Liver-specific glucocorticoid action in alcoholic liver disease:Study of glucocorticoid receptor knockout and knockin mice

Background and aim:Glucocorticoids are the only first-line drugs for severe alcoholic hepatitis(AH),with limited efficacy and various side effects on extrahepatic ***-targeting glucocorticoid therapy may have multiple advantages over systemic glucocorticoid for *** aim of this study was to determine the role of hepatocellular glucocorticoid receptor(GR)in alcohol-associated steatosis(AS)and AH.Materials and methods:AS was induced by a high-fat diet plus binge alcohol in adult male and female mice with liver-specific knockout(LKO)and heterozygote of *** was induced by chronic-plus-binge in middle-aged male mice with liver-specific knockin of *** in hepatic mRNA and protein expression were determined by quantitative real-time polymerase chain reaction and Western blot.Results:GR-LKO aggravated steatosis and decreased hepatic expression and circulating levels of albumin in both genders of AS mice but only increased markers of liver injury in male AS *** steatosis in GR-LKO mice was associated with induction of lipogenic genes and down-regulation of bile acid synthetic *** protein levels of GR,hepatocyte nuclear factor 4 alpha,and phosphorylated signal transducer and activator of transcription 3 were gene-dosage-dependently decreased,whereas that of lipogenic ATP citrate lyase was increased in male GR heterozygote and LKO ***,hepatic expression of estrogen receptor alpha(ERα)was induced,and the essential estrogen-inactivating enzyme sulfotransferase 1e1 was gene-dosage-dependently down-regulated in GR heterozygote and knockout AS mice,which was associated with induction of ERα-target ***-specific knockin of GR protected against liver injury and steatohepatitis in middle-aged AH mice.Conclusions:Hepatic GR deficiency plays a crucial role in the pathogenesis of AS induced by high-fat diet plus binge,and liver-specific overexpression/activation of GR protects against chronic-plus-binge-induced AH in middl

Extensive Resetting of Feldspar Pb Isotopic Composition in Archean–Paleoproterozoic Granitic Rocks from the Kongling Terrane,South China:Implications for Tracing Thermal Evolutionary History

Feldspar Pb isotopes have been widely used to trace magmatic formation and evolution ***,it remains unclear whether post-magmatic thermal events can affect feldspar Pb isotopic ***,the in situ Pb isotopic composition of feldspar hosted in granitic rocks(thirteen Archean and one Paleoproterozoic)from the northern Kongling terrane,Yangtze Craton,South China,is *** samples reveal a substantial variation in their Pb isotopic composition,spanning the gap between the 1.9 Ga and present-day geochrons,which indicates extensive resetting by later tectonothermal *** resetting was interpreted to have likely resulted from Paleoproterozoic and Neoproterozoic tectonothermal events related to the assembly and breakup of the Columbia and Rodinia *** results suggest that Pb isotopes should be used cautiously when tracing magma sources and petrogenesis in magmatic rocks that have experienced post-magmatic ***,the in situ Pb isotopic composition of feldspar in ancient granitoids may also potentially be used to reveal later tectonothermal *** extensive resetting of the Pb isotopic composition in feldspar by regional thermal events may also provide new insights into our understanding of the Pb isotope paradox.

Temperature dependence of mechanical properties and damage evolution of hot dry rocks under rapid cooling

Understanding the differences in mechanical properties and damage characteristics of granitoid under high temperatures is crucial for exploring deep geothermal *** study analyzes the evolution of the acoustic emission(AE)characteristics and mechanical parameters of granodiorite and granite after heating and water cooling by uniaxial compression and variable-angle shear tests under different temperature *** identify their changes in mesostructure and mineral composition with electron probe microanalysis and scanning electron *** show that these two hot dry rocks have similar diagenetic minerals and microstructure,but show significantly different mechanical and acoustic characteristics,and even opposing evolution trends in a certain temperature *** the temperatures ranging from 100℃to 500℃,the compressive and shear mechanical properties of granodiorite switch repeatedly between weakening and strengthening,and those of granite show a continuous weakening *** 600℃,both rocks exhibit a deterioration of mechanical *** damage mode of granite is characterized by initiating at low stress,exponential evolutionary activity,and intensified energy *** contrast,granodiorite exhibits the characteristics of initiating at high stress,volatile evolutionary activity,and intermittent energy release,due to its more stable microstructure and fewer thermal defects compared to *** the temperature increases,the initiation and propagation of secondary cracks in granodiorite are suppressed to a certain extent,and the seismicity and brittleness are *** subtle differences in grain size,microscopic heterogeneity,and mineral composition of the two hot dry rocks determine the different acoustic-mechanical characteristics under heating and cooling,and the evolution trends with *** findings are of great significance for the scientific and efficient construction of rock mass engineering by rationally utilizin

SS-LASS Zircon Dating Deciphering Multiple Episodes of Anatexis in a Deeply-Subducted Continental Crust:An Example from Sulu Orogen,China

‘Single shot'laser-ablation split-stream(SS-LASS)technique analyzing unpolished zircon grains makes their thin rims tenable for determination,which thus offers great potential in deciphering the timing of multiple and short-lived episodes of anatexis and metamorphism in deeplysubducted continental *** granitic gneisses in the deeply subducted continental crust undergoing considerable fluid-melt activities persist multistage growth of ***,a comparative study of SS-LASS and laser ablation inductively coupled plasma mass spectrometer(LA-ICP-MS)zircon dating was conducted on the granitic gneisses from the Sulu belt in this *** mostly show a core-mantle-rim structure with CL-bright rims thinner than 5μ*** LA-ICP-MS dating,relict magmatic zircon cores yield protolith ages of ca.756-747 Ma;whereas the dark mantles record synexhumation anatexis at ca.214 *** contrast,according to the U-Pb dates,trace element features,zircon crystallization temperatures and geological context,SS-LASS zircon petrochronology deciphers three episodes of anatectic events,as follows:(i)the first episode of anatexis at ca.218-217 Ma dominated by phengite-breakdown melting,likely facilitating the exhumation of the UHP slice from mantle depth;(ii)the second episode of anatexis at ca.193–191 Ma indicating part of northern Dabie-Sulu belt was still“hot”because of buried in the thickened orogenic crust at that time;(iii)the third episode of anatexis(ca.162–161 Ma)consistent with the intrusion ages(ca.161–141 Ma)of the Jurassic to Cretaceous granitoids in this orogen,suggesting the initial collapse of the orogenic root of the Sulu belt occurred at Late Jurassic due to the Izanagi plate initially subducting beneath the margin of Eastern *** study sheds new light upon the utilization of SS-LASS petrochronology deciphering multiple anatectic events in the deeply-subducted continental crust and supports us in better understanding the tectonic evolution of Dabie-

Ash removal from inferior coal via ammonium fluoride roasting and simultaneous yield of white carbon black

The quality upgrading and deashing of inferior coal by chemical method still faces great *** dangers of strong acid,strong alkali,waste water and exhaust gas as well as high cost limit its industrial *** paper systematically investigates the ash reduction and desilicification of two typical inferior coal utilizing ammonium fluoride roasting *** the optimal conditions,for fat coal and gas coal,the deashing rates are 69.02%and 54.13%,and the desilicification rates are 92.64%and 90.27%,*** molar dosage of ammonium fluoride remains consistent for both coals;however,the gas coal,characterized by a lower ash and silica content(less than half that of the fat coal),achieves optimum deashing effect at a reduced time and *** majority of silicon in coal transforms into gaseous ammonium fluorosilicate,subsequently preparing nanoscale amorphous silica with a purity of 99.90%through ammonia *** of the fluorine in deashed coal are assigned in inorganic minerals,suggesting the possibility of further fluorine and ash removal via *** research provides a green and facile route to deash inferior coal and produce nano-scale white carbon black simultaneously.

Conceptual design of a 714-MHz RFQ for compact proton injectors and development of a new tuning algorithm on its aluminium prototype

Radio frequency quadrupoles(RFQs),which are crucial components of proton injectors,significantly affect the performance of proton accelerator *** RFQ with a high frequency of 714 MHz dedicated to compact proton injectors for medi-cal applications is designed in this *** RFQ is designed to accelerate proton beams from 50 keV to 4 MeV within a short length of 2 m and can be matched closely with the downstream drift tube linac to capture more particles through a preliminary *** develop an advanced RFQ,challenging techniques,including fabrication and tuning method,must be evaluated and verified using a *** aluminium prototype is derived from the conceptual design of the RFQ and then redesigned to confirm the radio frequency performance,fabrication procedure,and feasibility of the tuning ***,a new tuning algorithm based on the response matrix and least-squares method is developed,which yields favorable results based on the prototype,i.e.,the errors of the dipole and quadrupole components reduced to a low level after several tuning *** from the conceptual design and techniques obtained from the prototype,the formal mechanical design of the 2-m RFQ is ready for the next manufacturing step.

Assessing the energy release characteristics during the middle detonation reaction stage of aluminized explosives

Afterburning behind the detonation front of an aluminized explosive releases energy on the millisecond timescale,which prolong the release of detonation energy and the energy release at different stages also shows significant ***,at present,there are few effective methods for evaluating the energy release characteristics of the middle reaction stage of such explosives,which can have a duration of tens to hundreds of *** present work demonstrates an approach to assessing the midstage of an aluminized explosive detonation based on a water push test employing a high degree of *** this method,the explosive is contained in a steel cylinder having one end closed that is installed at the bottom of a transparent water *** detonation,the gaseous products expand in one direction while forcing water ahead of *** resulting underwater shock wave and the interface between the gas phase products and the water are tracked using an ultra-high-speed framing and streak *** shock wave velocity in water and the expansion work performed by the gaseous detonation products were calculated to assess the energy release characteristics of aluminized explosives such as CL-20 and RDX in the middle stage of the detonation *** the middle stage of the detonation process of these aluminized explosives,the aluminum reaction reduced the attenuation of shock waves and increased the work performed by gas phase products.A higher aluminum content increased the energy output while the presence of oxidants slowed the energy release *** work demonstrates an effective means of evaluating the performance of aluminized explosives.

A New Method for the Aging Evaluation of Oil-Paper Insulation Using η-Butanol and Methanol

To improve the accuracy and efficiency of the aging life prediction and assessment of transformer oil-paper insulation,and to make up for the deficiencies of traditional characterizers,such as 2-furfural,carbon monoxide,and carbon dioxide,a method for the simultaneous determination of methanol,ethanol,n−propanol,and η-butanol in oil with a single injection is established by headspace-gas chromatography-mass *** measured results show that the determination limits of the four alcohol characterizers can be controlled to 10μg/kg *** on this method,the change patterns of the above four alcohols with thermal aging time and degree of polymerization are obtained through thermal aging experimental ***,η-propanol,and η-butanol in oil indicate nearly linear correlations with thermal aging time and degree of polymerization,similar to that of *** analyzing 52 sets of measured data of 500 kV EHV transformers in operation,η-butanol is found to have excellent performance,and a new method to evaluate the aging state of oil-paper insulation employing η-butanol and methanol is proposed along with the aging attention value *** measured data of 500 kV EHV transformers in operation indicate that the combination of η-butanol and methanol as the preferred characterizers can effectively compensate for the shortcoming of traditional characterizers in the early stages of aging,and the feasibility of the method is *** possible pathways for the generation of η-butanol by cellulose cleavage during the aging of oil-paper are proposed from the chemical structure of cellulose.

A global perspective on microbial risk factors in effluents of wastewater treatment plants

Effective monitoring and management of microbial risk factors in wastewater treatment plants(WWTPs)effluents require a comprehensive investigation of these risks.A global survey on microbial risk factors in WWTP effluents could reveal important insights into their risk *** study aims to explore the abundance and types of antibiotic resistance genes(ARGs),virulence factor genes(VFGs),the vector of ARG/VFG,and dominant pathogens in global WWTP *** collected 113 metagenomes of WWTP effluents from the Sequence Read Archive of the National Center for Biotechnology Information and characterized the microbial risk *** results showed that multidrug resistance was the dominant ARG type,while offensive virulence factors were the most abundant type of *** most dominant types of ARGs in the vector of plasmid and phage were both aminoglycoside resistance,which is concerning as aminoglycosides are often a last resort for treating multi-resistant *** baumannii was the most dominant pathogen,rather than Escherichia coli,and a weak negative correlation between Escherichia coli and two other dominant pathogens(Acinetobacter baumannii and Bacteroides uniformis)suggests that using Escherichia coli as a biological indicator for all pathogens in WWTP effluents may not be *** Getah virus was the most dominant virus found in global WWTP *** study presents a comprehensive global-scale investigation of microbial risk factors in WWTP effluents,providing valuable insights into the potential risks associated with WWTP effluents and contributing to the monitoring and control of these risks.
