Contribution of plant traits to the explanation of temporal variations in carbon and water fluxes in semiarid grassland patches

Aims Accurate prediction of spatiotemporal variations in carbon and water fluxes of heterogeneous landscape is critical to comprehensively address the effects of climate change and vegetation dynamics on landscape and regional carbon and water *** A field study was conducted to characterize the seasonal variations in gas fluxes and explore their relationships with abiotic and biotic factors in a small grassland *** carbon and water fluxes including net ecosystem exchange,gross ecosystem productivity,ecosystem respiration and evapotranspiration(ET)were measured for three types of grassland patches over a growing season using the closed chamber *** key plant trait variables were measured,based on which community weighted mean(CWM)and functional variance(FDvar)were *** Findings The results showed that the temporal variations in the carbon and water fluxes were regulated by meteorological,soil and community functional *** of the CWM and FDvar of plant trait measures greatly improved the degree of explanation of the predict *** leaf area and leafδ^(13)C content(Lδ^(13)C)were the most important trait variables in affecting the variations of the gas *** indices had greater importance than FDvar indices in predicting the variation of the C fluxes but FDvar indices were more important for ET than C *** findings demonstrated that mass ratio hypothesis and the complementary effects hypothesis are not mutually exclusive but have different relative importance for different ecosystem *** functional traits played important roles in predicting the spatiotemporal variations of carbon and water fluxes in semiarid grassland.

Lysine acetylation,a reversible and dynamic post-translational modification,plays pivotal roles in regulating many biological processes in ***,the information of lysine acetylation during flower development of woody angiosperm remains ***,we identified a total of 667 lysine acetylation sites in 467 proteins in flower buds of Catalpa *** acetylated proteins were mainly involved in the pathways of carbon metabolism,glycometabolism and oxidative *** functional enrichment analysis,61 and 13 acetylated proteins were involved in the glycometabolism and oxidative phosphorylation pathways,*** suggests that lysine acetylated proteins play critical roles in the energy metabolism ***,the protein–protein interactions were also highly enriched in the carbon metabolic *** analysis of lysine acetylome and transcriptome showed that transcript level changes of seven key lysine-acetylated proteins/genes involved in energy metabolism were significant different during floral ***,compared with the normal flowering variety,the expression levels of phosphoribulokinase,phosphoenolpyruvate carboxykinase,ATP synthase and pyruvate dehydrogenase complex genes were significantly up-regulated at the stages of vegetative and transition buds during early flowering of *** data represent the first lysine acetylome in the *** flowers,and can serve as an essential basis for exploring the function of lysine acetylated proteins in the flower bud development of plants.

Zinc finger protein A20 protects rats against chronic liver allograft dysfunction

AIM： To investigate the effect of zinc finger protein A20 on chronic liver allograft dysfunction in rats. METHODS： AIIogeneic liver transplantation from DA rats to Lewis rats was performed. Chronic liver allograft dysfunction was induced in the rats by administering low-dose tacrolimus at postoperative day （POD） 5. Hepatic overexpression of A20 was achieved by recom- binant adenovirus （rAd.）-mediated gene transfer ad- ministered intravenously every 10 d starting from POD 10. The recipient rats were injected with physiologi- cal saline, rAdEasy-A20 （1 × 109 pfu/30 g weight） or rAdEasy （1 × 109 pfu/30 g weight） every 10 d through the tail vein for 3 mo starting from POD 10. Liver tissue samples were harvested on POD 30 and POD 60. RESULTS： Liver-transplanted rats treated with only tacrolimus showed chronic allograft dysfunction with severe hepatic fibrosis. A20 overexpression ameliorated the effects on liver function, attenuated liver allograft fibrosis and prolonged the survival of the recipient rats. Treatment with A20 suppressed hepatic protein pro- duction of tumor growth factor （TGF）-β1, interleukin- 113, caspase-8, CD40, CD40L, intercellular adhesion molecule-i, vascular cell adhesion molecule-1 and E-selectin. A20 treatment suppressed liver cell apopto- sis and inhibited nuclear factor-KB activation of Kupffer cells （KCs）, liver sinusoidal endothelial cells （LSECs） and hepatic stellate cells （HSCs）, and it subsequently decreased cytokine mRNA expression in KCs and LSECs and reduced the production of TGF-β1 in HSCs. CONCluSION： A20 might prevent chronic liver allogra- ft dysfunction by re-establishing functional homeostasis of KCs, LSECs and HSCs.

目的系统评价先天性心脏病(congenital heart disease,CHD)青少年向成人过渡治疗及护理体验的质性研究,为制订CHD青少年向成人过渡治疗及护理管理方案提供参考。方法计算机检索PubMed、Embase、Web of Science、Cochrane Library、EBSCO、CINAHL、中国知网、万方数据库、维普数据库、中国生物医学文献数据库中关于CHD青少年向成人过渡治疗及护理体验的质性研究,检索时限为建库至2023年4月。使用澳大利亚乔安娜布里格斯研究所循证卫生保健中心质性研究质量评价标准(2016)评价文献质量,采用Meta整合方法对结果进行整合。结果共纳入9篇文献,提炼出49个研究结果,最终归纳为11个类别,综合为4个整合结果。①态度复杂,依恋和期待并存:依恋儿科医疗机构;期待过渡到成人医疗机构。②面临多种挑战:缺乏充分的向成人过渡的准备;父母退出疾病管理者角色;儿科、成人医疗机构的服务存在较大差异。③期望获得多种支持:期望获得医护人员全面的健康教育;期待医疗机构设立咨询门诊、实现病情交接;期望获得父母的陪伴和支持;期望获得同伴的理解和帮助。④感知到向成人过渡的益处:疾病管理能力增强;角色改变和个人成长。结论CHD青少年向成人过渡治疗及护理体验复杂,医护人员应关注其感受,鼓励其积极应对挑战,提供充足的信息,并联合父母及同伴给予支持,以促进青少年向成人顺利过渡。

Expression of Soluble Vascular Endothelial Growth Factor Receptor-2 and Its Effect on Proliferation of Vascular Endothelial Cells

Vascular endothelial growth factors（VEGFs） respectively bind to each of three receptor tyrosine kinases（RTKs）,known as Flt-1,KDR and *** VEGFs and their respective families of receptor tyrosine kinases（VEGFRs） are critical proteins which can regulate vascular development during angiogenesis,we decided to explore the inhibitory effects of soluble kinase insert domain-containing receptor（sKDR） on endothelial cells and *** RNA was extracted from human umbilical vein endothelial cells（HUVEC）,and cDNA of extracellular domains 1―4 was amplified and recombined with pQE40 *** being expressed,affinity purified,renatured and analyzed by Western blot,the sKDR was assayed for its effects on endothelial cells by [3-（4,5-dimethylthiazol-2-yl）2,5-diphenyltetrazolium bromide]（MTT）,and on angiogenesis by chick chorioallantoic membrane（CAM） *** cDNA of 1150 bp was obtained via real-time polymerase chain reaction（RT-PCR）,and sKDR was expressed by pQE40 procaryotic expression system,purified by sodium dodecyl sulfate-polyacrylamide gel electrophoresis（SDS-PAGE） analysis with only one band and proved by Western *** assay demonstrateds that sKDR could inhibit the VEGF-stimulated HUVEC from proliferation,and CAM experiment showed sKDR could block the VEGF-induced *** has the biological activity to bind with VEGF ligands and is a potential target for tumor anti-angiogenesis therapy.

Inhibition of cellular proliferation and induction of apoptosis in human esophageal carcinoma cell lines by extracts of Dioscorea bulbifera L and Chinese Angelica

Objective： To investigate the antiproliferative and apoptogenic activities of polysaccharides extracted from Dioscorea bulbifera L （DBP） and Chinese Angelica （CAP） and a 3：2 mixture of these compounds （DCCP） on two esophageal carcinoma cell lines, EC9706 and Ecal09. Methods： A MTT assay was used to detect the effects of DBP （10 μg/ml and 100 μg/ml）, DCCP（17 μg/ml and 170 μg/ ml） and CAP （10 μg/ml and 100 μg/ml） on the proliferation of EC9706 and Eca109 cells. DNA content analysis by flow cytometry was used to determine the cell cycle distribution, and Annexin V-FITC/PI stained fluorescence-activated cell sorter （FACS） was used to detect the apotosis rate of treated cells. Western blots were used to examine protein levels. Results： DBP and DCCP strongly inhibited the proliferation and viability of both the EC9706 and Eca109 cells. CAP enhanced the effects of DBP. DCCP primarily arrested the EC9706 and Eca109 cells at the G1 phase of the cell cycle. DCCP induced apoptosis in both esophageal carcinoma cell lines, and reduced the expression of plKBa and Bcl-2 proteins. Conclusion： DCCP triggered apoptosis in esophageal carcinoma cells by inhibiting the NF-KB signaling pathway.
