Oxygen(O_(2))-sensing matrices are promising tools for the live monitoring of extracellular O_(2) consumption levels in long-term cell *** this study,ratiometric O_(2)-sensing membranes were prepared by electrospinning,an easy,low-cost,scalable,and robust method for fabricating ***(ε-caprolactone)and poly(dimethyl)siloxane polymers were blended with tris(4,7-diphenyl-1,10-phenanthroline)ruthenium(II)dichloride,which was used as the O_(2)-sensing probe,and rhodamine B isothiocyanate,which was used as the reference *** functionalized scaffolds were morphologically characterized by scanning electron microscopy,and their physicochemical profiles were obtained by Fourier transform infrared spectroscopy,thermogravimetric analysis,and water contact angle *** sensing capabilities were investigated by confocal laser scanning microscopy,performing photobleaching,reversibility,and calibration curve studies toward different dissolved O_(2)(DO)*** sensing nanofibers showed a high response to changes in DO concentrations in the physiological-pathological range from 0.5%to 20%and good stability under ratiometric *** addition,the sensing systems were highly biocompatible for cell growth promoting adhesiveness and growth of three cancer cell lines,namely metastatic melanoma cell line SK-MEl2,breast cancer cell line MCF-7,and pancreatic ductal adenocarcinoma cell line Panc-1,thus recreating a suitable biological environment in *** O_(2)-sensing biomaterials can potentially measure alterations in cell metabolism caused by changes in ambient O_(2)content during drug testing/validation and tissue regeneration processes.

Carbon sequestration capability of Fagus sylvatica forests developing in the Majella National Park (Central Apennines,Italy)

Terrestrial ecosystems represent a major sink for atmospheric carbon(C) and temperate forests play an important role in global C cycling, contributing to lower atmospheric carbon dioxide(CO2) concentration through photosynthesis. The Intergovernmental Panel of Climate Change highlights that the forestry sector has great potential to decrease atmospheric CO2concentration compared to other sectoral mitigation activities. The aim of this study was to evaluate CO2sequestration(CO2S)capability of Fag us sylvatica(beech) growing in the Orfento Valley within Majella National Park(Abruzzo,Italy). We compared F. sylvatica areas subjected to thinning(one high-forest and one coppice) and no-management areas(two high-forests and two coppices). The results show a mean CO2S of 44.3 ± 2.6 Mg CO2ha-1a-1,corresponding to 12.1 ± 0.7 Mg C ha-1a-1the no-managed areas having a 28% higher value than the managed areas. The results highlight that thinning that allows seed regeneration can support traditional management practices such as civic use in some areas while no management should be carried out in the reserve in order to give priority to the objective of conservation and naturalistic improvement of the forest heritage.

Phenolic Acids Content and Nutritional Quality of Conventional, Organic and Biodynamic Cultivations of the Tomato CXD271BIO Breeding line (Solanum lycopersicum l.)

This study was undertaken to evaluate the influence of growing systems (conventional, organic and biodynamic performed with two types of manuring) on some nutritional traits of the tomato breeding line CXD271BIO (Solanum lycopersicum l.). Experimental fields were in the same geographical area and the cultivations were performed over five years. The content of macronutrients, minerals (Ca, K, Mg, P, Na), trace elements (Fe, Zn, Cu, Mn) and phenolic acids (chlorogenic, caffeic, p-coumaric and ferulic acids) was determined. The biodynamic A growing system showed the highest protein and carbohydrates content compared to the other growing systems. Data on minerals and trace elements content showed significant differences (mainly in Ca, Na, Fe and Zn content) between conventional and biodynamic tomatoes, whereas no major impact of the fertilization among organic and the two biodynamic growing systems was observed. Appreciable differences in phenolic acids biosynthesis were observed, with the year of harvest showing a marked effect especially on chlorogenic, p-cumaric and ferulic acids content. Conversely, the cultivation system did not show major influence. Principal Components Analysis (PCA), performed on mineral and phenolic acids content, substantiated the large effect of the year of harvest, also suggesting an effect of the conventional cultivation system.

Conservation of Endangered Species: Androsace mathildaelevier (Primulaceae) in Central Italy

Climate change influences the distribution of species, causing the displacement of distribution areas, both in altitude and latitude. The potentially most climate change-susceptible taxa are those of mountain peaks, with limited distribution areas, such as rare or endemic species. In this paper, we deepened the study of Androsace mathildae levier, a plant that lives in Central Italy over 2500 m asl. This species, classified as a rare plant according to the Red list of Plants of Italy, is included in Appendix I of the Bern Convention, Annex II and IV Habitats Directive 92/43 CEE, and is protected under Regional law n. 45/79 of the Abruzzo Italian Region. In order to preserve A. mathildae, morphological observations and ex situ preservation have been performed. Results indicate that the germination of seeds increases in the presence of extracts of humic acids obtained by the decomposition of the lower leaves of the plant. The presence of Pyrenophora bromi (Ascomyta) on the leaves as a decomposition agent, has never been reported either on A. mathildae or at these altitudes. A number of experiments to obtain callus induction and neomorphogenesis were conducted to identify suitable procedures for in vitro stabilization. The obtained shoots had a high number of lateral buds (6.3 per plant, on average) which were used to increase the number of clones present in the growth chamber within a few sub-culture passages. This in vitro material will be used in cryopreservation and to improve the restocking activity of A. mathildae.

The main of the research was to analyze the leaf metal accumulation capability of Platanus acerifolia (Aiton) Willd., Ailantus altissima (Mill.) Swingle, Robinia pseudoacacia L. and Quercus ilex L., largely distributed in Rome. In addition, metal concentration was analyzed in the soil, sampling sites were chosen in historical parks (A sites) and high traffic level sites (B sites). The results highlight significant higher leaf and soil metal concentrations in B than in A sites. The ratio between metal concentration in leaves and soils (Biological Absorption Coefficient, BAC) for all the considered sites was significantly different among the species. le="font-family:Verdana;">Morphological and anatomical leaf traits of the considered species show significant differences in A and B sites in response to traffic level. Overall, the results highlight the importance of the selection of tree species in urban areas for their ability to lower pollution levels.

随着重金属污染日趋严重,环境中的重金属进入食物链,从而给动物和人类带来危害。近年来的研究表明,不是所有的重金属都会被动物和人类吸收,生物吸收重金属的量取决于重金属的存在形态。能够被生物吸收的形态称为生物可利用态,所以通过测量生物可利用形态的浓度了解重金属的生物有效性显得尤为重要。测量生物可利用浓度的方法有很多,薄膜扩散梯度技术是其中的一种。本文采用羧甲基纤维素钠(CMC)为结合相、醋酸纤维素渗析膜为扩散相,组装成来了 CMC-DGT装置。对作为结合相的CMC浓度进行了优选,CMC作为结合相的最佳浓度为0.015mol/l。镉离子通过作为扩散相的醋酸纤维素渗析膜的扩散系数为1.097x10-6cm2/s。探索了镉离子浓度对CMC-DGT及Rhodeinae鱼的镉吸收量的影响。随着镉离子浓度的增大,鱼鳃、鱼体和CMC-DGT累积的量也随之增加。鱼鳃累积镉的量远大于鱼体累积镉的量。通过实验得到了 CMC-DGT累积镉的量与鱼鳃和鱼体吸收镉的量的相关性。鱼鳃累积量(y1)和CMC-DGT累积量(x1)的相关关系为:y1=3.8014x1+72.65,R2=0.7444,鱼体累积量(y2)和CMC-DGT累积量(x2)的相关关系为:y2=0.3179x2+7.6442,R2=***-DGT具有预测镉对Rhodeinae鱼得生物有效性的潜力。

life sciences such as medical, nutritional, and those related to agrarian biotechnologies are experiencing a constant increase. Therefore, food consumption is today related to health state. Aiming to create an innovative spinneret system that defines the tight correlation between high nutritional and nutraceutical value of foods and their beneficial effects on consumer’s health state, we tested the effects related to the administration of 40 g of the Tonda Gentile Romana Italian hazelnut cultivar (Viterbo, Italy). To this end, we enrolled 24 subjects for a period of 14 weeks, based on a crossover design. Although several papers and reviews on hazelnut intake benefits are available, few studies have investigated the effects of hazelnut-enriched diet on plasma key parameters commonly used to ascertain the individual’s well-being status. The primary endpoint of the trial was to assess lDl change related to hazelnut administration, which resulted in significant beneficial effects on cholesterol parameters (lDl dropped from 133.09 mg/dl to 119.18 mg/dl). Interestingly, despite within the normal ranges, uric acid and serum creatinine mean values significantly decreased after hazelnut intake. A significant increment was also observed for AlT (from 30.9 U/l to 35.22 U/l, P = 0.01), while GGT significantly decreased from 38.04 mg/dl at T0 to 35.27 mg/dl at T1 (P = 0.001). No significant changes were also observed in azotemia, serum iron, glycemia, AST, total bilirubin, Hb, WBCs, RBCs, platelet count, and total plasma proteins. All the blood parameters analyzed appeared to be reversible after 6 weeks from the end of the treatment. Mann-Whitney U test did not show any significant differences in T0 - T1 Δ% lDl considering gender, age and baseline lDl. These findings may be of help in the development of strategies for personalized nutrition and diseases management.

Partial characterization of the N-linked oligosaccharide structures on P-selectin glycoprotein ligand-1 (PSGl-1)

PSGl-1, a specific ligand for P-, E- and l-selectin, was isolated from in vivo [3H]-glucosamine labeled Hl- 60 cells by a combination of wheat germ agglutinin-agarose and P- or E-selectin-agarose chromatography. N- linked oligosaccharides were released from the purified, denatured ligand molecule by peptide: N-glycosidase F treatment and, following separation by Sephacryl S-200 chromatography, partially characterized using lectin, ion-exchange and size-exclusion chromatography in combination with glycosidase digestions. The data obtained suggest that the N-glycans on PSGl-1 are predominantly core-fucosylated, multiatennary complex type structures with extended, poly- N- acetyllactosamine contaniing outer chains. A portion of the outer chains appears to be substituted with fucose indicating that the N-glycans, in addition to the O-glycans on PSGl-1, may be involved in selectin binding.
