Flow cytometric assessment of the effects of chlorine, chloramine, and UV on bacteria by using nucleic acid stains and 5-cyano-2,3-ditolyltetrazolium chloride

Flow cytometry （FCM） has been widely used in multi-parametric assessment of cells in various research fields, especially in environmental sciences. This study detected the metabolic activity of Escherichia coli and Staphylococcus aureus by using an FCM method based on 5-cyano-2,3-ditolyltetrazolium chloride （CTC）; the accuracy of this method was enhanced by adding SYTO 9 and 10% R2A broth. The disinfection effects of chlorine, chloramine, and UV were subsequently evaluated by FCM methods. Chlorine demonstrated stronger and faster destructive effects on cytomembrane than chloramine, and nucleic acids decomposed afterwards. The metabolic activity of the bacteria persisted after the cytomembmnewas damaged as detected using CTC. Low-pressure （LP） UV or medium-oressure （MP） UV treatments exerted no significant effects on membrane permeability. The metabolic activityof the bacteria decreased with increasing UV dosage, and MP-UV was a stronger inhibitor of metabolic activity than LP-UV. Furthermore, the membrane of Gram-positive S. aureus was more resistant to chlorine/chloramine than that of Gram-negative E. coil In addition, S. aureus showed higher resistance to UV irradiation than E. coli.

Comparison of membrane fouling in ultrafiltration of down-flow and up-flow biological activated carbon effluents

Fouling during ultrafiltration of down- and up-flow biological activated carbon effluents was investigated to determine the roles of polysaccharides, proteins, and inorganic particles in ultrafiltration membrane fouling. During ultrafiltration of down- flow biological activated carbon effluent, the trans-membrane pressure was £26 kPa and the permeate flux was steady at 46.7 L∙m–2∙h–1. However, during ultrafiltration of up-flow biological activated carbon effluent, the highest trans-membrane pressure was almost 40 kPa and the permeate flux continuously decreased to 30 L∙m–2∙h–1. At the end of the filtration period, the normalized membrane fluxes were 0.88 and 0.62 for down- and up-flow biological activated carbon effluents, respectively. The membrane removed the turbidity and polysaccharides content by 47.4% and 30.2% in down- flow biological activated effluent and 82.5% and 22.4% in up-flow biological activated carbon effluent, respectively, but retained few proteins. The retention of polysaccharides was higher on the membrane that filtered the down- flow biological activated effluent compared with that on the membrane that filtered the up-flow biological activated carbon effluent. The polysaccharides on the membranes fouled by up-flow biological activated carbon and down- flow biological activated effluents were spread continuously and clustered, respectively. These demonstrated that the up-flow biological activated carbon effluent fouled the membrane faster. Membrane fouling was associated with a portion of the polysaccharides (not the proteins) and inorganic particles in the feed water. When there was little difference in the polysaccharide concentrations between the feed waters, the fouling extent was exacerbated more by inorganic particles than by polysaccharides.

Cooling rate is a key factor that can drastically affect the phase transformation and thermal stress of duplex stainless steels. Therefore, in this research, different sand moulds were used to explore the influence of cooling rate on the solidification of the 2304 duplex stainless steel (DSS). The macro and micro structures of the 2304 DSS were investigated. Small equiaxed grains are obtained in chromite sand mould sample with a lower pouring temperature and a higher cooling rate, whereas coarse columnar and equiaxed grains are found in silica sand and refractory powder mould samples. The size of austenite phase is significantly increased with decreasing cooling rate, while the ferrite phase content ranging from 51.6% to 53.9% does not change obviously. In addition, the linear contraction of the 2304 DSS decreases from 2.34% to 1.09% when the mean cooling rate above 1,173 K increases from 0.99 K·s-1 to 3.66 K·s-1.

Traditional asphalt rejuvenators,like aromatic oil(ao),are known to be effective in improving the low-temperature properties and fatigue performances of aged SBS(styrene-butadiene-styrene)modified asphalt(SBSMA)binders and ***,these rejuvenators inevitably compromise their high-temperature properties and deformation resistances because they dilute asphalt binder but do not fix the damaged structures of aged *** this study,a highly-active chemical called polymerized 4,4-diphenylmethane diisocyanate(PMDI)was used to assist the traditional ao asphalt *** physical and rheological characteristics of rejuvenated SBSMA binders and the moisture-induced damage and rut deformation performances of corresponding mixtures were comparatively *** results showed that the increasing proportion of ao compromises the hightemperature property and hardness of aged SBSMA binder,and an appropriate amount of PMDI works to compensate such losses;3%rejuvenator at mass ratio of ao:PMDI=70:30 can have a rejuvenated SBSMA binder with a high-temperature performance similar to that of fresh binder,approximately at 71.4°C;the use of ao can help reduce the viscosity of PMDI rejuvenated SBSMA binder for improving its workability;PMDI can help improve the resistance of ao rejuvenated SBSMA binder to deformation,especially at elevated temperatures,through its chemical reactions with aged SBS;moisture induction can enhance the resistance to damage of rejuvenated mixtures containing ao/PMDI or only PMDI;and the rejuvenator with a mass ratio of ao:PMDI=70:30 can lead the rejuvenated mixture to meet the application requirement,with a rut depth of only 2.973 mm,although more PMDI can result in a higher resistance of rejuvenated mixtures to high-temperature deformation.

Conventional repairing methods for asphalt pavement have some inconveniences,such as insufficient strength,and are typically *** address these issues,this study proposes a new technological method to design and prepare a high-performance assembled asphalt concrete block for fast repair of the potholes.A series of composite modified asphalt binders with 10%crumb rubber(CR)and different dosages(0%,1%,3%,5%)of polyurethane(PU)are examined to determine the optimized ***,the corresponding asphalt mixtures are prepared for further comparison and assessment of engineering properties,such as moistureinduced damage,high-temperature deformation,and low-temperature cracking *** test results show that PU can significantly improve the high-temperature performance and hardness of(crumb rubber modified asphalt)CRMA binder;3%PU contributes allowing the resistance of CRMA mixture to moisture-induced damage at higher levels,particularly under water whole immersion;as 3%PU is added,the high-temperature rutting deformation resistance of the CRMA mixture increases significantly,and the low-temperature anti-cracking properties are also improved ***,the innovatively designed high-quality assembled fast-repairing asphalt concrete block is recommended as an appropriate option for highway maintenance.

AIM To investigate the role of apolipoprotein E (apoE) polymorphism in the lithogenesis of gallstone and the hereditary pathogenesis of the *** Polymerase chain reaction (PCR)was used to study apoE phenotypes and allelefrequencies in patients with gallstones and control, and the fasting serum lipids of subjectswere also measured by enzymatic *** The levels of triglyceride (TG) andvery low density lipoprotein cholesterol (VLDLC) were much higher in Ez/, patients than that inE,/, control. E,/, patients were accompanied withremarkably low levels of high density lipoproteincholesterol (HDLC) and its subforms. But in E,/#patients there were only slight changes in levelsof VLDLC and low density lipoprotein cholesterol (LDL--C).CONCluSION Different apoE phenotype patientswith gallstones have different cheracteristics ofdyslipidemia and the average level of serum lipids in patients with gallstones are higher thansubjects without gallstones in the same apoEgene phenotype. EZ allele is possibly one of thedangerous factors in the lithogenesis of cholecystolithiasis.

Reconstruction of Place Spirit in the Street Protection of Hekou Town of Qianshan

Place spirit originates from ancient Rome, and emphasizes whole fusion of objective material environment and subjective consciousness. The introduction of "place spirit" concept in protection work of old town is conducive to further understanding complex relationship between reconstruction of objective material environment and reappearance of traditional culture connotation. The street of Hekou Town of Qianshan is historical one, and its protection and planning are conducting. The street is full of rich marketplace breath and has certain feature. In this paper, started from protection status quo of Hekou Town, we preliminarily discuss how to effectively pass historical information in the protection process and maintain place spirit of old town in modern living environment.

Android is the mobile operating system most frequently targeted by malware in the smartphone ecosystem,with a market share significantly higher than its competitors and a much larger total number of *** of malware before being published on official or unofficial application markets is critically important due to the typical end users'widespread security *** this paper,a novel feature selection method is proposed along with an Android malware detection *** feature selection method proposed in this study makes use of permissions,API calls,and strings as features,which are statically extractable from the Android executables(APK files)and it can be used in a machine learning process with different algorithms to detect malware on the Android platform.A novel document frequency-based approach,namely Delta IDF,was designed and implemented for feature *** IDF was tested upon three universal benchmark datasets that contain Android malware samples and highly promising results were obtained by using several binary classification algorithms.
