Mass Spectrometric Analyses Reveal a Central Role for Ubiquitylation in Remodeling the Arabidopsis Proteome during Photomorphogenesis

The switch from skotomorphogenesis to photomorphogenesis is a key developmental transition in the life of seed plants. While much of the underpinning proteome remodeling is driven by light-induced changes in gene expression, the proteolytic removal of specific proteins by the ubiquitin-26S proteasome system is also likely paramount. Through mass spectrometric analysis of ubiquitylated proteins affinity-purified from etiolated Arabidopsis seedlings before and after red-light irradiation, we identified a number of influential proteins whose ubiquitylation status is modified during this switch. We observed a substantial enrichment for proteins involved in auxin, abscisic acid, ethylene, and brassinosteroid signaling, peroxisome function, disease resistance, protein phosphorylation and light perception, including the phytochrome （Phy） A and phototropin photoreceptors. Soon after red-light treatment, PhyA becomes the dominant ubiquitylated species, with ubiquitin attachment sites mapped to six lysines. A PhyA mutant protected from ubiquitin addition at these sites is substantially more stable in planta upon photoconversion to Pfr and is hyperactive in driving photomorphogenesis. However, light still stimulates ubiquitylation and degradation of this mutant, implying that other attachment sites and/or proteolytic pathways exist. Collectively, we expand the catalog of ubiquitylation targets in Arabidopsis and show that this post- translational modification is central to the rewiring of plants for photoautotrophic growth.

Origin of high particle number concentrations reaching the St. louis, Midwest Supersite

Ultrafine particles are associated with adverse health effects. Total Particle Number Concentration（TNC） of fine particles were measured during 2002 at the St. louis — Midwest supersite. The time series showed overall low level with frequent large peaks. The time series was analyzed alongside criteria pollutant measurements and meteorological observations. Multiple regression analysis was used to identify further contributing factors and to determine the association of different pollutants with TNC levels. This showed the strong contribution of sulfur dioxide（SO2） and nitrogen oxides（NO x） to high TNC levels. The analysis also suggested that increased dispersion resulting from faster winds and higher mixing heights led to higher TNC levels. Overall, the results show that there were intense particle nucleation events in a SO2 rich plume reaching the site which contributed around 29% of TNC. A further 40% was associated with primary emissions from mobile sources. By separating the remaining TNC by time of day and clear sky conditions,we suggest that most likely 8% of TNC are due to regional nucleation events and 23% are associated with the general urban background.

Hepatic intra-arterial infusion of yttrium-90 microspheres in the treatment of recurrent hepatocellular carcinoma after liver transplantation: A case report

Hepatocellular carcinoma （HCC） recurs with a reported frequency of 12%-18% after liver transplantation. Recurrence is associated with a mortality rate exceeding 75%. Approximately one-third of recurrences develop in the transplanted liver and are therefore amenable to local therapy. A variety of treatment modalities have been reported including resection, transarterial chemoembolization （TACE）, radiofrequency ablation （RFA）, ethanol ablation, cryoablation, and external beam irradiation. Goals of treatment are tumor control and the minimization of toxic effect to functional parenchyma. Efficacy of treatment is mitigated by the need for ongoing immunosuppression. Yttrium-90 microspheres have been used as a treatment modality both for primary HCC and for pre-transplant management of HCC with promising results. Twenty-two months after liver transplantation for hepatitis C cirrhosis complicated by HCC, a 42-year old man developed recurrence of HCC in his transplant allograft. Treatment of multiple right lobe lesions with anatomic resection and adj.vant chemotherapy was unsuccessful. Multifocal recurrence in the remaining liver allograft was treated with hepatic intra-arterial infusion of yttrium-90 microspheres （SIR-Spheres, Sirtex Medical Inc., Lake Forest, IL, USA）. Efficacy was demonstrated by tumor necrosis on imaging and a decrease in alpha-fetoprotein （AFP） level. There were no adverse consequences of initial treatment.

Irrigation Termination Thermal Time and Amount on Cotton Lint Yield and Fiber Quality

Cotton irrigation in the Texas High Plains (THP) is often dictated by the well capacity and not by the water needs of the crop. The source of irrigation-water is the Ogallala aquifer and in many areas of the THP, the water table has declined to well capacities that deliver 1.3 to >7.6 mm/d. There is plenty of information on cotton responses to irrigation frequency and amount;however, information on when to terminate irrigation and its effect on cotton lint yield and fiber quality is scarce. Our obj.ctive was to evaluate over a 4-year period three irrigation termination thermal times corresponding to cumulative daily heat units (∑HU) of 890 °C, 1000 °C and 1110 °C from crop emergence, and three levels of irrigation (2.5, 5.1 and 7.6 mm/d) on cotton lint yield and fiber quality. Irrigation was applied with a sprinkler system on a 3-day frequency in Lubbock, TX. Results showed that on average the 7.6 mm/d level produced the most cotton lint yield regardless of the irrigation termination thermal time. Terminating cotton at 1000- °C ∑HU resulted in water savings of 25 to 50 mm for the 2.5 and 5.1 mm/d levels without significantly affecting lint yield. For the 7.6 mm/d and terminating at 890- °C ∑HU resulted in water savings of 100 to 115 mm. Average fiber length statistically increased with termination thermal time and level. This effect was most significant in years with the least rain and warmer air temperature. Micronaire increased with the termination thermal time in years with >500 mm of rain. Average fiber length uniformity and fiber strength were minimally affected by termination thermal time. As irrigation level increased, the average micronaire decreased, and fiber strength increased for the 5.1 and 7.6 mm/d irrigation. We concluded that in the THP for well capacities that deliver 2.5 - 5.1 mm/d irrigation can be terminated when the ∑HU reaches about 1000 °C from emergence without impacting cotton lint yield.
