We present and compare twoﬂexible and effective methodologies to predict disturbance zones ahead of underground tunnels by using elastic full-waveform *** methodology uses a linearized,iterative approach based on misﬁt gradients computed with the adjoint method while the other uses iterative,gradient-free unscented Kalmanﬁltering in conjunction with a level-set *** the former does not involve a priori assumptions on the distribution of elastic properties ahead of the tunnel,the latter intro-duces a massive reduction in the number of explicit model parameters to be inverted for by focusing on the geometric form of potential disturbances and their average elastic *** imaging methodologies are validated through successful reconstructions of simple *** an application,we consider an elastic multiple disturbance *** using identical synthetic time-domain seismo-grams as test data,we obtain satisfactory,albeit different,reconstruction results from the two inversion *** computa-tional costs of both approaches are of the same order of magnitude,with the gradient-based approach showing a slight *** model parameter space reduction approach compensates for this by additionally providing a posteriori estimates of model parameter uncertainty.

traditional Chinese medicine,such as tripterygium wilfordii and Paeonia lactiflora,has potential values in treating systemic sclerosis(SSc)and other autoimmune diseases,while their toxic side effect elimination and precise tropical drug delivery are still ***,we present multiple traditional Chinese medicine integrated photoresponsive black phosphorus(BP)microneedles(MNs)with the desired features for the SSc *** employing a template-assisted layer-by-layer curing method,such MNs with triptolide(tP)/paeoniflorin(Pae)needle tips and BP-hydrogel needle bottoms could be well *** combined administration of tP and Pae can not only provide anti-inflammatory,detoxification,and immunomodulatory effects to treat skin lesions in the early stage of SSc but also remarkably reduce the toxicity of single drug ***,the additive BPs possess good biocompatibility and near-infrared(NIR)responsiveness,imparting the MN photothermal-controlled drug release *** on these features,we have demonstrated that the traditional Chinese medicine integrated responsive MNs could effectively improve skin fibrosis and telangiectasia,reduce collagen deposition,and reduce epidermal thickness in the SSc mouse *** results indicated that the proposed Chinese medicine integrated responsive MNs had enormous potential in clinical therapy of SSc and other diseases.

Capsular contracture is a prevalent and severe complication that affects the post-operative outcomes of patients who receive silicone breast *** present,prosthesis replacement is the major treatment for capsular contracture after both breast augmentation procedures and breast reconstruction following breast cancer ***,the mecha-nism(s)underlying breast capsular contracture remains *** study aimed to identify the biological features of breast capsular contracture and reveal the potential underlying mechanism using RNA *** tissues from 12 female patients(15 breast capsules)were divided into low capsular contracture(LCC)and high capsular contracture(HCC)groups based on the Baker ***,41 lipid metabolism-related genes were identified through enrichment analysis,and three of these genes were identified as candidate genes by SVM-RFE and LASSO *** then compared the proportions of the 22 types of im-mune cells between the LCC and HCC groups using a CIBERSORt analysis and explored the Cor-relation between the candidate hub features and immune ***,PRKAR2B was positively correlated with the differentially clustered immune cells,which were M1 macro-phages and follicular helper t cells(area under the ROC=0.786).In addition,the expression of PRKAR2B at the mRNA or protein level was lower in the HCC group than in the LCC *** molecular mechanisms were identified based on the expression levels in the high and low PRKAR2B *** findings indicate that PRKAR2B is a novel diagnostic biomarker for breast capsular contracture and might also influence the grade and progression of capsularcontracture.

A small fraction of NOx（ts in CO2feedstock（e.g. exhausted gas）, which can significantly reduce the efficiency of CO2electroreduction by ～30%. Hence, electrochemical denitrification is the precondition of CO2electroreduction. the pH effect is a key factor, and can be used to tune the selectivity between N2and N2O production in electrochemical denitrification. However, there has been much controversy for many years about the origin of pH dependence in electrocatalysis. to this end, we present a new scheme to accurately model the pH dependence of the electrochemical mechanism. An extremely small pH variation from pH 12.7 to pH 14 can be accurately reproduced for N2O production. More importantly,the obviously different pH dependence of N2production, compared to N2O, can be attributed to a cascade path. In other words, the N2was produced from the secondary conversion of the as-produced N2O molecule（the major product）, instead of the original reactant NO. this is further supported by more than 35 experiments over varying catalysts（Fe, Ni, Pd, Cu, Co, Pt and Ag）, partial pressures（20%, 50% and100%） and potentials（from -0.2 to 0.2 V vs. reversible hydrogen electrode）. All in all, the insights herein overturn long-lasting views in the field of NO electroreduction and suggest that rational design should steer away from catalyst engineering toward reactor optimization.

the photovoltaic/thermal(PV/t)system is a promising option for countering energy *** improve the performance of PV/t systems,compound parabolic concentrators(CPCs)and phase-change materials(PCMs)were jointly applied to construct a concentrating photovoltaic/thermal system integrated with phase-change materials(PV/t-CPCM).An open-air environment is used to analyze the effects of different parameters and the intermittent operation strategy on the system *** results indicate that the short-circuit current and open-circuit voltage are positively correlated with the solar irradiance,but the open-circuit voltage is negatively correlated with the temperature of the PV *** the solar irradiance is 500 W⋅m^(−2) and the temperature of the PV modules is 27.5℃,the short-circuit current and open-circuit voltage are 1.0 A and 44.5 V,*** solar irradiance results in higher thermal power,whereas the thermal efficiency is under lower solar irradiance(136.2-167.1 W⋅m^(−2) is twice under higher solar irradiance(272.3-455.7 W⋅m^(−2))).In addition,a higher mass flow rate corresponds to a better cooling effect and greater pump energy *** the mass flow rate increases from 0.01 to 0.02 kg⋅s^(-1),the temperature difference between the inlet and outlet decreases by 1.8℃,and the primary energy-saving efficiency decreases by 0.53%.the intermittent operation of a water pump can reduce the energy consumption of the system,and the combination of liquid cooling with PCMs provides better thermal regulation and energy-saving effects under various conditions.

Glypican-3 is a protein encoded by the Glypican-3 gene located on human X chromosome (Xq26), composed of two subunits, a 40 kDa N-terminal subunit, and a 30 kDa C-terminal subunit. Glypican-3 is a currently potential target molecule for liver cancer treatments because of its over-expression and growth effects on hepatocellular carcinoma (HCC). this study examined the expression and purification of a C-terminal subunit of Glypican-3 protein (GPC3-Csub) due to its application in both diagnosis and therapy for hepatocellular carcinoma. the gene encoding for GPC3-Csub was successfully cloned into plasmid pEt28a fused with an affinity tag composed of six consecutive histidine residues (His-tag). Recombinant protein GPC3-Csub was expressed in Escherichia coli BL21 (DE3) in the condition of adding 3% ethanol with IPtG induction. GPC3-Csub was extracted using repeated freeze-thaw cycles with lysozyme, and inclusion bodies were solubilized by 8M Urea, SDS 10% in pH 12. His-tag fused GPC3-Csub proteins allowed it to be purified by affinity chromatography method using the Nickel-nitrilotriacetic acid (Ni-NtA) column. High expression of GPC3-Csub was confirmed by Coomassie staining and western-blot. GPC3-Csub could be isolated with a Ni-NtA column and have a purity of about 90%.

Background In Viet Nam, tuberculosis (tB) represents a devastating life-event with an exorbitant price tag, partly due to lost income from daily directly observed therapy in public sector care. thus, persons with tB may seek care in the private sector for its flexibility, convenience, and privacy. Our study aimed to measure income changes, costs and catastrophic cost incurrence among tB-affected households in the public and private *** Between October 2020 and March 2022, we conducted 110 longitudinal patient cost interviews, among 50 patients privately treated for tB and 60 tB patients treated by the National tB Program (NtP) in Ha Noi, Hai Phong and Ho Chi Minh City, Viet Nam. Using a local adaptation of the WHO tB patient cost survey tool, participants were interviewed during the intensive phase, continuation phase and post-treatment. We compared income levels, direct and indirect treatment costs, catastrophic costs using Wilcoxon rank-sum and chi-squared tests and associated risk factors between the two cohorts using multivariate *** the pre-treatment median monthly household income was significantly higher in the private sector versus NtP cohort (USD 868 vs USD 578;P=0.010). However, private sector treatment was also significantly costlier (USD 2075 vs USD 1313;P=0.005), driven by direct medical costs which were 4.6 times higher than costs reported by NtP participants (USD 754 vs USD 164;Pthis resulted in no significant difference in catastrophic costs between the two cohorts (Private: 55% vs NtP: 52%;P=0.675). Factors associated with catastrophic cost included being a single-person household [adjusted odds ratio [(aOR=13.71; 95% confidence interval (CI): 1.36–138.14;P=0.026], unemployment during treatment (aOR=10.86; 95%CI: 2.64–44.60;PtB-related stigma (aOR=37.90; 95%CI: 1.72–831.73;P=0.021).Conclusions Persons with tB in Viet Nam face similarly high risk of catastrophic costs whether treated in the
