Recently, two researches by Doucet et al. and Collin et al. used functional neuroimaging as a tool to improve the management of schizophrenia and other psychotic disorders [1, 2]. We would like to highlight several issues in relation to cardiopulmonary comorbidity, radiomics and machine learning, and therapeutic regimens, along with their clinical implications.

Information flow among auditory and language processing-related regions implicated in the pathophysiology of auditory verbal hallucinations（AVHs） in schizophrenia（SZ） remains unclear. In this study, we used stochastic dynamic causal modeling（s DCM） to quantify connections among the left dorsolateral prefrontal cortex（inner speech monitoring）, auditory cortex（auditory processing）, hippocampus（memory retrieval）, thalamus（information filtering）, and Broca＇s area（language production） in 17 first-episode drug-na？¨ve SZ patients with AVHs, 15 without AVHs, and 19 healthy controls using resting-state functional magnetic resonance ***, we performed receiver operating characteristic（ROC） analysis and correlation analysis between image measures and symptoms. s DCM revealed an increasedsensitivity of auditory cortex to its thalamic afferents and a decrease in hippocampal sensitivity to auditory inputs in SZ patients with AVHs. The area under the ROC curve showed the diagnostic value of these two connections to distinguish SZ patients with AVHs from those without AVHs. Furthermore, we found a positive correlation between the strength of the connectivity from Broca＇s area to the auditory cortex and the severity of AVHs. These findings demonstrate, for the first time, augmented AVHspecific excitatory afferents from the thalamus to the auditory cortex in SZ patients, resulting in auditory perception without external auditory stimuli. Our results provide insights into the neural mechanisms underlying AVHs in SZ. This thalamic-auditory cortical-hippocampal dysconnectivity may also serve as a diagnostic biomarker of AVHs in SZ and a therapeutic target based on direct in vivo evidence.

Association of Optic Radiation Integrity with Cortical Thickness in Children with Anisometropic Amblyopia

Previous studies have indicated regional abnormalities of both gray and white matter in amblyopia. However, alterations of cortical thickness associated with changes in white matter integrity have rarely been reported. In this study, structural magnetic resonance imaging and diffusion tensor imaging （DTI） data were obtained from 15 children with anisometropic amblyopia and 15 age- and gender-matched children with normal sight. Combining DTI and surface-based morphometry, we examined a potential linkage between disrupted white matter integrity and altered cortical thickness. The fractional anisotropy （FA） values in the optic radiations （ORs） of children with anisometropic amblyopia were lower than in controls （P 〈 0.05）. The cortical thickness in amblyopic children was lower than controls in the following subregions： lin- gual cortex, lateral occipitotemporal gyrus, cuneus, occip- ital lobe, inferior parietal lobe, and temporal lobe （P 〈 0.05, corrected）, but was higher in the calcarine gyrus （P 〈 0.05, corrected）. Node-by-node correlation analysis of changes in cortical thickness revealed a significant association between a lower FA value in the OR and diminished cortical thickness in the following subregions： medial lingual cortex, lateral occipitotemporal gyrus, lat- eral, superior, and medial occipital cortex, and lunate cortex. We also found a relationship between changes of cortical thickness and white matter OR integrity in amblyopia. These findings indicate that developmental changes occur simultaneously in the OR and visual cortex in amblyopia, and provide key information on complex damage of brain networks in anisometropic amblyopia. Our results also support the hypothesis that the pathogenesis of anisometropic amblyopia is neurodevelopmental.

＜正＞Objective Orexins,including orexin A and B,are neuropeptides expressed by the neurons of lateral hypothalamus and play multiple roles in feeding,sleeping,rewarding,*** is recently reported that the number of orexin neurons can be negatively affected with the progression of Parkinson’s disease（PD）,but the detailed time dependency is not clear so *** we investigated the schedule of orexin neurons’ loss in PD rats by immunohistochemical and ELISA methods. Methods PD rats were made by injection of 6-OHDA into the medial prefrontal *** post operation of 4 d,7 d,10 d, 2 w,3 w,5 w,7 w and 9 w,cerebrospinal fluid（CSF） of each rats was obtained through cisterna magna for ELISA assay and the rats were then sacrificed for immunohistochemical staining of tyrosine hydroxylase（TH） in midbrain and orexin A in *** TH neurons in substantia nigra pars compacta decreased significantly at 4 d after operation and nearly totally disappeared at 1 w after *** contrast to this,the number of orexin A neurons decreased at 7 w after operation and reached only half of the control at 9 *** to this tendency,the concentration of orexin A in the CSF of 9 w group was significantly reduced by 25%as compared with that of *** Collectively,these results suggest that the degeneration of orexin neurons in hypothalamus is far preceded by that of TH neuron and may thus underlie the sleep disorders in PD patients in the later phase of the disease.
