Endotoxins(also known as lipopolysaccharides(LPS)) are undesirable by-products of recombinant proteins,purified from Escherichia coli.LPS can be considered stable under a wide range of temperature and pH,making their removal one of the most difficult tasks in downstream processes during protein purification.The inherent toxicity of LPS makes their removal an important step for the application of these proteins in several biological assays and for a safe parenteral administration.Immobilized metal affinity chromatography(IMAc) enables the affinity interactions between the metal ions(immobilized on the support through the chelating compound) and the target molecules,thus enabling high-efficiency separation of the target molecules from other components present in a mixture.Affinity chromatography is applied with ca2+-iminodiacetic acid(IDA) to remove most of the LPS contaminants from the end product(more than90%).In this study,the adsorption of LPS on an IDA-ca2+ was investigated.The adsorption Freundlich isotherm of LPS-IDA-ca2+ provides a theoretical basis for LPS removal.It was found that LPS is bound mainly by interactions between the phosphate group in LPS and ca2+ ligands on the beads.The factors such as pH(4.0 or 5.5) and ionic strength(1.0 mol/L) are essential to obtain effective removal of LPS for contaminant levels between endotoxin' concentration values less than100 EU/mL and 100 000 EU/mL.This new protocol represents a substantial advantage in time,effort,and production costs.

Affinity and ion exchange conventional chromatography have been used to capture erythropoietin(EPO)from mammalian cell culture supernatant.currently,chromatographic adsorbent perfusion is available,however a limited number of applications have been found in the literature.In this work,three anion exchange chromatographic supports(gel,membrane and monolithic)were evaluated in the capture step of the recombinant erythropoietin purification process.The influences of load and flow rate on each support performance were analyzed.Also the purity of the EPO molecules was determined.A productivity analysis,as a decision tool for larger scale implementation,was done.As a conclusion,the evaluated supports are technically suitable to capture EPO with adequate recovery and good purity.However,the monolithic column admits high operating velocity,showing the highest adsorption capacity and productivity.

comparison of cIM~ c4-HLD monolithic column with Sartobind phenyl membrane column for pIDKE2 purification

The main component of the center for Genetic Engineering and Biotechnology(cIGB)candidate vaccine against Hepatitis c virus(HcV)is the pIDKE2 plasmid.The current designed downstream process for the production of pIDKE2 fulfils all regulatory requirements and renders the required quantities of pharmaceuticalgrade plasmid DNA(pDNA)with 95%purity.The advantages of this procedure include high plasmid purity and the elimination of undesirable additives,such as toxic organic extractants and animal-derived enzymes.However,yields and consequently the productivity of the process are low.Previous work demonstrated that the most critical step of the process is the reverse phase chromatography,where conventional porous particle resins are used.Therefore,to increase the process productivity,alternative technologies such as membranes and chromatographic monoliths were tested as alternative options for this critical step.Here,a comparison between the behaviors of cIM~ c4-HLD and Sartobind phenyl matrices was performed.To obtain higher productivities and purities,the dynamic binding capacities and selectivities were evaluated.The results showed that both matrices had a similar capacity for pIDKE2 plasmid,but the separation of pDNA isoforms using cIM~ technology was much better than that with Sartobind.Additionally,the optimal conditions for loading plasmid DNA on a cIMc4-HLD 800-mL monolithic column in a real production process were determined.These optimizations will allow production levels to satisfy the high plasmid consumption demanded by clinical trials.

Anion exchange chromatography is the most popular chromatographic method for plasmid separation.POROS R1 50 is a perfusio n chro mato graphic suppo rt w hich is a reversed phase matrix and is an alternative to co nventio nal o nes due to its mass transfer pro perties.The adso rptio n and elutio n o f the pIDKE2 plasmid o nto reversed phase POROS R1 50 w as studied.Langmuir iso therm mo del w as adjusted in o rder to get the max imum adso rptio n capacity and the disso ciatio n co nstant fo r POROS R1 50-plasmid DNA(pDNA) system.Breakthro ugh curves w ere o btained fo r vo lumetric flo w s betw een 0.69-3.33 mL/min,given dynamic capacity up to 2.3 times higher than tho se repo rted fo r io nic ex change matrix used during the purificatio n pro cess o f plasmids w ith similar size to that o f pIDKE2.The efficiency w as less than 45% fo r the flo w co nditio ns and initial co ncentratio n studied,w hich means that the suppo rt w ill no t be o perated under saturatio n circumstances.

Origin of Traditional chinese Medicine in cuba in the 19th century from Its Main Exponents and Some Notable Medical Descendants in the 20th century

The cuban people are made up of three major migratory currents, the chinese are one of them. They brought their culture, the methods, and procedures of traditional chinese medicine(TcM) in the 19th century. Few were able to return and so they created families in cuba;some of their descendants dedicated themselves to medicine. In order to investigate the practices that were predecessors of TcM in cuba in the 19th century, a qualitative phenomenological research was carried out, reviewing what was published by various sources, applying documentary analysis, logical historical analysis, abstraction, synthesis, and systematization of the results on the regularities of the work and human behavior of chinese doctors in the cuban 19th and 20th centuries. This made it possible to identify six chinese doctors in the 19th century in cuba who gave rise to the beginning of some practices of TcM in cuba, and five from the 20th century, descendants of coolies who dedicated themselves to other specialties of medicine. It was found that despite their geographical and time disperse, they were all notorious for their outstanding professional and human behavior, with a trail of accumulated successes in achieving “almost the impossible” with the patient. They have left their mark on cuban culture.

Frailty and acute coronary syndrome: does gender matter?

There are important sex-related differences in elderly patients with acute coronary syndrome(AcS).Women are older,more frequently frail,and present more comorbidities than men.Atypical symptoms at presentation are also more common in female patients,they are leaded to a delayed diagnosis and treatment.coronary angiography and subsequent revascularization are frequently underused in elderly women and they tend to receive less guidelines-recommended therapies.The prognosis in elderly frail women with AcS is poor,and it is with high mortality and readmissions rates.Bleeding is recurrent ischemic events in which it is more frequent in women than in men.Recovery time might be long,and a multidisciplinary approach is desirable to improve prognosis and quality of life.Further studies are needed in order to clarify the benefit of the different therapies in the group of frail women,and this is particularly true for revascularization,as scientific evidence in this group is very scarce.

The projection of world population growth with concurrent generation of large volumes of agroindustrial waste that negatively affect the environment is of great concern.Therefore,this review article describes the nexus between concepts of circular Bioeconomy,Zero Waste Technology,Sustainable Development,Biorefineries,and alternatives and research efforts to generate less environmental impact.A brief analysis of the Ecuadorian industry and exports is described,emphasizing the fact that,to improve the Ecuadorian trade balance,it is necessary to increase industrial competitiveness.It is important to have emerging technologies and innovation in order to promote the replacement of fossil-derived raw materials with renewable raw materials and develop more environmentally friendly processes and industries.This paper analyses the state of biomass research and its transformation in Ecuador,together with current pretreatment research on biomass to obtain bioproducts and biofuels in a biorefinery that promotes clean production for the extraction of phytochemicals using green solvents,such as deep eutectic solvents;and technologies to recover high-value added materials with enhanced properties.In conclusion,the need to develop technologies and markets to commercialize high value-added products coming from biorefineries is highlighted,as this will increase the income both in rural and urban areas and will strengthen the productivity and profitability of the Ecuadorian agroindustry.Our goal through this analysis is to improve Ecuador’s trade balance while also contributing to the circular bioeconomy that promotes sustainable development.

Seed Dispersal of citharexylum tetramerum and Ziziphus penduculata by carnivorous in a Xerophilous Scrub at Tehuacan, Puebla, Mexico

Seed dispersion reduces the depredation rate and increases genetic flow. Some species of carnivora consume fruits as a standard component of their diet, so they become potential seed dispersers. In Mexico, a few studies evaluated carnivorous as seed dispersers, especially in dry ecosystems. citharexylum tetramerum and Ziziphus pedunculata are endemic plants from the Tehuacán-cuicatlán’s Biosphere Reserve (TcBR);however, there are very little data about dispersal seeds for both species. We assessed the germination of seeds ingested by carnivores obtained from feces. We compared them with seeds from fruits as a control group. At the Metropolitan University laboratory, we germinated seeds, from: plants, scats, and fruits, and we used Kruskall Wallis Test to compare percentage and germination rate. c. tetramerum seeds ingested by a carnivorous decreased in germination percentage and rate. In contrast, the percentage and rate of germination of Z. pedunculata seeds from carnivorous feces were higher than the control group. However, Z. pedunculata control seeds did not germinate during tests, but at the end of the tests, we found that 90% of control seeds were not dead, so they were dormant seeds. carnivores interact as legitime dispersers for both plants because they are keeping the viability of seeds. Still, we only showed that Z. pedunculata gut carnivores broke with the dormancy of seeds. We consider that in future research, it could be essential to identify other animals that consume fruits of those plants and if it is possible to follow seeds after they have been removed. Those points could help to get better understanding of those endemic plants from Tehuacán.

Adsorption of Mn^(2+) from aqueous solution using Fe and Mn oxide-coated sand

The adsorption of Mn2＋ onto immobilized Mn-oxide and Fe-oxide adsorbent such as manganese oxide-coated sand 1 （MOcS1）, manganese oxide-coated sand2 （MOcS2）, iron oxide-coated sand2 （IOcS2）, and manganese and iron oxide-coated sand （MIOcS）was investigated. The effects of pH （5.5 to 8.0） and temperature （25 to45°c） on the equilibrium capacitywere examined. Equilibrium studies showed that there is a good fitwith both Freundlich and Langmuir isotherm,which indicates surface heterogeneity and monolayer adsorption of the adsorbents. Kineticdata showed high correlationwith the pseudo second-order model,which signifies a chemisorption-controlled mechanism. The activation energies, activation parameters （ΔG＊,ΔH＊,ΔS＊）, and thermodynamic parameters （ΔG0 , ΔH0 , ΔS0 ） confirmed that adsorptionwith MIOcSwas endothermic and more spontaneous at higher temperaturewhile an opposite trendwas observed for the other adsorbents. Thermodynamic studies showed that adsorption involved formation of activated complex,where MOcS1 and MIOcS follow a physical-chemical mechanism,while MOcS2 and IOcS2 follows purely chemical mechanism.
