Piwi like RNA-mediated gene silencing 1 gene as a possible major player in gastric cancer

AIM To establish a permanent piwi like RNA-mediated genesilencing 1(PIWIL1) gene knockout in AGP01 gastric cancer cell line using CRISPR-Cas9 system and analyze phenotypic modifications as well as gene expression *** CRISPR-Cas9 system used was purchased from Dharmacon GE Life Sciences(Lafayette, CO, United States) and permanent knockout was performed according to manufacturer's recommendations. Woundhealing assay was performed to investigate the effect of PIWIL1 knockout on migration capability of cells and Boyden chamber invasion assay was performed to investigate the effect on invasion capability. For the gene expression analysis, a one-color microarray-based gene expression analysis kit(Agilent Technologies, santa Clara, CA, United States) was used according to the protocol provided by the manufacturer. RESULTS PIWIL1 gene knockout caused a significant decrease in AGP01 migration capacity as well as a significant decrease in cell invasiveness. Moreover, functional analysis based on grouping of all differentially expressed m RNAs identified a total of 35 genes(5 up-regulated and 30 down-regulated) encoding proteins involved in cellular invasion and migration. According to current literature, 9 of these 35 genes(DOCK2, ZNF503, Pde4 D, ABL1, ABL2, LPAR1, SMAD2, WASF3 and DACH1) are possibly related to the mechanisms used by PIWIL1 to promote carcinogenic effects related to migration and invasion, since their functions are consistent with the changes observed(being up-or down-regulated after knockout). CONCluSION Taken together, these data reinforce the idea that PIWIL1 plays a crucial role in the signaling pathway of gastric cancer, regulating several genes involved in migration and invasion processes; therefore, its use as a therapeutic target may generate promising results in the treatment of gastric cancer.

Stresses Analysis on a Rail Part

The EFVM （Vitoria Minas Railroad） is one of the main railways in Brazil. It transports freight trains of ore, 220 wagons each. These wagons have 2 boogies of 2 axles each and 32 metric tons on metre gauge. Elastic strains were measured on a special part of this railway due to these trains. The main load to evaluate stresses and strains was a G 16 Locomotive, a C-C kind from Vale, a Brazilian Company. The measurements were obtained by dynamic deflectometer installed on a main line of this railway, near Ipatinga, a city from Minas Gerais, one of Brazil states. This track was equipped to obtain stresses under an equal repeated static load A simulation of the stresses was made under critical strain by Ferrovia 1.0 software. It was also made an evaluation of unequal results from neighbor sleepers taking in comparison two equipped parts of this railway, one with compacted ballast and no compaction to the other. The results were strain limited, avoiding breakage or damage to the studied rails. This work analyses these measurements focusing on the improvement of track quality.

Moringa oleifera Lam(Moringaceae) is a plant with high nutritional and medicinal value. Native to India, it is now widely distributed throughout tropical and subtropical regions of the world. Its different parts are sources of proteins, vitamins and minerals and present different pharmacological and biotechnological potential. Moreover, M. oleifera seeds are widely used in water and effluent treatment, for their coagulation, flocculation and sedimentation properties, their ability of improving water quality, by reducing organic matter and microbial load, with special applicability in intensive animal production systems, such as aquaculture. In addition, due to its high nutritional value and several medicinal properties, this tree may act as a nutritional and medical alternative for socially neglected populations. In this context, this review gathers information on M. oleifera, emphasizing its chemical constituents, nutritional, pharmacological and antimicrobial properties, applications in the treatment of water effluents, and ecological and social aspects.

Objective:To investigate the in vitro antimicrobial potential of extracts of stem,leaves,flowers,pods and seeds of Moringu okifera(***) against Vibrio *** hatchery water and the prawn Mmrobrachium ***:The ethanol extracts of stem,leaves,pods and seeds and chloroform extract of flowers of *** were tested against Vibrio cholerue(***) serogroups non-O1/non-O139(n=4).Vibrio vulnificus(n=l) and Vibrio mimicus(n=1).Escherichia coli(***)(ATCC(?) 25922) was used as quality *** species were obtained from Macmbrachium amazonicum prawns and from hatchery water from prawn *** Minimum Inhibitory Concentration(MIC) was determined by broth microdilution ***:The best result was obtained with the ethanol extract of pods,which inhibited three strains of the ***,Vibrio vulnificus,Vibrio minimis and ***(MIC range 0.312-5.000 mg/mL).The chloroform extract of flowers was effective against all *** strains and ***(MIC range 0.625-1.250 mg/mL).However,the ethanol extracts of stem and seeds showed low effectiveness in inhibiting the bacterial ***:The extracts of pods,flowers and leaves of *** have potential for the control of Vibrio *** studies are necessary to isolate the bioaetive compounds responsible for this antimicrobial activity.

Growth Hormone Replacement Therapy in Patients without Adult Growth Hormone deficiency: What Answers Do We Have So Far?

Growth hormone (GH) and insulin-like growth factor 1 (IGF-1) have been suggested as “anti-aging” therapies, or for improving quality of life with aging. In this study, we focus on the actions of GH in the main organs and organ systems of the human body, like skeletal muscle, bones and brain, particularly in regard to data and research on the use of GH replacement therapy in adults without growth hormone deficiency, especially elderly patients. Several different studies have been carried out to show what the effects and side effects of GH replacement in healthy people and what would be the impact in quality of life and life span. In this review, we demonstrate what answers we have so far about the effects of GH replacement in many organs and systems in healthy people.

Evaluation between Two Brazilian Railway Tracks

Most of Brazilian railways were built more than 100 years ago. Some of them were submitted to rebuilding processes while others were just overloaded by additional layers of ballast. Nowadays, Brazil is going through a new railway transport impulse in relation to the increase of load, despite of the necessary supply capacity. For this reason, there were developed evaluations from some Brazilian railways in order to determine their operational conditions. This work shows a comparative analysis of results from two parts of studied old railway, aiming to determine minimum features to enable them to accept higher load axis. One of these studied old railway parts did not have a sub-ballast layer in contrast to the other one. The strains and stresses of these old railway track parts were generated by the same locomotive.

Individuation and the Feminine Soul: Study of Feminine Archetypal Images Carried out on Surgical Patients

This article is the result of a research carried out in a private hospital of Belo Horizonte, Brazil. The patient care model of the Mastology department includes an interdisciplinary staff that aims for the comprehensive treatment of these surgical patients. The research is based on clinical approach, under the framework of Analytical Psychology. Feminine archetypal images are symbolically represented by the Main Goddesses of the Olympus. It is natural for one of these expressions to prevail throughout a woman’s life, or even in specific moments of her life, but it is important to recognize that all of them are part of the feminine soul. We can, therefore, define and understand the Goddesses from Greek Mythology symbolically as being representative of the various feminine psychogenic forces that are projected. In reality, they are internal images that can be used to help know and understand personal issues and changes that are necessary in a woman’s life, enabling personal growth and the possibility to identify herself with the more authentic elements of her being. In this work experience, the feminine force showed itself to be a force of creation, of expression and of overcoming.

Enterobacteria and Vibrio from Macrobrachium amazonicum prawn farming in Fortaleza,Ceará,Brazil

Objective:To investigate the isolation of enterobacteria associated with Macrobrachium amazonicum(***) farming and evaluate the in vitro antimicrobial susceptibility of Vibrio ***:Strains were isolated from female *** prawns and environmental and hatchery *** assays were used to identify bacterial genera and those belonging to the genus Vibrio were submitted to further analyses for species identification,through Vitek 2 automated system and *** test was performed according to Clinical Laboratory Standards ***:The following genera of enlerobacteria were recovered:Enterobacter(n=11),Citrobacter(n=10),Proteus(n=2),Serratia(n=2),Kluyvera(n=2),Providencia(n=2),Cedecea(n=1),Escherichia(n=1).Edwardsiella(n=1) and BulliauxeUa(n=1).As for Vibrio,three species were identified:Vibrio cholerae non-O1/non-O139(n=4).Vibrio vulnificus(***)(n=1) and Vibrio mimicus(n=1).Vibrio *** minimum inhibitory concentrations values within the susceptibility range established by Clinical Laboratory Standards Institute for almost all antibiotics,except for ***,which presented intermediate profile to ***:Enterobacteria do not seem to be the most important pathogens associated with *** farming,whereas the recovery of Vibrio *** larviculture,with emphasis on Vibrio cholerae and ***,deserves special attention due to their role as potentially zoonotic aquaculture-associated ***,the intermediate susceptibility of *** to ampicillin reflects the importance of monitoring drug use in prawn farming.

Economic and Financial Viability in Spanish Thermal Power Station

This article tracked, identified, and demonstrated the economic and financial viability of the renewable energy sector from biomass in a Spanish Thermal Power Plant. The methodology used came from Theoretical postulates （GITMAN, 2004; VAN HORNE, 1993） called, Technical Preparation of capital budgets （Economic and Financial Analysis） called, payback, Net Present Value, and Internal Rate of Return）. The economic and financial viability results demonstrated were in two stages： the first identified that the plant is viable according to the payback. The second traced the economic and financial viability from the thermal energy production hypothesis equivalent to electricity power in the amount of 7,699.08 MWh, which resulted in the following indicators： payback of seven years and three months, NPV of R$ 369,064.98, and the IRR of 12.46%. Therefore, managers would have to make a user raising campaign in the ratio of 24.66%, taking into account of an unused capacity of 25%. These indicators were calculated using the following financial data： initial investment of R $ 3,654,731.86, operational income of R $1,262,264.16, operational inputs of R$ 503,639.59, and capital cost of 10.95% over 20 years. This industrial plant is located in the city of Cuellar in northern Spain, having two employees working eight hours a day; it produces only thermal power, represented in this study by MWh of electric power. This industrial plant produces power by burning forest residues from tree pruning. It was concluded that, given the research question developed in this paper--is the study plant profitable from the economic and financial point of view？--The response was identified by calculating the economic and financial indicators that the plant studied is viable from the proposed of maximizing sales revenues to 7,699.08 MWh.

Reactive oxygen species(ROS)production is a by-product of mitochondrial activity and is necessary for the acquisition of the capacitated state,a requirement for functional ***,an increase in oxidative stress,due to an abnormal production of ROS,has been shown to be related to loss of sperm function,highlighting the importance of an accurate detection of sperm ROS,given the specific nature of this *** this work,we tested a variety of commercially available fluorescent probes to detect ROS and reactive nitrogen species(RNS)in human sperm,to define their *** both flow cytometry(FC)and fluorescence microscopy(FM),we confirmed that MitoSOX™Red and dihydroethidium(DHE)detect superoxide anion(as determined using antimycin A as a positive control),while DAF-2A detects reactive nitrogen species(namely,nitric oxide).For the first time,we also report that RedoxSensor™Red CC-1,CellROX®Orange Reagent,and MitoPYl seem to be mostly sensitive to hydrogen peroxide,but not ***,mean fluorescence intensity(and not percentage of labeled cells)is the main parameter that can be reproducibly monitored using this type of methodology.
