Nutlin-3a.is a.MDM2 a.ta.onist a.d preclinica. drug tha. a.tiva.es p53. Cells with MDM2 gene a.plifica.ion a.e especia.ly prone to Nutlin-3a.induced a.optosis, though the ba.is for this is unclea.. Glucose meta.olism ca. inhibit a.optosis in response to Nutlin-3a.through mecha.isms tha. a.e incompletely understood. Glucose meta.olism through the pentose phospha.e pa.hwa. （PPP） produces Na.PH tha. ca. protect cells from potentia.ly letha. rea.tive oxygen species （ROS）. We compa.ed a.optosis a.d glucose meta.olism in ca.cer cells with a.d without MDM2 gene a.plifica.ion trea.ed with Nutlin-3a. a.optosis in MDM2-a.plified cells wa. a.socia.ed with a.reduction in glycolysis a.d the PPP, reduced Na.PH, increa.ed ROS, a.d depletion of the tra.scription fa.tor SP1, which norma.ly promotes PPP gene expression. In contra.t, glycolysis a.d the PPP were ma.nta.ned or increa.ed in MDM2 non-a.plified cells trea.ed with Nutlin-3a. This wa. dependent on p53-media.ed a.T a.tiva.ion a.d wa. a.socia.ed with ma.ntena.ce of SP1 a.d continued expression of PPP genes. Knockdown or inhibition of a.T, SP1, or the PPP sensitized MDM2-non-a.plified cells to a.optosis. The da.a.indica.e tha. p53 promotes a.T a.d SP1-dependent a.tiva.ion of the PPP tha. protects cells from Nutlin-3a.induced a.optosis. These findings provide insight into how glucose meta.olism reduces Nutlin-3a.induced a.optosis, a.d a.so provide a.mecha.ism for the heightened sensitivity of MDM2-a.plified cells to a.optosis in response to Nutlin-3a.
