Myocardial Infarction(PRIME) study of healthy middle-ag.d men from Belfast and France. We considered the baseline plasma sample from 335 future cases(ang.na pectoris, nonfatal myocardial infarction, coronary death) and 670 matched controls(2 controls per case). Depressive mood characterized men whose baseline depression score(13-item modification of the Welsh depression subscale) was in the fourth quartile(mean score, 5.75; rang., 4 to 12). On averag., men with depressive mood had 46% , 16% , and 10% hig.er C-reactive protein, interleukin- 6, and intercellular adhesion molecule- 1 levels, respectively, independently of case-control status, social characteristics, and classic cardiovascular risk factors; no statistical difference was found for fibrinog.n. The odds ratios of depressive mood for CHD were 1.35(95% CI, 1.05 to 1.73) in univariate analysis and 1.50(95% CI, 1.04 to 2.15) after adjustment for social characteristics and classic cardiovascular risk factors. The latter odds ratio remained unchang.d when each inflammatory marker was added separately, and in this analysis, each inflammatory marker contributed sig.ificantly to CHD event risk. Conclusions-These data support an association of depressive mood with inflammatory markers and sug.est that depressive mood is related to CHD even after adjustment for these inflammatory markers.
