C-reactive protein decreases interleukin-8 production in human endothelial progenitor cells by inhibition of p38 MAPK pathway

Background C-reactive protein （CRP） has been reported to damage the vascular wall by inducing endothelial dysfunction and inflammation, and it is also speculated to have a role in attenuating angiogenic functions of human endothelial progenitor cells （EPCs）. Interleukin-8 （IL-8） is an important mediator of the paracrine mitogenic effect of EPCs which has direct angiogenic effects on mature endothelial cells. We, herein, investigated the direct effect of CRP on IL-8 production and gene expression in cultured human EPCs. Methods EPCs were isolated from the peripheral venous blood of healthy male volunteers. Cells were cultured in EndoCultTM liquid medium in the absence and presence of CRP at clinically relevant concentrations （5 to 25 μg/ml） for different durations （3 to 48 hours）. IL-8 protein and mRNA of cultured EPCs were evaluated using ELISA and real-time PCR. Results The results showed that CRP at a concentration of 10 μg/ml significantly reduced IL-8 secretion of cultured EPCs with a peak at 25 μg/ml, and also decreased mRNA expression in EPCs with a peak at 12 hours. In addition, preincubation of EPCs with SB203580, an inhibitor of p38 mitogen-activated protein kinase （MAPK） decreased CRP inhibition of IL-8 mRNA expression at 12 hours in EPCs. Conclusions Our study, for the first time, demonstrates that CRP directly inhibits EPCs IL-8 secretion, a key cytokine player of angiogenesis induced by EPCs. Inhibition occurred in part via an effect of CRP to active the p38 MAPK signal transduction pathway in EPC. The ability of CRP to inhibit EPCs IL-8 secretion may represent an important mechanism that further links inflammation to cardiovascular disease.

AIM:To investigate macrophage migration inhibitory factor(MIF) expression and its clinical relevance in gastric cancer,and effects of MIF knockdown on proliferation of gastric cancer cells. METHODS:Tissue microarray containing 117 samples of gastric cancer and adjacent non-cancer normal tissues was studied for MIF expression by immunohistochemistry(IHC) semiquantitatively,and the association of MIF expression with clinical parameters was analyzed. MIF expression in gastric cancer cell lines was detected by reverse transcriptionpolymerase chain reaction(RT-PCR) and Western blot. Two pairs of si RNA targeting the MIF gene(MIF si-1 and MIF si-2) and one pair of scrambled si RNA as a negative control(NC) were designed and chemically synthesized. All si RNAs were transiently transfected in AGS cells with OligofectamineTM to knock down the MIF expression,with the NC group and mock group(OligofectamineTM alone) as controls. Esophageal squamous cell carcinoma(ESCC) is known for its rapid progression and poor outcomes. China has the highest incidence and mortality in the world. Diagnoses made at early stages and accurate staging are associated with better outcomes, all of which can play a significant role in the selection of treatment protocols. ESCC is staged according to the widely accepted TNM system. Common imaging modalities used in staging ESCC before treatment include endoscopy, computed tomography(CT), positron emission tomography(PET) and magnetic resonance imaging(MRI). Endoscopic ultrasound is useful for staging tumor depth and nodal status. Narrow band imaging is valuable for early stage disease assessment. CT and PET provide additional valuable information regarding node and metastasis staging. The ability of MRI to delineate ESCC is continuously being improved and adds information regarding locoregional status to routine examinations.

Background:Enlarged retropharyngeal lymph nodes(RLNs)are very common in patients with nasopharyngeal carcinoma(NPC)undergoing *** most suitable treatment option for enlarged RLNs depends on the pathological ***,RLN sampling is difficult and imminent in the clinic *** recently developed a novel minimally invasive technique termed endoscopic ultrasound-guided fine needle aspiration(EUS-FNA)for sam-pling RLN tissues sufficient for pathological or cytological ***:We enrolled 30 post-radiotherapy patients with NPC with suspected RLN metastasis detected via mag-netic resonance imaging(MRI).The EUS probe was introduced into the nasopharynx via the nostrils,and EUS was then used to scan the retropharyngeal space and locate the RLN in the anterior carotid ***-FNA was subsequently *** safety and efficacy of using EUS-FNA to sample the RLN tissues were ***:Strips of tissue were successfully sampled from all patients using *** the 30 patients,23 were confirmed to have cancer cells in the biopsied tissues via pathology or cytology examinations with 1 EUS-FNA biopsy *** seven cases without confirmed cancer cells were subsequently reanalyzed by using another EUS-FNA biopsy session,and two more cases were confirmed possessing cancer *** other five patients without con-firmed cancer cells were closely followed with MRI every month for 3 *** follow-up for 3 months,three patients were still considered cancer-free due to the presence of RLNs with stable or shrinking *** rest two patients who showed progressive disease underwent a third EUS-FNA biopsy procedure and were further confirmed to be cancer *** the whole cohort reported here,the EUS-FNA procedure was not associated with any severe ***:EUS-FNA is a safe and effective diagnostic approach for sampling tissues from the RLNs in patients with suspected recurrent NPC.

Structure and properties of SEBS/PP/OMMT nanocomposites

Nanocomposites based on styrene-b-(ethylene-co-butylenes)-b-styrene triblock copolymer(SEBS), polypropylene (PP) and organomontmorillonite (OMMT) were prepared using melt intercalation by a twin mill. The nanostructure of SEBS/PP/OMMT hybrids was characterized by X-ray diffractometry(XRD) and transmission electron microscopy(TEM). The d001 space of OMMT and SEBS/PP/OMMT hybrid increases from 1.30 nm to 3.52 nm and 4.06 nm, respectively which indicate, that the C16 and SEBS/PP could intercalate in the silicate layers and expand the basal spacing. The formation of intercalated structure was also confirmed by TEM. The results show that the individual silicates are well dispersed in the elastomer matrix. The glass transition temperature (tg) of PS block of SEBS was investigated by dynamic mechanical analysis (DMA). It is shown that there are remarkable enhancements in tg of PS block containing a small mass fraction of OMMT. tg of PS in SEBS of the nanocomposites has been increased about 39℃when containing 4% OMMT. The influence of OMMT on the dynamic storage moduli is temperature dependent. Below 50℃the SEBS/PP/OMMT composites with higher OMMT contents show higher dynamic storage moduli. OMMT has the function of reinforcing SEBS/PP elastomers. However, the influence of OMMT on the mechanical properties of the nanocomposites is not significant according to the tensile test and tear test.
