Elastic com.osites containing liquid inclusions exist widely in nature and engineering fields. The volum.tric response of liquid inclusions is im.ortant in m.ny cases, such as an isolated cell em.edded in an extracellular m.trix and an oil pocket em.edded within shale. In this study, we developed a m.del for describing the volum.tric response of an ellipsoidal liquid inclusion. Specifically, we investigated the volum.tric response of an ellipsoidal liquid inclusion em.edded in a three-dim.nsional (3D) m.trix through an analytical expression of the volum.tric response. We perform.d param.tric analysis and found that loading along the shortest axis can induce the m.st volum. change, while loading along the longest axis can induce the least volum. change. We also found that the volum.tric response decreases with increasing Poisson ratio of the m.trix. These results could be used to understand som. cell behavior in a 3D m.trix, for exam.le, cell alignm.nt under m.chanical load.

Different from.canonical ubiquitin-like proteins, Hub1 does not form.covalent conjugates with substrates but binds proteins noncovalently. In Socchorom.ces cerevisioe, Hub1 associates with spUceosom.s and m.diates alternative splicing of SRCI, without affecting pre-m.NA splicing generaity. Hum.n Hub1 is highty sim.lar to its yeast hom.tog, but its cellular function rem.ins largely unexplored. Here, we show that hum.n Hub1 binds to the spliceosom.l protein Snu66 as in yeast; however, unlike its 5. cerevisioe hom.los, hum.n Hub1 is essential for viability. Prolonged in vivo depletion of hum.n Hub1 leads to various cellular defects, including splicing speckle abnorm.lities, partial nuclear retention of m.NAs, m.totic catastrophe, and consequently cell death by apoptosis. Early consequences of Hub1 depletion are severe splicing defects, however, only for specific splice sites leading to exon skipping and intron retention. Thus, the ubiquitin-iike protein Hub1 is not a canonlcal spliceosom.l factor needed generally for splicing, but rather a m.dulator of spliceosom. perform.nce and facilitator of alternative splicing.

Four awl-substituted tripyrrinone copper（II） com.lexes were synthesized and characterized as to their electro- chem.cal and spectroelectrochem.cal properties. The exam.ned com.ounds are represented as （YPh）3TPO2Cu, where TPO2 is a dianion of the tripyrrinone and Y is a CH3O, CH3, H or CI substituent on the para-position of each phenyl ring of the com.ound. The neutral com.ounds are ESR active and show a typical d9 Cu（II） signal in frozen CH2C12 solution. Each Cu（II） com.ound undergoes three reductions between -0.61 and -1.90 V and two oxidations between 1.00 and 1.24 V in CH2Cl2 containing 0.1 m.l/L tetra-n-butylam.onium.perchlorate. The potentials for oxidation are sim.lar to those of copper tetraarylporphyrins but the reduction potentials are positively shifted as com.ared to the corresponding tetraarylporphyrins. Thus, the potential difference between the first one-electron oxidation and first one-electron reduction （the HOm.-LUm. gap） of （YPh）3TPO2Cu, which ranges from.1.68 to 1.75 V, is sim.lar to that of copper corroles （1.73-1.85 V）, but sm.ller than the separation of 2.30-2.37 V for the copper tetraarylporphyrins under the sam. solution conditions.

Sulfur is essential for plant growth and developm.nt, and the m.lecular system. for m.intaining sulfur and thiol m.tabolism.are tightly controlled. From.a biochem.cal perspective, the regulation of plant thiol m.tabolism.high- lights nature＇s ability to engineer pathways that respond to m.ltiple inputs and cellular dem.nds under a range of con- ditions. In this review, we focus on the regulatory m.chanism. that form.the m.lecular basis of biochem.cal sulfur sensing in plants by translating the intracellular concentration of sulfur-containing com.ounds into control of key m.tabolic steps. These m.chanism. range from.the sim.le （substrate availability, therm.dynam.c properties of reactions, feedback inhi- bition, and organelle localization） to the elaborate （form.tion of m.ltienzym. com.lexes and thiol-based redox switches）. Ultim.tely, the dynam.c interplay of these regulatory system. is critical for sensing and m.intaining sulfur assim.lation and thiol m.tabolism.in plants.

Gene Expression Profiling of Hum.n Epiderm.l Keratinocytes in Sim.lated m.crogravity and Recovery Cultures

Sim.lated m.crogravity （Sm.） bioreactors and DNA m.croarray technology are powerful tools to identify ＂space genes＂ that play key roles in cellular response to m.crogravity. We applied these biotechnology tools to investigate Sm. and post-Sm. recovery effects on hum.n epiderm.l keratinocytes by exposing cells to Sm. for 3, 4, 9, and 10 d using the high aspect ratio vessel bioreactor followed by recovery culturing for 15, 50, and 60 d in norm.l gravity. As a result, we identified 162 differentially expressed genes, 32 of which were ＂center genes＂ that were m.st consistently affected in the tim. course experim.nts. Eleven of the center genes were from.the integrated stress response pathways and were coordinately downregulated. Another seven of the center genes, which are all m.tallothionein m.-Ⅰ and m.-Ⅱ isoform., were coordinately up-regulated. In addition, HLA-G, a key gene in cellular im.une response suppression, was found to be significantly upregulated during the recovery phase. Overall, m.re than 80% of the differentially expressed genes from.the shorter exposures （≤4 d） recovered in 15 d; for longer （≥9 d） exposures, m.re than 50 d were needed to recover to the im.act level of shorter exposures. The data indicated that shorter Sm. exposure duration would lead to quicker and m.re com.lete recovery from.the m.crogravity effect.
